1.The research development of PI3K/AKT/mTOR signaling pathway in Her-2 resistance for the treat-ment of breast cancer

Her-2 targeting therapy is an important part of the treatment for breast cancer with Her -2 overexpression.However,the therapeutic effectiveness is largely influenced by Her -2 resistance.Previous studies have confirmed that PI3K/AKT/mTOR signaling pathway is activated during the process of drug resistance to Her-2-targeting therapy.Therefore,investigating the PI3K/AKT/mTOR signaling pathway and drugs -targeted pathway are important for the treatment of breast cancer .
2.New Research of Nerve Growth Factor on Fracture Healing.

Fracture healing has long been a major concern for orthopedists.Currently,about 10% of fracture patients still have poor fracture healing or bone nonunion.In recent years,research has found that nerve growth factor(NGF)can promote fracture healing.This article reviews the mechanism and research progress of NGF in promoting fracture healing.NGF can promote vascular regeneration and nerve growth at callus and plays an important role in the proliferation and migration of bone cells.New animal experiments and clinical trials have confirmed the role of NGF in promoting fracture healing and further revealed its possible mechanism of action.Further research on NGF can provide new ideas for promoting fracture healing,especially for treating nonunion.

3.The correlation of plasma PAF with cortisol and their roles in the estimating of sepsis prognosis

Objective To investigate the correlation of platelet activating factor(PAF)with cortisol(Cor) ,as well as their rela-tionship with patients after sepsis prognosis .Methods 102 patients with sepsis admitted in the ICU of Chongqing Emergency Med-ical Center from April 2012 to August 2013 were enrolled into sepsis group ,and 40 cases of volunteers served as control group .All the patients were divided into survival group and non-survival group ,according to their prognosis during hospitalization .Plasma PAF and Cor level in control group ,survival group and non-survival group were compared with each other ,and acute physiology and chronic health evaluationⅡ(APACHEⅡ)in survival group and non-survival group were compared .The correlations of plasma PAF with Cor level in control group ,survival group and non-survival group were analyzed ,respectively .Results Plasma PAF and Cor level in survival group and non-survival group were significantly higher than that in control group(P<0 .05) .Plasma PAF ,Cor lev-el and APACHEⅡ scores were higher in non-survival group than that in survival group(P<0 .05) .The correlation of plasma PAF with Cor level in control group was not significant(P>0 .05) ,while the plasma PAF level positively correlated with Cor level in survival group and non-survival group(P<0 .05) .Curves of receiver operating characteristics(ROC)showed that PAF ,Cor and A-PACHEⅡ score could be used as predictors of mortality during hospitalization(Area=0 .708 ,0 .715 ,0 .787) .Conclusion The plas-ma PAF level positively correlates with Cor level in patient with sepsis .PAF ,Cor and APACHEⅡ score have certain guiding signif-icance for the assessment of prognosis during hospitalization in patients with sepsis .
4.Changes in serum PSA before and after treatment for acute urinary retention due to benign prostate hyperplasia

Objective To evaluate the effect of catheterization on the serum prostate specific antigen (PSA) in acute urinary retention (AUR) patients due to benign prostate hyperplasia (BPH). Methods The data of 35 patients with AUR due to BPH were analyzed, and the changes in serum PSA values were compared before and after catheterization. The effect of AUR on serum PSA in BPH patients was discussed. Results It has been found that serum PSA declined from 9.93?8.85ng/ml to 6.46?5.75ng/ml after catheterization for the patients of AUR with BPH, and the values showed a significant difference (P0.05) in the patients in whom the serum PSA ≤4ng/ml, while a significant difference existed in both the group of patients in whom the serum PSA was 4~10ng/ml (from 5.95?1.43ng/ml to 4.09?2.00ng/ml, P10ng/ml (from 18.93?8.49ng/ml to 11.49?6.37ng/ml, P4ng/ml.
5.Experimental study of homogenous bladder transplantation in rats

