1.Effects of interleukin-1βon MMP-13 expression in rat chondrocytes and its regulation of miR-27b

Objective To observe the effect of interleukin-1β(IL-1β) on expression of Matrix Metalloproteinases 13 (MMP-13) in rat chondrocytes and its regulation of miR-27b. Methods Chondrocytes were extracted from 7 Wistar male rats. Expression of MMP-13 were examined by Western blot at 0 h, 24 h, 48 h after IL-1βstimulation. Differential miRNAs expression profiles were examined by miRNAs microarray. The most obviously down-regulated miRNAs were confirmed by quantitative Real-time PCR. Targeted regulation relationship between miR-27b and MMP-13 was set up by Luciferase re?porter gene experiments. Results Expression of MMP-13 in rat chondrocytes was increased at a timely dependent manner upon IL-1βstimulation(P<0.05);Microarray revealed 36 miRNAs whose expression changed, among which 6(miR-27b, miR-31, miR-26a, miR-26b, miR-23, miR-204)were especially obvious. Real-time PCR confirmed that miR-27b was the one whose expression level were most down-regulated. Transient co-transfection of miR-27b mimics with luciferase expres?sion plasmids resulted in significant repression of luciferase activity in rat chondrocytes (P < 0.05). Conclusion IL-1βstimulation result in down-regulation of miR-27b and up-regulation of MMP-13 expression. MiR-27b and MMP-13 show targeted regulation relationship.

3.Comparison of two extraction techniques for larger chemically aceilular nerve allografts

[Objective]To compare two preparing procedures for larger chemically acellular nerve allografts (CANA). [Methods]The sciatic nerves of pigs were exposed by a muscle-splitting incision and were isolated free of the underlying fascia. The 60-mm-long segments of the nerve were obtained. They were treated according to the following decellularization processes.In group I,the nerve segments were treated with 7% Triton-100 solution and 7% sodium deoxycholate for two times.In group II,another protocol was created with the detergents Triton X-200,sulfobetaine-16,and sulfobetaine-10 for two times. The degrees of decellularization,activity of laminin,degrees of demyelination,and integrity of the nerve fiber tube were observed under microscope and were assessed by a scoring system.[Results]In both experimental groups the activity of laminin was present and the degrees of decellularization were complete. As for the demyelization of the nerve segments,the myelin sheath in Group II was partially preserved,but it completely disappeared in Group Ⅰ. The structure of the nerve fiber tube in Groups Ⅰ and Ⅱ were not as integral as that in the normal group.[Conclusion]It may be a better method for the larger CANA,to be treated with TritonX-100 and sodium deoxycholate during the decellularization procedure.

6.A further discussion on acupuncture treatment plan of facial neuritis in Evidence-based Guidelines of Clinical Practice in Acupuncture and Moxibustion.

The treatment plan of facial neuritis in Evidence-based Guidelines of Clinical Practice in Acupuncture and Moxibustion (2011 edition) is discussed, and case information of facial neuritis during the recent five years in department of acupuncture and moxibustion, PLA General Hospital, is retrospectively analyzed. In accordance with anatomy of the facial nerve to form the acupuncture prescription, the detailed diagnosis and treatment method for facial neuritis are introduced. The advantages of the diagnosis and treatment method for facial neuritis are summarized, hoping to establish a more comprehensive, standardized and unified treatment plan.

8.Relationship between homocysteine and the levels of plasma folic acid, vitamin B_(12) and lipid in patients with acute cerebral infarction

