1.Preparation and ectopic bone formation activity of recombinant human bone morphogenetic protein-2 loaded chitosan nanospheres in vivo

Objective To prepare recombinant human bone morphogenetic protein-2(rhBMP-2)loaded chitosan nanospheres and to evaluate their ectopic bone formation activity in vivo. Methods The rhBMP-2 loaded chitosan nanospheres were prepaid using the ionic crosslinking method with TPP as a crosslinking agent.The morphological properties and particle size distribution were evaluated by transmission electron microcopy(TEM) and particle size analyzer.An intramuscular ectopic bone formation test was carried out to compare the ectopic bone formation activity in 24 SD rabbits that were randomized into 4 even groups.rhBMP-2 loaded chitosan nanospheres (1 mg),rhBMP-2 (1 mg),chitosan nanospheres and nothing were implanted into the muscle pouch at the left thigh,respectively,in groups A,B,C and D. Results The rhBMP-2 loaded chitosan nanospheres were well distributed and spherical in shape.The average particle diameter was 230.0 nm,the entrapment efficiency was 66.87% ± 4.58%,and the loading rate was 33.44 ± 2.29 μg/mg.The mean ALP activity was respectively 1.94 ± 0.35 kat/g,1.48 ± 0.56 kat/g,0.20 ±0.07 kat/g and 0.18 ±0.06 kat/g in groups A,B,C and D,with a significant difference between the 4 groups( F =42.959,P =0.000).The mean Ca2 + content was respectively 5.20 ± 1.42 μg/mg,3.80 ± 1.40 μg/mg,0.19 ± 0.08 μg/mgand 0.20 ± 0.08 μg/mg in groups A,B,C and D,with a significant difference between the 4 groups ( F =39.242,P =0.000).Groups A and B were obviously higher than groups C and D and group A was obviously higher than group B in terms of ALP activity and Ca2+ content. Conclusion The rhBMP-2 loaded chitosan nanospheres prepared by ionic crosslinking method show a good property and obvious ectopic bone formation activity.
2.The correlation between infectious complications after radical resection of gastric cancer and the changes of T lymphocyte subsets in the patients with gastric cancer

Objective Our retrospective study is aimed to discuss the relationship between the curve changes of T lymphocyte subsets before and after the operation and the infectious complications after radical resec -tion of gastric cancer .Methods Clinical data of patients with gastric cancer received treatment at Central Hospi -tal of Panjin City,Liaoning Province from 2012 to 2014 were retrospectively analyzed .The enrolled patients were divided into two groups according to the infectious complications ,observe group ( patients with infectious complica-tions) and control group ( patients without infectious complications ) .The results of T lymphocyte subsets in two groups were compared .Results A total of 70 patients was retrospective analyzed , including 33 pationts in ob-serve group and 37 pationts in control group.At the 4th day after operation,the levels of CD3,CD4 in the observe group were obviously lower than those in control group .There was no statistical difference at the other time points . At the 4th and 9th day after operation,the levels of CD8,CD4/CD8 rate in the observe group were obviously lower than those in control group .There was no statistical difference at the other time points .Conclusion There is re-markable relationship between the curve changes of T lymphocyte subsets before or after the operation and the in -cidence of infectious complications after radical resection of gastric cancer .
3.Returning Path of Medical Humanities Spirit Based on the Current Situation of Medical Humanities Teaching

Therefore , it needs to be nourished by rich humanistic spirit .At present , the generalization of medical technology , the abnormal development of medicine socialization and the less care from humanities to medical and many other reasons lead to the lack of humanistic spirit of medicine .To correct the phenomenon of the disorder, we should explore the effective means actively to provide powerful support for the returning of the medical humanism .There-fore, using an appropriate method to strengthen the humanities education of medical students and attention in the classroom will be regarded as good path in medical humanities spirit returning .
4.Progress and prospect of applications of silk fibroin in construction of tissue engineering scaffold.

With the development of tissue engineering, a variety of forms of silk fibroin (SF) scaffolds has been applied to research of constructing variety of organization based on cells, which has become scientific focus in recent years. In this paper we introduced the source and structure of SF and the fabrication method of the scaffold, and also address the SF application progress in several relevant fields of tissue engineering, such as bone, cartilage, skin, blood vessel and nerves. Finally, we discuss the future leading prospect of the SF in order to provide reference for subsequent research.