Objective To establish a model of homogenous bladder transplantation without blood vessel anastomosis in rat,and to investigate the outcome of the transplanted grafts.Methods 20 newborn SD rats were used as donors,and a total of 20 Wistar rats of 5-week-old were used as the reeipiemts for transplantation of homogenous bladder.The grafts were harvested from the SD rats,and they were wrapped with the greater omentum of the recipients without blood vessel anastomosis.Immunosuppressive agent was given postoperatively.10 rats were sacrificed on the 7th day and the remainings rats were sacrificed on the 14th day after bladder transplantation.The transplanted bladder grafts were observed grossly and microscupicully.Results Of 20 rats undergone homogenous bladder transplantation,19 rats were alive and the grafts were in good condition microscopically.There was no obvious rejection reaction in the transplanted bladders.The bladder mucosa,blood vessels and muscle layers of the grafts appeared to be normal microscopically.Conclusions The homogenous bladder transplantation into the omentum without blood vessel anastomosis in rats is considered to be a kind of stable animal model,with a higher survival rate of the tramsplanted bladder.
6.Long-term complications of continent ileocolic urinary reservoir with in situ appendix conduit through umbilicus

Objective To evaluate the long-term complications of continent ileocolic reservoir with in situ appendix conduit through umbilicus and their management.Methods We reviewed 37 patients with bladder diseases who underwent the surgery of continent ileocolic diversion with in situ appendix conduit through umbilicus and followed up for 85 to 136 months(mean 109 months).Long-term postoperative complications were documented.Results Complications occurred in 15 cases(40.5%),including stone formation in urinary pouch in 2 cases,efferent limb stricture and difficulty in catheterization in 3 cases,ureter stricture and hydronephrosis in 7 cases,foreign matter in urinary pouch in 2 cases,efferent limb mucosa exstrophy in 1 case.These complications were correctly treated and good results were achieved.Conclusion Long-term complications of continent ileocolic diversion with in situ appendix conduit through umbilicus were related to nursing in daily life after operation and unsuited design in operation.Nursing in daily life is considered very important in preventing postoperative complications.
7.DEVELOPMENT OF IFA METHOD FOR DETECTING ANTIBODIES OF SARS CORONAVIRUS

Objective To develop a diagnostic test based on indirect immunofluorescence assay(IFA) to detect special antibodies in the serum of SARS patients, thus to provide a reference material for confirmation of the clinical diagnosis of SARS. Methods SARS coronavirus GZ01 and BJ01 strains isolated in our laboratory were used to infect Vero E6 cells. When CPE reached 25%, cells were trypsinized and transferred to 10 well slides in a quantity of 40?l with a cell density of 2?10 7 /ml. After 4 hour incubation at 37℃,the slides were fixed with acetone, and IFA was used to detect antibodies in serum samples, which were obtained from 154 SARS patients and 14 non SARS patients with respiratory disease, as well as 100 healthy volunteers. Results IFA method for detecting antibodies of SARS coronavirus was developed. Sera from one hundred and forty two out of 154 clinically diagnosed patients were IFA positive, with a positive rate of 92 3%. Sera from 14 non SARS patients with respiratory disease and 100 healthy persons were all IFA negative. Conclusion The IFA method we developed was sensitive and specific in detecting SARS antibodies in serum, and was a reliable test for laboratory diagnosis of SARS coronavirus.
8.Effects of Nitrogen and Phosphorus on Growth of Microcystis aeruginosa Strains

100∶1. The specific growth rate of microcystis and P concentration in different P concentration and different N/P ratio cultures decreased with the culture time. Conclusions The initial P concentration affects the microcystis yield significantly, N/P ratio should be concerned in confirming the limiting factor. To control the P concentration in water through different methods will be the effective way to control water bloom.
9.Analysis of correlations between blood lactic acid, lactate clearance rate and emergency stratification as well as prognosis in patients

