1.EFFECT OF ZINC DEFICIENCY ON THE GENE EXPRESSION OF VITAMIN D RECEPTOR AND CaBP IN DUODENAL MUCOSA OF RATS

Objective: To study the effect of zinc deficiency(ZD)on the gene expression of vitamin D receptor(VDR)and CaBP in duodenal mucosa of rats. Methods: Thirty weaning male rats were randomly divided into three groups:ZD,paired-fed(PF),zinc adaquate(ZA). After 15 d feeding, took the duodenal mucosa to extract RNA,and measured the levels of VDR mRNA and CaBP mRNA in duodenal mucosa by real time fluorescent quantity PCR. Results: The VDR gene expression of ZD group was downregulated 88.89% as that of PF group,and 50.00% as that of ZA group. CaBP gene expression of ZD group was downregulated 80.16% as that of the PF group,and 88.50% as that of ZA group. Conclusion: Zinc deficiency, by changing the activity of VDR and the mRNA expression of VDR,affects the transcription of the target gene CaBP,and then the absorption of calcium that causes allo-osteogenesis.
2.Study of HCV genotype and histopathology on post-transplant recurrent hepatitis C

Objective To explore the universal pathologic changes as well as the characteristic features of post-transplant hepatitis C caused by different HCV genotypes and study the relationship between pathologic change with HCV genotypes.Methods Fifty cases of HCV-related post-transplant hepatitis C with confirmative serum HCV RNA positivity and HBV DNA negativity between January 2001 to April 2002 were collected from Starzl transplant institute, Pittsburgh university. Liver biopsy from all cases was obtained at the same time. The parameters of histology activity index (HAI), hepatic fibrosis stage (HFS), liver steatosis (LS) and rejection were evaluated. The data were analyzed by SAS 6.12 statistical software using Wilcoxon method.Results Several HCV genotypes or subgenotypes such as 1a, 1b, 2, 3a and 4 were separated at this group. Although there was no statistical difference in HAI, HFS between individual genotype groups, there was significant difference in LS. 3.Clinical studies transurethral holmium laser treatment of ureteral orifice cyst and stones

Objective Discussion transurethral holmium laser treatment of ureteral orifice cyst and stones efficacy and safety . Methods A retrospective analysis of 23 cases of transurethral holmium laser treatment in patients with ureteral orifice cysts and stones (minimally invasive group) and 11 cases of open surgical excision of the cyst and stone -treated patients (open group), two groups were compared operative time, blood loss , catheterization time, hospital stay, postoperative complication rate .Results Minimally invasive surgery patients time (20.0 ±5.6)min, blood loss (15.0 ±2.3)ml, ambulation time (10.0 ±2.5)h, catheterization time (2.3 ± 0.2)d, length of hospital stay (5.0 ±0.6)d; open group of patients were (90.0 ±10.3)min, (80.0 ±12.7)ml, (48.0 ±5.3)h, (7.0 ±1.1)d, (11.0 ±1.7)d, two groups of indicators differences were statistically significant ( P <0.05).Postoperative complica-tions occurred in patients with minimally invasive group was 8.6%, down from 45.4% in the control group, the difference was statisti-cally significant ( P <0.05).Conclusions Transurethral holmium laser treatment of ureteral orifice cysts and stones with less trau -ma, less bleeding, shorter operative time, rapid recovery, the advantages of the low rate of complications , treatment of ureteral orifice cysts and stones safe and effective method .
4.Epidemiological analysis on leprosy in China, 2001-2010

ObjectiveTo study the epidemiological characteristics of leprosy in China,2001-2010.MethodsData were collected from the database of the national system of leprosy surveillance.A descriptive and comparative analysis was performed.ResultsTotally,15 507 new leprosy cases were detected from 2001 to 2010 with an average case detection rate of 0.118 per 100 000 population.Among these cases,2.7% were children under 15 years,86.5% multibacillary,and 22.5% suffered from grade 2 disability.From 2001 to 2010,a total of 1506 relapse cases were detected,and relapse occurred after multi-drug therapy(MDT) in 464 of these cases.There were significant differences between low and high endemic areas in the proportion of children under 15 years,females,immigrant patients among newly detected patients as well as the proportion of cases of relapse after MDT among all the relapse cases.By the end of 2010,the registered leprosy cases were 6032 with a prevalence rate of 0.450 per 100 000 population,among whom 2886 were under MDT.ConclusionsThe leprosy case detection rate continued to decrease in China from 2001 to 2010 with an unequal distribution.The pocket areas were in Yunnan province,Guizhou province,Sichuan province,Guangdong province,Hunan province and Tibet Autonomous Region.It is warranted to enhance the control of leprosy and reduce the prevalence of disability due to leprosy.
6.Differences of Plasma NT-proBNP Level in Patients With Left Heart Failure Combining or not Combining Pulmonary Hypertension

Objective: To observe the relationship between plasma levels of NT-proBNP and pulmonary hypertension in patients with left heart failure (LHF).
Methods: A total of 391 LHF patients were enrolled in our study, according to pulmonary arterial pressure, the patients were divided into 3 groups: Control group, the patients without pulmonary hypertension,n=102, Mild pulmonary hypertension group,n=94 and Moderate to severe pulmonary hypertension group,n=195. Plasma levels of NT-proBNP, left ventricular diameter (LVD), right ventricular diameter (RVD) and left ventricular ejection fraction (LVEF) were examined and compared among 3 groups.
Results: Plasma levels of NT-proBNP and RVD in Mild pulmonary hypertension group and Moderate to severe pulmonary hypertension group were higher than Control group, those indexes in Moderate to severe pulmonary hypertension group were higher than Mild pulmonary hypertension group,P<0.05-0.001. While LVD and LVEF were similar among 3 groups, P>0.05.
Conclusion: Plasma levels of NT-proBNP were increased in LHF patients combining pulmonary hypertension, the changing trend elevated with the increased pulmonary hypertension accordingly.

