2.The pathogenesis of ischemic preconditioning to warm ischemia reperfusion injury of hepatocytes in rats

Objective To explore the pathogenesis of ischemic preconditioning to warm ischemia reperfusion injury of hepatocytes in rats. Methods Ninety SD rats were randomly divided into three experimental groups:sham operation group (group A), warm hepatic ischemia/reperfusion group(group B and group C). Group C was given ischemic preconditioning treatment. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were detected 0 h, 2 h, 6 h, 12 h and 24 h after ischemia reperfusion injury. Levels of TNF-α and IL-1β were tested detected by ELISA. Levels of malondialdehyde (MDA) and superoxide dismutase (SOD) of hepatocytes were detected at the same time points. Mitochondrial membrane potential was examined to assess ischemia reperfusion injury of hepatocytes in rats using chart of intensity of JC-1 in mitochondria. Results The serum levels of ALT, TNF-α, IL-1β, and MDA were significantly higher in hepatic warm ischemia reperfusion group and ischemic preconditioning group than those in sham operation group (P<0.05). Values of prothrombin activity and cholinesterase were significantly lower in liver warm ischemia reperfusion group and ischemic preconditioning group than those of sham operation group (P<0.05). The SOD level of liver was significantly lower in warm ischemia reperfusion group and ischemic preconditioning group than that in sham operation group. The indexes were better in ischemic preconditioning group than those of warm ischemia reperfusion group (P<0.05). The mitochondrial membrane potential level of liver cells reached the lowest value 0 hours after ischemia and reperfusion, and then increased gradually within 24 hours (P<0.05). And the level of mitochondrial membrane potential of liver cells was significantly higher in ischemic preconditioning group than that in warm ischemia reperfusion group (P<0.05). Conclusion Ischemic preconditioning may play a protective role in warm ischemia-reperfusion injury in rats. Ischemic preconditioning may significantly decrease the levels of ALT, AST, TNF-α, IL-1βand MDA, and increase the SOD activity in hepatocytes. Thedamage of mitochondrial membrane potential is decreased after ischemic preconditioning, which might be the pathogenesis of ischemic preconditioning to warm ischemia reperfusion injury of hepatocytes in rats.
3.Distinguishing value of serum aldosterone level for patients with different types of atrial fibrillation

Objective:To explore distinguishing value of serum aldosterone level for different types of atrial fibrilla-tion (AF) .Methods:A total of 153 AF patients were selected ,including 99 patients with paroxysmal AF (paroxys-mal AF group) and 54 patients with persistent AF (persistent AF group) .Serum aldosterone concentration was measured ,and its correlation with two types of AF was analyzed .Results:Compared with patients with paroxysmal AF ,there was significant rise in serum aldosterone level [ (234.3 ± 69.6) pg/ml vs .(302.7 ± 78.3) pg/ml , P<0.01] in patients with persistent AF .Correlation analysis indicated that aldosterone level was positively correlated with atrial diameter and left ventricular ejection fraction (r=0.896 ,0.831 ,P<0.01 both) .Conclusion:Serum al-dosterone level is helpful to distinguishing paroxysmal atrial fibrillation and persistent atrial fibrillation ,and posses-ses certain guiding significance for outcome .
4.Application of moving average method in the quality control of coagulation function measurement

Objective To discuss the feasibility of moving average method (X‐B method) in the quality control of coagulation function measurement .Methods Fluctuation average value tendency of prothrombin time (PT ) ,activated partial thromboplastin time (APTT) ,thrombin time (TT) and fibrinogen (FIB) of clinical specimen ,detected by using Stago Compact analyzes ,were ana‐lyzed ,and compared cryochemic plasma method .Results Average value of PT ,APTT ,TT and FIB changed in small ranges .Tend‐ency of quality control chart of X‐B method and cryochemic plasma method were basically consistent .Conclusion X‐B method ,u‐sing plasma of patients ,could be used for the quality control of PT ,APTT ,TT and FIB examination and for the monitoring of the stability of analyzers .