Objective To investigate the correlations between the level of blood lactic acid (Lac),lactate clearance rate (LCR) and emergency stratification Ⅰ or Ⅱ as well as the prognosis in patients.Methods A retrospective analysis was conducted.The clinical data of 370 critically ill patients with emergency stratification Ⅰ or Ⅱ accompanying with hyperlactacidemia admitted to emergency center of People's Hospital of Wuwei City during January 2013 to April 2015 were analyzed.The patients were allocated into two groups:Lac ≥ 10 mmol/L (n =181) and Lac 4-10 mmol/L (n =189).Base excess (BE),acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) score and hospital mortality were compared between the two groups.The differences in initial Lac,6-hour LCR and APACHE Ⅱ score were compared between survival group and death group as well as emergency stratification Ⅰ and Ⅱ groups.The correlation between initial Lac,6-hour LCR and APACHE]Ⅱ score was analyzed by Pearson correlation method.Results ① With the increase in Lac level,the negative deviation extent of BE and APACHE Ⅱ score in critical patients were gradually increased [BE (mmol/L):-16.74 ± 8.21 vs.-5.98 ± 8.43,APACHE Ⅱ score:27.6 ± 5.6 vs.20.1 ± 4.8],and hospital mortality was increased [76.79% (139/181) vs.43.39% (82/189),all P < 0.01].② The initial Lac and APACHE Ⅱ score of the death group were significantly higher than those of the survival group [initial Lac (mmol/L):8.81 ± 4.71 vs.4.43 ± 2.82,APACHE Ⅱ score:23.6 ± 5.6 vs.17.3 ± 3.7],and 6-hour LCR was significantly decreased [(12.26 ± 6.47)% vs.(35.16± 10.63)%,all P < 0.01].③ Patients in emergency stratification Ⅰ group had a higher initial Lac and a higher APACHE Ⅱ score but a lower 6-hour LCR level than those in emergency stratification Ⅱ group [initial Lac (mmol/L):8.7±2.6 vs.6.8±2.0,APACHE Ⅱ score:25.2±6.3 vs.16.3±4.7,6-hour LCR:(14.8±4.7)% vs.(33.5±5.8)%,both P < 0.01].④ It was shown by correlation analysis that initial Lac was significantly positively correlated with APACHE Ⅱ score (r =0.731,P =0.017) in 370 emergency critical ill patients,while 6-hour LCR was negatively correlated with APACHE Ⅱ score (r =-0.694,P =0.010).Conclusions The early arterial blood Lac of patients with emergency stratification Ⅰ was significantly higher than emergency stratification Ⅱ,and the 6-hour LCR in patients with emergency stratification Ⅱ was significantly higher than emergency stratification Ⅰ.Furthermore,the Lac level and LCR were simple and easy to implement as compared with APACHE Ⅱ score in emergency critical ill patients.
10.Effects of Botulinum Toxin Type A on Proliferation, Apoptosis and ERK/MAPK Signaling Pathway of Hypertrophic Scar Fibroblasts

Objective To investigate the effects of botulinum toxin type A on the proliferation,apoptosis and ERK/MAPK signaling pathway of human hypertrophic scar fibroblasts in vitro.Methods Fibroblasts were isolated from human hypertrophic scar and cultured by tissue explant method.The cells were divided into control and botulinum toxin type A groups.The botulinum toxin group A was cultured in DMEM medium containing 0.4U/L botulinum toxin type A.The control group was cultured with DMEM alone.Cell proliferation was measured by CCK8 kit at days 1,3,5 and 7 and compared within groups.Apoptosis of fibroblasts was detected by flow cytometry which treated with botulinum toxin and control group.Western blot was used to detect the relative expression of collagen Ⅰ,p-ERK1/2 and total ERK protein in fibroblasts of control,botulinum toxin type A and U0126 groups.Results Botulinum toxin type A could significantly inhibit the proliferation of fibroblasts,the number of cells was only 68.9% in the control group at 7th days.The apoptotic rate of botulinum toxin type A was 35.9%,which was significantly higher than that of the control group.ERK 1/2 protein expression in the three groups was not significantly different (P > 0.05),while p-ERK 1/2 expression in the control group was still significantly higher,but the p-EKR 1/2 in botulinum toxin type A and U0126 group was significantly inhibited.Collagen Ⅰ was highly expressed in the control group,which was significantly higher than that in the botulinum toxin type A and U0126 groups (P < 0.05).Conclusion Botulinum toxin type A can inhibit the ERK/MAPK signaling pathway and increased fibroblasts apoptosis,decreased proliferation activity and collagen Ⅰ secretion.