Objective:To investigatethe transdermal delivery characteristics of methylphenidate hydrochloride (MPH) in vitro. Methods: Characteristics of MPH crossing nude rats skin were studied with Franz diffusion cells. A high performance liquid chromatographic (HPLC) method was established to determine the concentration of MPH crossed the skin. The permeability coefficient (P), steady state flux (J) and lag time(LT) for MPH through the skin of nude rats treated with various enhancers were compared with those of control. Objective:To study the inhibited function of CTLA4 Ig on T cell activation and its mechanism Methods:After separating fresh PBMC,the effects of CTLA4 Ig on T lymphocyte transformation and mixed lymphocyte reaction were analyzed,and its inhibitory effect on the CD25 expression on T cell surface was observed At last,the changes of the nuclear protein in active T cell were analyzed by the EMSA method.Results:CTLA4 Ig has an inhibitory effect on the lymphocyte transformation,MLR and CD25 expression on T cell surface,and it can inhibite the activation of RE/AP binding factor related to IL 2 expression. Conclusion:CTLA4 Ig inhibit the activation of T cell by multipathway,one of the mechanism might be its inhibitory effect on the activation of RE/AP binding factor.
9.Effect of resistance nodulation division efflux pumps on reduced susceptibility to tigecycline in carbapenem-resistant Acinetobacter baumanii

Objective To investigate the effect of resistance nodulation division (RND) efflux pumps on reduced susceptibility to tigecycline in carbapenem-resistant Acinetobacter baumanii.Methods Totally 631 isolates of Acinetobacter baumanii were collected from 16 hospitals in 7 provinces in 2010.Genes oxa-51 and oxa-23 were detected by PCR method,and the ST profiles were determined by multilocus sequence typing (MLST).The disk susceptibility assay was used to determine the inhibition zone diameters of β-lactams,aminoglycosides,macrolides,tetracyclines,carbapenems,tigecycline and polymyxin.The minimum inhibitory concentration (MIC) of tigecycline was determined by E-test in strains with inhibition zone diameters ≤ 12 mm on tigecycline.The expression of operon genes adeB,adeG and adeJ was determined with efflux pump inhibitor NMP (N-methyl-2-pyrrolidone) for detection of efflux pump inhibitor phenotype.The isolates of Acinetobacter baumanii which were resistant both to tigecycline and carbapenems and with the inhibited phenotype of efflux pump inhibitor were collected as the experiment group,the isolates which were susceptible to tigecycline but resistant to carbapenems were collected as the control group,and ATCC 19606 was used as the reference strain.The expressions of adeABC,adeFGH and adeIJK were quantified by q-PCR at the transcriptional level.Genes adeR,adeS and adeL were amplified and sequenced using PCR method to find polymorphic locus and insertion sequences.Results There were 32 isolates of Acinetobacter baumanii with reduced susceptibility to tigecycline and carbapenem-resistant.Eight isolates were with the inhibited phenotype by efflux pump inhibitor.And 4 strains which were susceptible to tigecycline but resistant to carbapenems were selected as the control.The expressions of adeABC in A518,Z1219 and A527 of experiment group were 13-fold,5-fold and 7-fold higher than reference strain ATCC19606,respectively.The expressions of adeFGH and adeIJK were up-regulated slightly in some isolates.Transcript of adeABC was not found in control group strains A207 and A1731,and the expressions of adeABC,adeFGH and adeIJK were not up-regulated in other isolates.The single nucleotide polymorphism (SNP) was detected in adeR (E220K) and adeS (A130D),respectively.ISAab1 insertion sequence was identified in adeS of adeABC-over expressed isolates.No mutation was found in adeL.Conclusion High expression of adeABC pump may play an important role in tigecycline resistance in carbapenem-resistant Acinetobacter baumannii,mainly due to the insertion of ISAba1 sequence in its regulator gene adeS,but other mechanism of tigecycline resistance may not be excluded.
10.GSTM1 polymorphism,smoking,alcohol drinking and oral cancer susceptibility in Asians:A meta-analy-sis

Objective:To evaluate the association of glutathione S-transferase M1 (GSTM1 )polymorphism,smoking and alcohol drinking with oral cancer susceptibility in Asians by meta-analysis.Methods:A literature search of PubMed,Chinese BioMedical, Wanfang,VIP and CNKI databases from inception to July 30th,2013 was conducted.Crude odd ration (OR)with 95% confidence intervals (95%CI)was calculated.Results:27 case-control studies were assessed,and the results showed that the association be-tween GSTM1 null genotype and oral cancer susceptibility was significant in Asians (OR=1.31,95%CI:1.18-1.45,P<0.001), especially in South Asians (OR=1.35,95%CI:1.20 -1.52,P<0.001).Significant associations between oral cancer risk and smoking and alcohol drinking were found in East Asians (Smoking:OR=1.70,95%CI:1.36 -2.13,P<0.001;Alcohol drink-ing:OR=1.54,95%CI:1.24 -1.90,P<0.001).Conclusion:GSTM1 null genotype may be associated with increased oral cancer risk in Asians.Smoking and alcohol drinking confer significant susceptibility to oral cancer in East Asians.
