Chinese Journal of Integrated Traditional and Western Medicine 2011;31(3):386-390
OBJECTIVETo observe the effect of sequential method of nourishing yin and tonifying yang on serum sex hormone levels in rats with polycystic ovary syndrome (PCOS), and to explore its possible mechanisms.
METHODSPCOS rat models were established by injecting dehydroepiandrosteron (DHEA) subcutaneously. A total of 70 21-day female SD rats were randomly divided into seven groups, ten in each group: the normal control, the model control, the yin nourishing, the yang tonifying, the sequential method, the Westem medicine, and the combined medication. Rats in all groups were treated by gastrogavage for twelve successive days. Serum testosterone (T) and estradiol (E2) were detected by chemoluminescence method. Serum androstenedione (A), follicle stimulating hormone (FSH), luteinizing hormone (LH), insulin (INS), and insulin-like growth factor-I (IGF-I) were detected by radioimmunoassay (RIA).
RESULTSSerum levels of A, T, and E2 were significantly higher in the model control group than in the normal control group (P < 0.05, P < 0.01). Serum levels of A and T were obviously lower in the sequential group than in the model control group with significant difference (P < 0.01). The serum E2 level was obviously lower in the sequential group than in the model control group (P < 0.05). Serum levels of INS and IGF-I were significantly higher in the model control group than in the normal control group (P < 0.05). Serum levels of INS and IGF-I were significantly lower in the sequential group than in the model control group (P < 0.01).
CONCLUSIONSThe sequential method of nourishing yin and tonifying yang can lower serum levels of A, T, E2, INS, and IGF-I, and reverse its endocrine disturbance in PCOS rats. One of its mechanisms in decreasing serum levels of A, T, E2 might be possibly through lowering serum levels of INS and IGF-I.

OBJECTIVETo observe the effect of the combination of Bushen Xuguan Recipe (BXR) and Ruan-jian Huayu Recipe (RHR) on the matrix metalloproteinases-2 (MMP-2) expression of rats with endometriosis (EMS), and to study the pathogenesis and mechanism of EMS.
METHODSThe EMS rat model was established. Successfully modeled female SD rats were randomly divided into the combination group (BXR+RHR), the RHR group, and the model group. And a normal control group was set up. After 4 weeks of medicinal treatment, rats were sacrificed in the oestrus cycle. MMP-2 distribution and the staining density in ectopic and eutopic endometrium of the rats were observed under microscope. The MMP-2 expression was determined by immunohistochemical assay (SP method).
RESULTSThe MMP-2 expression in the ectopic endometrial tissues of rats in the model group were significantly higher than that of the normal control group (P<0.01). Compared with the model group, the MMP-2 expression decreased in the combination group and the RHR group (P<0.05). The therapeutic effect of the combination group was most obvious by observing the distribution of MMP-2 and changes of positive staining.
CONCLUSIONSThe occurrence of EMS was correlated with increased activities of MMP-2. The combination method could inhibit the activities of MMP-2 in the ectopic endometrial tissues of EMS rats more effectively than RHR alone, which might be achieved through improving the endocrine environment, and reducing the "invasion" capability of the ectopic endometrium.

The Medical Journal of Malaysia 2016;71(3):154-155
Background: Angioleiomyoma of the nasal cavity is an
extremely rare benign neoplasm. It usually occurs in the
lower extremities. Up to date, only few cases of
angioleiomyoma have been reported. First case of
angioleiomyoma of nasal cavity was reported in 1966. We
report a rare case of angioleiomyoma arising from the right
maxillary sinus.
Case Report: A 43-year-old lady presented with recurrent
epistaxis and right nasal obstruction for two months
duration. Clinical examination revealed a huge right nasal
mass obstructing the right nasal cavity. The tumour was
excised completely via endoscopic endonasal surgical
approach. Histopathological examination confirmed the
tumour is sinonasal angioleiomyoma. Postoperatively, she
recovered well without any recurrence after a year of followup.
Conclusion: This tumour has an excellent prognosis and
recurrence is extremely rare if excised completely.

A novel simple, sensitive fluorescence immunosensing method based on aptamer-plasmid complex amplification was developed. This method utilized the specific recognition between antibody and antigen as well as aptamer-plasmid complex and the intercalation of fluorescence dye SYBR Green Ⅰ in the groove of duplex plasmid DNA in detection of Platelet-Derived Growth Factor BB (PDGF-BB). The immunoassay was performed in the microtiter wells in which rabbit anti PDGF-BB antibody was immobilized. The PDGF BB analyte was captured by the primary antibody and then sandwiched by the aptamer-plasmid DNA complex. The introduction of fluorescence dye SYBR Green Ⅰ allows for the detection of the sandwiched immunocomplex of antibody/anigen/aptamer-plasmid complex. Under the optimized conditions of salt concentration, ratio of aptamer to PUC19,and SYBR Green Ⅰ concentration, the proposed method offers a linear detection range from 0.2 μg/L to 200 μg/L with a detection limit of 0.1μg/L.

Objective To detect the mutations of COL7A1 gene in three cases of dystrophic epidermolysis bullosa pruriginosa (DEBP). Methods Clinical data were collected from 3 patients with DEBP. Skin lesions were obtained from these patients and subjected to transmission electron microscopy. DNA was extracted from the peripheral blood of the 3 patients, their 16 relatives, and 150 unrelated normal human controls, and PCR was performed to amplify all the exons and flanking sequences of COL7A1 gene followed by sequencing. Results The patient 1 and 2 had family history, whereas the case 3 was sporadic. Transmission electron microscopy showed tissue cleavage beneath lamina densa in case 1 and slightly decreased anchoring fibrils in some areas of the lesions in case 1 and 3. Three heterozygous mutations of COL7A1 gene, i.e., c. G6734T, c.G6859A and c. G5318T, which leaded to three amino acid mutations, i.e., p. G2245V, p. G1773V and p. G2287R, were found in patient 1, 2 and 3 respectively. Of them, p. G2245V and p. G1773V were novel mutations. The mutations strictly cosegregated with the phenotype in the patients of family 1 and 2. No mutation was detected in the unaffected parents of patient 3 or the 150 unrelated healthy controls. Conclusions The p. G2245V, p. G2287Rand p. G1773V mutations of COL7A1 gene may be responsible for the phenotype of DEBP in the three cases, and of them, p. G2245V and p. G1773V have never been reported.
