2.Analysis of the risk factors related to early neurological deterioration in patients with recurrent cerebral infarction

Objective To study the risk factors related to early neurological deterioration (END) in patients with recurrent cerebral infarction (RCI). Methods Three hundreds and twenty-six RCI patients admitted to our hospital from January 2014 to March 2015 were divided into END group (n = 66) and non-END group (n =260). The clinical and laboratory data were collected, and their cerebrovascular lesions were accessed according to the angiographic findings. Results (1) Single factor analysis showed the level of cholesterol was significantly higher in the END group than that of the non-END group (P < 0.05). The proportion of patients suffering from smoking, arterial occlusions, early onset stroke associated pneumonia (SAP) and hyponatremia was also higher in the END group (all P < 0.05). (2) Logistic regression analysis showed that arterial occlusions (OR = 2.137, 95%CI 1.093~4.181), SAP (OR=2.001, 95%CI 1.023~3.912) and hyponetremia (OR=2.467, 95%CI 1.217~ 5.000) were independent risk factors for END of RCI patients (all P < 0.05). Conclusion Arterial occlusions, SAP and hyponatremia may be used as predictors of the early development of RCI patients.

Objective To establish an in vitro skin sensitization test,human cell line activation test (h-CLAT),based on THP-1 cell line (a human acute monocytic leukemia cell line),and to assess the sensitizing potency of plant raw materials of chemical and cosmetic products by this in vitro skin sensitization test.Method THP-1 cells were cultured in vitro and exposed to 11 reference skin sensitization chemicals and 9 samples,by monitoring the cell viability,cell surface marker CD54 /CD86 and relative fluorescence intensity of cells surface after the cells was exposures to the substances,and to discover whether there is a positive reaction.At the same time,Buehler test was used to validate the results of samples tested by h-CLAT.Results 11 reference chemicals were distinguished correctly by h-CLAT.Among the 9 samples tested,7 samples were recognized as negative sensitizer and 2 plant extracted substances were identified as suspicious skin sensitizer.The qualitative classification of the 9 samples by h-CLAT test was consistent with the results obtained by animal test.Conclusions The h-CLAT-in vitro test can be used to replace some animal tests for the prediction of soluble skin sensitizing substances.
5.Clinical analysis of fungus infection in acute exacerbation of chronic obstructive pulmonary disease

Objective To explore the risk factors and targeted therapy for fungus infection in acute exacerbation of chronic obstructive pulmonary disease (COPD) pulmonary.Methods 83 acute exacerbations of COPD patients with pulmonary fungus infection were selected as the observation object(COPD fungal infection group),according to the time sequence of random month by month in acute exacerbations of COPD no secondary fungal infection of the 80 patients hospitalized patients as control group,analysis of risk factors lead to fungal infection.Results Two groups in age,live ICU time,antibiotic use time,hormone use time,albumin level mechanical ventilation people with diabetes mellitus,merger,as cor pulmonale had statistically significant differences (P < 0.05) ; The conditional Logistic multiariate analysis,age,ICU patients live time,antibiotics use time,hormone use time,albumin level was acute exacerbations of COPD factors such as pulmonary fungus infection independent risk factors(P <0.01) ;Fungus infection were combined with antimycotic,cured 69 cases (83.13%) improved 12 cases (14.46%),there was no change in 1 case(1.20%),death 1 case (1.20%).Conclusion Patients older age,live long,long time ICU antibiotics and hormones and the low level of albumin is acute exacerbations of COPD pulmonary fungus infection independent risk factors,timely diagnosis and prognosis of patients with antifungal treatment is good.
7.Effect of hydrogen sulfide postconditioning on systolic function of left ventricle during myocardial ischemia-reperfusion in rats

