1. Effect of autophagy-related gene Beclin 1 on growth of xenografts of human lung adenocarcinoma A549 cells in nude mice

Objective: To investigate the effect of autophagy-related gene Beclin 1 on growth of xenografts of human lung adenocarcinoma A549 cells in BALB/c nude mice. Methods: The recombined plasmids pRNAT-U6.2/Lenti-si423 and pLenex-Beclin 1 were transiently transfected by lipofectin regeant into A549 cells, respectively. The expression levels of Beclin 1 mRNA and protein in A549 cells were detected by real-time fluorogenic quantitative-PCR (RFQ-PCR) and Western blotting, respectively. The proliferation rate of A549 cells transfected with recombinant vector pRNAT-U6.2/Lenti-si423 or pLenex-Beclin 1 was determined by MTT assay. The A549 cells expressing Beclin 1 or with Beclin 1 silencing were subcutaneously injected into right axillary region of nude mice. The growth rate and size of xenograft tumor were observed, and the expression levels of Beclin 1 mRNA and protein in xenograft tumor were determined by RFQ-PCR and immunohistochemistry, respectively. Results: The expression levels of Beclin 1 mRNA and protein were increased in A549 cells transfected with recombinant vector pLenex-Beclin 1, and the cell proliferation in vitro was inhibited. The expression levels of Beclin 1 mRNA and protein were decreased in A549 cells transfected with recombinant vector pRNAT-U6.2/Lenti-si423. As compared with Beclin 1-silencing group, the growth rate of subcutaneous xenograft was slowing down with lower weight and smaller volume and the relative expression levels of Beclin 1 mRNA and protein were both higher in Beclin 1-overexpression group. Conclusion: Autophagy-related gene Beclin 1 can inhibit the growth of xenograft of human lung adenocarcinoma A549 cells in nude mice, and it may become a new target for tumor therapy. 2.Clinical study on taxol plus 5-fluorouracil/leucovorin combination chemotherapy for patients with advanced gastric cancer

Objective The current study was designed to evaluate the efficacy and toxicity of taxol plus fluorouracil(TF)chemotherapy in patients with advanced gastric cancer.Methods 55 patients with ad- vanced gastric cancer were treated with TF schedule.A treatment cycle lasted 21 days.The efficacy was eval- uated after 2~3 cycles of the chemotherapy.Results These 55 patients were evaluated the efficacy and the toxicity.The total efficiency rate of TF schedule was 45.45 %.Clinical Benefit Response(CBR)were evaluated in 35 patients.Ache remission rate was 86.6 %.The main side effect of toxicity was neutropenia.The total re- sponse neutropenia is 56.4 %.The aboveⅢstage was 10.9 %.The rate of nausea and vomiting was 43.6 %. Conclusion The combination of taxol and fluorouracil is a high response protocol with mild toxicity.TF schedule has high efficiency for advanced gastric cancer,especially for the patients who had had chemothera- py with fluorouracil.
3.Treating Severe Pseudobulbar Palsy with Acupuncturing Lianquan Point Combined with Rehabilitation

Objective To observe the effect of acupuncturing Lianquan point combined with rehabilitation on severe pseudobulbar palsy.Methods Sixty patients with severe pseudobulbar palsy were divided into the control group and therapy group with 30 cases in each group. The patients of the control group were treated with medication, nasal feeding and vein nutrition. The patients of the therapy group were added with acupuncturing Lianquan point combined with rehabilitation besides methods the control group used. After two periods of treatment, the effects of two groups were compared.Results The overall efficient rate was 96.7% in the therapy group, and 73.3% in the control group, the former was obviously superior to the latter ( P<0.05).Conclusion Acupuncturing Lianquan point combined with rehabilitation can improve the therapeutic effect of severe pseudobulbar palsy.
6.The construction of driver' s personality trait model of armed police force

Objective To investigate the personality characteristics of the drivers in the Armed Police Force ( CAPF), as well as construct a personality traits model for drivers and evaluation criteria. Methods Adopting Sixteen Personality Factor Questionnaire(16PF) to collect data from 1187 CAPF drivers in Beijing,Sichuan,Inner Mongolia and Yunnan. Results The paper concludes that career personality traits of CAPF drivers are made up of 4 factors: stability and self-control, self-motivation and cooperation, fantasy and openness, intelligence and candor, which explain 50.13% of the total variance. Conclusion The four factors basically construct the personality trait model of CAPF drivers. They could also be of great significance in recruiting, training and evaluating drivers.
9.Transvaginal ultrasound guided methotrexate injection for unruptured early tubal pregnancy

2 000 mIU/ml,combined use of mifepristone(100 mg,twice daily,for 3 days) was employed. Results The treatment succeeded in 28 patients,with a success rate of 87.5%.Acute lower abdominal pain occurred in 4 patients and laparoscopic salpingectomies were carried out.The serum ?-hCG levels decreased by 23%~35% at 3 days after injection,by 38%~62% at 1 week,and to normal levels(≤6.1 mIU/ml) at 2~4 weeks.The mass completed subsided at 30~55 days after injection. Conclusions Transvaginal ultrasound guided methotrexate injection for the management of unruptured early tubal pregnancy is simple,safe, and effective.Further investigations should be carried out in respect of the recovery of tubal anatomy and physiological functions.