6.Study on clinical anatomy of the accessory nerve,the great auricular nerve and the transverse nerve of neck in neck levelⅡ-Ⅳ

OBJECTIVE To study the anatomicallandmarks of the accessory nerve (AN) , the great auricular nerve (GAN)and the transverse nerve of neck (TN) in neck level Ⅱ-Ⅳ and provide a reliable surgical basis for treatment of functional neck dissection in neck level Ⅱ-Ⅳ. METHODS The courses of the AN, GAN and TN were studied in 42 patients (56 sides) who underwent lateral neck dissection. Meanwhile, the correlations among the three nerves above and their surrounding structures were observed. RESULTS After emerging from the inferior border of the posterior belly of digastric muscle, the AN descends obliquely downward and backward before the transverse process along with the sternocleidomastoid branch of occipital artery to the upper part of the deep surface of the sternomastoid muscle. The distance from the mastoid process to the point where AN enters the sternocleidomastoid muscle is (4.93?0.75) cm. The point where AN emerges from sternocleidomastoid muscle (point A) located above the point where the GAN exits the sternocleidomastoid muscle (point B). The distance between the two points is (1.04?0.59) cm while the distance from point A to the middle point of clavicle (point C) is (8.09?0.65) cm. The GAN leaves the cervical plexus at the posterior border of the sternocleidomastoid muscle and run anteriorly between the lateral surface of this muscle and the deep surface of platysma. It is (6.37?0.73) cm long from point B to point D where the GAN send out posterior aurem branches, auricular lobule branches and anterior auricular branches. The distance from point B to point C is (7.67?1.00) cm. Most of GANand external jugular vein run nearly in parallel and the distance between them is (1.02?0.61) cm. The point where the TN emerges from the posterior border of the sternocleidomastoid muscle is below point B. Then the TN which is below the external jugular vein runs almost horizontally over the surface of the sternocleidomastoid muscle to sector branches in the lateral neck and part of its branches are distributed in the lateral neck after going across the anterior jugular vein. 7.Effect of Some Amino Acids in Gentamicin Production

Gentamicin production at home and abroad was fermentation cycle long,production rate low,production costs high and short of competitiveness.Added a little quantity of lysine,glycine,tyrosine,methionine into the medium of gentamicin producing Micromonospora purpurea during inoculation could improve the metabolic capability of microbe,shorten fermentation cycle and increase production rate of gentamicin.The new method was successfully developed in shaking flask test and applied to 5L glass fermentor for one month.Average data of one month showed that the fermentation time per cycle was shortened about 40% and the yield of gentamicin per fermentor increased about 14% in comparison with the conventional technique.The production rate of gentamicin was increased 30% ~95%(according to the capability of strain).The quality of the end product gentamicin conformed to CP2000,BP2000 and USP26 pharmacopoeia.Therefore,the new method may reduce production costs of gentamicin remarkably,it shows strong competitiveness in the marketplace.
8.The visual cortex plasticity of adult rat after binocular form deprivation by pattern visual evoked potential

Background The visual evoked potential (VEP) is an important functional index to assess visual cortex plasticity.Monocular form deprivation(MD) of rat is a classic model to study visual cortex plasticity change.Utilizing VEP technique record the shift of ocular dominance in adult rats model is of an important significance for explore the treating opportunities of amblyopia.Objective The present study was to observe the pattern VEP changes in different ages of Long-Evens rats and the adult rats after binocular form deprivation.Methods Thirty-six healthy SPF Long-Evens rats were divided into PW3,PW4,PW5,PW6 groups according to the postnatal weeks,and PW7 rats included 36 SPF Long-Evens rats.The left eye lids of the rats were sutured in PW3,PW6 and PW7 rats for 3,5,7 days respectively,and PVEP in both left and right eye were recorded to assess the rat age that visual cortex plasticity ended.Bilateral PVEP were recorded in PW7 rats,and the bilateral eyelids were sutured to establish the bilateral form deprivation models.The right eyes were opened in 7,10,14 days,and the left eyes were opened in the next 3,5,7 days respectively and the PVEPs were recorded again to find the shift of ocular dominance and whether binocular form deprivation induce the visual cortex plasticity in adult rats.Results The P100amplitudes of the left eyes were gradually declined and those of the right eyes were raised from 3 to 7 days after MD of the left eyes in rats of PW3 group in comparison with before MD ( P<0.01 ).In PW6 groups,no significant changes in P100amplitudes of both the right and left eyes were found in the third day after MD,but significant raise in the right eyes and lowing in the left eyes were seen in the 5 and 7 days after MD in comparison with before MD(P<0.01 ).No any P100changes in both eyes were found in 3-7 days after MD compared with before MD in PW7 rats (P>0.05).The C/I ratio (contralateral VEP amplitude to ipsilateral VEP amplitude in occluded eyes) in the rats of PW3,PW4 and PW5 groups were 1.07±0.15,1.16±0.16 and 1.14±0.15 respectively in 3 days after MD,showing the significant lowing in comparison with before MD (2.69±0.45,2.58±0.4,2.62±0.32) (P<0.01),but those of PW6 and PW7 were unchanged (2.80±0.48 vs 2.90±0.46,2.59 ±0.36 vs 2.90±0.46,P>0.05 ),indicating the absence of ocular dominance plasticity in the adult rats.In rats of PW7 with binocular form deprivation for 14 days,a significant decrease was observed in the C/I ratio in rats with next MD for 3 days,demonstrating that the visual cortex plasticity was reactivated after 14 days of binocular form deprivation in PW7 rats( 1.33±0.18 vs 2.70±0.45,P<0.01 ).Conclusions PVEP can be recorded in Long-Evens rats.It is a major index for identifying the shift of ocular dominance in the Long-Evens rats.Binocular form deprivation can reflect the visual cortex plasticity in the adult rats.
9.Effect of adipose-derived stem cells modified by insulin-like growth factor I gene on TWEAK/Fn14 signaling pathway

