Clinical Medicine of China 2012;28(10):1053-1055
Objective To observe the changes and clinical significances of ischemia modified albumin (IMA),cardiac triponin T (cTnT) and creatine kinase MB (CK-MB) in transient myocardial ischemia induced by radio frequency ablation ( RFA ) in patients with arrhythmia. Methods One hundred and twelve patients with arrhythmia undergoing RFA and 91 patients undergoing CAG were evaluated with blood levels of IMA,cTnT and CK-MB before and 3 and 6 hours after operation. Results Serum levels of IMA at 3 h [ (80.62 ± 7.53 )U/ml ] and 6 h [ (76.37 ± 9.21 ) U/ml ] after operation rose significantly (P < 0.05 ) in the 112 cases of RFA compared with the preoperative [ ( 45.42 ± 7.00 ) U/ml ].While the levels of IMA were not significantly different at 3 and 6 hours after CAG in the 91 patients compared with before sugery ( P > 0.05).The levels of cTnT and CK-MB remained unchanged between the two groups after the procedures ( P > 0.05 ).Conclusion The levels of IMA were quickly increased after myocardial ischemia induced by RFA and this change occurred earlier than that of cTnT.IMA is an early diagnostic biochemical marker of myocardial ischemia.

Objective To determine the expression of Notch pathway receptors (Notch1 and Notch4) and ligands (Jagged1 and Dll4) in cutaneous malignant melanoma (CMM) tissues, and to preliminarily explore the role of the Notch signaling pathway in the pathogenesis of CMM.Methods Immunohistochemical study was performed to determine the expression pattern and intensity of Notch1,Notch4,Jagged1 and Dll4 in 40 paraffin-embedded CMM specimens and 15 paraffin-embedded pigmented nevus specimens.Statistical analysis was carried out by chi-square test and Spearman rank correlation analysis with the SPSS 21.0 software. Results Notchl was detected in 31 (77.5%) of 40 CMM specimens,as well as in 3 of 15 pigmented nevus specimens, and the positive rates significantly differed between the two groups (x2 =15.281,P < 0.001).However,no significant difference in the expression intensity of Notch1 was observed between 18 in situ melanoma tissues and 22 invasive melanoma tissues (x2 =0.631,P =0.427).In addition, the positive rates of Notch4,Jagged1 and Dll4 were also significantly higher in the CMM group than those in the pigmented nevus group (all P < 0.05),and the expression intensity of Notch4,Jagged1 and Dll4 significantly differed between in situ and invasive melanoma tissues (all P < 0.05).In CMM tissues, the expression of Notch1 was positively correlated with that of Jagged1 (rs =0.350,P =0.027) and Dll4 (rs =0.562,P < 0.001),while the expression of Jaggedl was negatively correlated with that of Dl14 (rs =-0.734,P < 0.001).Conclusion Abnormality of the Notch signaling pathway may be involved in the pathogenesis of melanoma, but further researches are still needed to elucidate the detailed mechanism.

Objective To discuss the effect of daidzein on hepatocellular carcinoma SMMC7721 cell proliferation CD133 expression on tumor stem cell. Methods Hepatocellular carcinoma SMMC7721 cells were cultured, digested and passaged, and divided into six groups with different drug:control group with no daidzein,100 μg/mL daidzein group,200μg/mL daidzein group,300μg/mL daidzein group,400μg/mL daidzein group ,500μg/mL daidzein. The inhibition ratio, hexokinase, alkaline phosphatase and CD133 levels in SMMC7721 cell were detected and compared at 24 h,48 h,72 h among those groups. Results The inhibition ratio was increased by Daidzein dose increasing, and decreased apparently by times extending, especially in 400μg/mL and 500μg/mL daidzeingroups.Compared with control group, the hexokinase and alkaline phosphatase activity and CD133 expression were decreased apparently in groups treated with daidzein(P<0.01).The more the dose, the higher the drop(P<0.01).Conclusion Daidzein can inhibit hepatocellular carcinoma SMMC7721 cell proliferation, and inhibit CD133 expression on tumor stem cell.