5.Transplantation of rabbit embryonic full-thickness retina into the subretinal space of rats

Objective To study the transplantation of embryonic full-thickness retina, including neural retina and retinal pigment epithelial (RPE), into the subretinal space of rats and to investigate the development of the grafts. Methods A total of 34 eyes from 3-week-old pigmented rabbit embryos were treated with the dispase to obtain pieces of the full-thickness retina, and then grafts were injected into the subretinal space of the right eyes of 20 Wistar rats and 14 RCS (Royal college of surgeons) rats. Host rats were sacrificed at 3, 7, 14, and 28 d after transplantation. Sections were stained with hematoxylin eosin and immunohistochemistry. Results The rabbit embryonic retina was capable of developing and differentiating in the subretinal space of the host rats. Conclusion The rabbit embryonic retina is capable of developing and differentiating in the subretinal space of host rats. Transplantation of xenografts of embryonic full-thickness retina is practicable, but the methods of transplantation need to be improved.
6.Clinical significance of clock-drawing test and MMSE in Alzheimer's disease

ObjectiveTo observe the clinical sense of clock-drawing test(CDT) and Mini Mental Status Examination(MMSE) in Alzheimer's disease(AD). MethodsMMSE and CDT were used to assess the intellectual ability in AD group and control group. ResultsThe orientation force, immediate memory, ability of calculation, short-term memory and speech ability in AD group were significantly decreased than that of the control group(P<0.05). The score of CDT in AD group was markedly suppression than that of control group(P<0.01). ConclusionThe CDT and MMSE are the ideal cognitive screening test to determine the degree in Alzheimer's disease.
7.Regulation of transcription of rRNA synthesis by Apak under hypoxic conditions

Objective To explore the effect and mechanism of Kruppel-associated box(KRAB) type zinc-finger protein Apak on the transcription of ribosomal RNA(rRNA) under hypoxic conditions.Methods Real-time quantitative PCR(qRT-PCR) was used to detect the level of rRNA and the effect of Apak on the transcription of rRNA.The expression and phosphorylation level of proteins were examined by Western blotting.Immunofluorescence assay was used to observe the location of Apak.Results Under hypoxia (0.3%O2),the transcription level of rRNA increased first and then decreased in Apak wild type cells.In Apak knock-out cells,the transcription level of rRNA kept decreasing under hypoxic conditions.The repression of Apak on rRNA synthesis and the nucleolus location of Apak disappeared,and the expression and phosphorylation level of Apak were down-regulated under hypoxic conditions for 3 h.Conclusion Under hypoxic conditions,the disapperance of Apak repression on rRNA transcription could lead to a temporary increase in rRNA.

9.Analysis of the PICU critically ill children with pediatric critical score and pediatric death risk score and their value

Objective To investigate the pediatric intensive care unit (PICU) in critically ill children with pediatric critical score (PCIS) and pediatric death risk score (PRISM),comparing its value.Methods A total of 100 cases of matching children with acute respiratory distress syndrome (ARDS) to stay in our hospital during 2007.07 ~2012.07 was retrospectively analyzed.Check PICU most serious disease in this group of children,living PICU most severely ill when diagnosed with acute respiratory distress syndrome (ARDS),when it was suffering from ARDS most serious line PCIS with PRISM score,and then using the Logistic multiple regression to analyze the scores of the two scoring ability as predicting the ARDS death in children with risk factors,and using two-factor analysis of variance of the two scoring methods by judging linear correlation existence.Results (1)PCIS with PRISM score showed a negative correlation (r=-0.6031,P <0.01),the linear regression equation y =-0.2389x +74.816 (P<0.01).(2) There was no statistical difference between the ARDS death group and survival group (P > 0.05) ; By PCIS PRISM score LOGISTIC regression analysis of the risk of death in children,there was no statistical difference (P > 0.05).Conclusions PCIS and PRISM score cannot be the standards to forecast the death of the ARDS patients,but can provide an objective referring standard of ARDS's treatment.
10.Establishment of Anterior Instability of Gienohumeral Joint on Rabbits

Objective To establish the animal model(rabbit) of shoulder instability for providing the experimental basis of related studies.Methods shoulder joints of 6 skeletally matured New Zealand white rabbits were measured with the anatomical characteristics and joint motion range in order to determine the operative approach,the similarities of skeletal structure,surrounding muscles,joint capsule and ligaments between rabbit and human were compared.Eighteen rabbits,weighing between 2.5 and 3kg were randomized into 2 groups(experimental group 12 and control 6).In experimental group,instability model was established by means of the operation procedure as follows:the glenohumeral joint of right shoulder was exposed by cutting the subscapularis tendon,then the anteroinferior labrum and surrounding capsular ligament were excised.For control group,sham operation was performed by only curing the skin and superficial tissue,the subscapularis tendon and glenoid structures was kept intact.The specimens of 6 rabbits were harvested 6 weeks after operation.The remaining rabbits of both experimental and control groups were killed 12 weeks post the operation for specimens processing.The motion range and instability were measured in all specimens.Results The gross anatomy of shoulder ioint in rabbit is similar to human.In experimental group,the stability is showed less than control,grade 2 anterior instability was found.The motion range was restricted in all directions,such as external and internal rotation,extension and abduction compared to control group.Conclusion Rabbit is suitable to establish animal model for shoulder instability and related iniuries,the animal model which was designed by this study could provide valuable information for the research of anterior shoulder instability.