Objective To investigate the differences of the plasma homocysteine (Hcy) between the patients with acute cerebral infarction and the normal controls, and the relationship between the levels of plasma Hcy and folic acid, vitamin B_(12) and lipids, and the relationship between clinical symptoms in patients with acute cerebral infarction. Methods A total of 91 patients with first-ever acute cerebral infarction and 100 controls without cerebrovascular accident were included. Their ratio factors such as age, sex, hypertension and diabetes did not have significant differences. Enzymatic cycling was used to detect plasma Hcy levels. Chemiluminescence was use to detect folic acid and vitamin B_(12) levels. Biochemical analyzer was used to measure lipid levels. The patients with acute cerebral infarction were evaluated by the National Institutes of Health Stroke Scale (NIHSS). Results The plasma Hcy level was significant higher in the acute cerebral infarction group than that in the control group (21.22 ±7.29 μmol/L vs. 13.19 ± 2.13 μmol/L) (P < 0.05); the plasma Hcy level in the acute cerebral infarction group was significantly negatively correlated with folic acid (r = - 0.307,P <0.05) and vitamin B_(12) (r = - 0.270, P <0.05). It was significantly positively correlated with low-density lipoprotein (r =0.282, P <0.05), and it was significantly negatively correlated with high-density lipoprotein (r = -0.219, P <0.05). The mean value of the plasma Hcy in acute cerebral infarction group increased with the increase of NIHSS scores. Conclusions Hyperhomocysteinemia is an independent risk factor for acute cerebral infarction. The levels of folic acid and vitamin B_(12) decreased with the increase of Hcy levels. The higher the Hcy levels,the more serious the clinical symptoms are. Hcy may increase the risk of ischemic cerebrovascular disease by influencing the lipid metabolism.
9.Evaluation on clinical efficacy of hyperbaric oxygenation in treatment of sudden deafness :A Meta-analysis

Objective:To evaluate the clinical efficacy of treatment of sudden deafness using hyperbaric oxygenation, and to provide guidance for its treatment. Methods:The studies about sudden deafness treated with hyperbaric oxygenation combined with drug were retrieved in Cochrane Library, PubMed , MEDLINE, Wanfang Database and CNKI up to July 2016.RavMan 5.3 provided by Cochrane was used for Meta-analysis.Results:Thirty-nine studies which were randomized controlled trials (RCTs)were included, a total of 4 599 cases,of which 2 418 cases received hyperbaric oxygenation therapy combined with drug treatment as treatment group and 2 181 cases received drug treatment alone as control group.A total of 36 studies about the comparison of clinical efficacy between treatment group and control group were included in the Meta-analysis, the result showed that treatment group had a significant clinical efficacy than control group(RR:1.26,95%CI:1.22-1.30,P<0.0001).A total of 6 studies about the improvement of the pure tone average(PTA)in the two groups were included in the Meta-analysis, the PTA gain was significant better in hyperbaric oxygenation group(WMD:11.8,95%CI:4.77-18.83,P<0.000 1).A total of 11 studies about the stage of using hyperbaric oxygenation therapy were included in the Meta-analysis, the result suggested that the early use of hyperbaric oxygenation therapy had better effect than the late use of hyperbaric oxygenation (RR:1.42,95%CI:1.31-1.53,P<0.000 01).Conclusion:The clinical efficacy of hyperbaric oxygenation combined with drug therapy is better than drug therapy alone, the PTA gain is significantly improved and the introduction of hyperbaric oxygenation therapy at the earlier stage shows better results.
10.Discriminatory Potential of The Chinese version of ADAS-Cog for Mild versus Moderate Alzheimer Disease

Objective:To examine the discriminatory potential of the ADAS-Cog, Chinese version for mild and moderate Alzheimer's disease (AD). Methods:Patients with AD (199 mild AD and 106 moderate AD) meeting the NINCDS-ADRDA criteria of probable AD were recruited. The Chinese version of ADAS-Cog was administered to all AD subjects. Results:The total score of ADAS-Cog and the score of each ADAS-Cog item differed significant-ly between mild and moderate AD groups, with higher scores in moderate AD group. GLM analysis showed insignifi-cant influence of age or educational level on the ADAS-Cog total score. These results indicated that ADAS-Cog could represent the distinctive profiles of cognitive impairment between mild and moderate AD. The results of Logis-tic regression analysis showed that the item score of orientation and constructional praxis as well as the ADAS-Cog total score could classify mild and moderate AD efficiently, with a sensitivity of 78% - 82% and a specificity of 70% - 73%. Conclusion:Our results indicate that the Chinese version of ADAS-Cog is useful for staging of AD. It is recom-mended that the Chinese version of ADAS-Cog be introduced for monitoring the AD drug therapeutic efficacy on cognitive impairment among Chinese AD patients.psychiatry Alzheimer's disease