Objective To investigate the effect of hydrogen sulfide postconditioning on the systolic function of left ventricle during myocardial ischemia-reperfusion (IR) in rats. Methods Part Ⅰ Adult male SD rats weighing 200-250 g were anesthetized with pentobarbital 40 mg/kg and heparin 250 U/kg. Their hearts were excised and perfused in a Langendorff apparatus with K-H solution saturated with 95% O2-5% CO2 at 37 ℃. Forty isolated rat hearts were randomly divided into 5 groups ( n = 8 each) after 20 min of equilibration: control group (group C); IR group; sodium hydrosulfide 1,10, 100 μmol/L postconditioning group (group SP1, SP10, SP100 ).In group Cthe hearts were perfused continuously for another 100 min. In group IR, the hearts were reperfused for 60 min after 40 min ischemia induced by 10 ml/kg ST. Thomas solution. In group SP1 , SP10 and SP100 the hearts were perfused with K-H solution containing sodium hydrosulfide 1, 10, 100 μmol/L for 2 min before reperfusion.LVDP and ± dp/dtmax were recorded at the end of equilibration and reperfusion. Part Ⅱ Cardiomyocytes were isolated from the male SD rats (weighing 200-250 g) and then cultured in CO2 incubator for 4 h. Sixty-four dishes of cultured myocytes were randomly divided into 4 groups( n = 16 each): control group (group C), hypoxia/reoxygenation group (group HR), hydrogen sulfide postconditioning group (group SP) and hypoxia postconditioning group (group HP). Group C were cultured continuously for 2 h. Group HR, SP and HP were exposed to 1 h hypoxia (95%N2-5%CO2 ) followed by 1 h reoxygenation. In group SP 10 μmol/L sodium hydrosulfide was added and the myocytes were then incubated for 2 min before reoxygenation. In group HP the cultured myocytes were expased to 3 min reoxygenation followed by 3 min hypoxia for 3 times before the 1 h reoxygenation. Mitochondrial membrane potential and F-actin expression were determined. Results Part Ⅰ Compared with group C, LVDP and ± dp/dtmax were significantly decreased at the end of reperfusion in group IR (P < 0.05), while no significant difference was found in group SP1 , SP10 and SP100(P >0.05). Compared with group IR, LVDP and ± dp/dtmax were significantly increased in group SP ( P < 0.05). There was no significant difference in LVDP and ± dp/dtmax among group SP1, SP10 and SP100(P >0.05). Part H Compared with group C, the mitochondrial membrane potential was significantly decreased in group HR and HP, and the expression of F-actin was significantly up-regulated in group HR, SP and HP ( P < 0.05). Compared with group HR, the mitochondrial membrane potential was significantly increased and the expression of F-actin up-regulated in group SP and HP ( P < 0.05 ). There were no significant difference in the mitochondrial membrane potential and expression of F-actin between group SP and HP ( P >0.05).Conclusion Hydrogen sulfide postconditioning can improve left ventricular systolic function during IR in rats by stabilizing mitochondrial membrane potential and promoting aggregation of F-actin.
8.Targeting therapy of SLE associated thrombocytopenia with labeled antibody in vitro

Objective To discuss the feasibility and effect of labeled antibody on targeting therapy of systemic lupus erythematosus(SLE) associated thrombocytopenia.Methods MTX-IVIG conjugate was prepared by indirect and direct cross linking,and the binding ability of it to Fc fragment was detected by indirect immunofluor-escence.The killing activity of the conjugate was detected by MTT method using murine macrophage with Fc receptor and strain U937 of human monocytic leukemia cell as targets.Results Conjugation showed stronger cytotoxicity upon target cells than free MTX,and it showed less cytotoxic effect on Fc receptor negative cells compared with the positive ones.IVIG-HSA-MTX restrained the phagocyte of macrophage.The killing effect of IVIG-HSA-MTX was significantly stronger than that of IVIG-MTX.Conclusion The conjugation can show a highly specific cytotoxicity upon mononuclear-macrophage in vitro.
9.HIGH LEVEL EXPRESSION AND ACTIVITY DETECTION OF SINGLE CHAIN IMMUNOTOXIN 183B_2ScFvPE38 AGAINST OVARIAN CARCINOMA

Objective To prepare the immunotoxin protein (183B 2ScFvPE38) which might be useful in immuno guided therapy for ovarian carcinoma and study the activity of the protein. Methods The methods of ELISA and cytotoxicity were used to study the immunotoxin after induced with IPTG and the activity of the immunotoxin. Results The expressed fusion proteins were detected mostly as inclusion bodies at high level, and soluble immunotoxins were also observed. The results showed liable activity of antibody part and toxic part. Conclusion The recombinant fusion protein 183B 2ScFvPE38 keeps the activity of both components and might be of great use in the future to deal with ovarian carcinoma. [
10.Integrated CAMVA and BCOP methods to predict eye irritation caused by cosmetics

Objective To explore the use of integrated two methods in vitro in prediction of eye irritation caused by cosmetics.Method Chorioallantoic membrane vascular assay ( CAMVA), bovine corneal opacity and permeability (BCOP) and Draize rabbit eye irritation test were used to determine the predictive potential of eye irritation of 60 kinds of cosmetics.Results CAMVA method was able to distinguish 41 non-irritant samples and 18 irritant samples.BCOP method was able to predict 35 non-irritant samples , 21 mild-moderate irritant samples and 4 severe irritant samples . Combination of CAMVA and BCOP methods could obviously improve the identification ability of irritation , and the classification consistency with Draize rabbit eye irritation testing reached 98.3%.Conclusions The integrated test strategy combined BCOP with CAMVA can be used to appropriately predict ocular irritation of cosmetics , with a prediction range covering non-irritant to severe irritant samples .