BACKGROUND:Because insulin-like growth factor I has the ability to induce mesenchymal stem cells into chondrocytes, we hypothesized that the chondrogenic diferentiation of adipose-derived stem cels can be improvedvia insulin-like growth factor I transfection. OBJECTIVE:To investigate the influence of insulin-like growth factor I transfection on chondrogenic potential of adipose-derived stem cels in vitroand tumor necrosis factor-like weak inducer of apoptosis (TWEAK)/fibroblast growth factor-inducible 14 (Fn14) signaling pathway. METHODS:Recombinant lentivirus plasmid pLVX-IGF-I-IRES-ZsGreenl was constructed and transferred into passage 3 adipose-derived stem cels, and then these stem cels were induced to diferentiate into chondrocytes (experimental group 1). Meanwhile, the cels transfected with pLVX-IRES-ZsGreenl were taken as green fluorescent protein/adipose mesenchymal stem cel group (experimental group 2), and those with no transfection acted as control group. RESULTS AND CONCLUSION:The mRNA expression of TWEAK was reduced in the experimental group 1 as compared with the other two groups, but the mRNA expressions of insulin-like growth factor I, Col2a1 and Sox9 were up-regulated in the experimental group 1. At the same time, the protein expression of matrix metaloproteinase-3 and TWEAK were down-regulated, while the protein expression of Col2a1 was increased in the insulin-like growth factor I-transfected cels in contrast to the cels modified with pLVX-IRES-ZsGreenl or with no transfection. These findings indicate that pLVX-IGF-I-IRES-ZsGreenl transfection of adipose-derived stem cels results in a higher expression of insulin-like growth factor I, and down-regulates the expression of TWEAK mRNA and protein, which improves the diferentiation of adipose-derived mesenchymal stem cels into chondrocytes.
10.Human adipose-derived mesenchymal stem cells from the infrapatellar fat pad:isolation, culture and identification

BACKGROUND:Infrapatelar fat pad is often partialy resected in the knee surgery, which can be used as an important source of adipose-derived mesenchymal stem cels. OBJECTIVE: To explore the strategies of isolation, culture, and identification of adipose-derived mesenchymal stem cels from the infrapatelar fat pad and to detect the expression of cel surface markers of human adipose-derived stem cels. METHODS: Infrapatelar fat pad was obtained from patients undergoing knee arthroscopy surgery, and attached cels were obtained from adipose tissue by using colagenase I. Cels were cultured in 10% low-sugar DMEM. Stem cels proliferation was detected by means of MTT and then, cel growth curve was made. The obtained cels were induced and differentiated into adipocytes and osteocytes. Expressions of cel surface markers CD29 and CD44 were detected. RESULTS AND CONCLUSION:A few of attached cels were observed after cultured 24 hours. Cels proliferated faster and exhibited spindle shape after 1 week. Cel adherence and proliferation were speeded up after subculture. Growth curve of cels exhibited that the passages 5 and 2 cels had higher reproductive activity than passage 8 cels. The obtained cels can be induced and differentiated into adipocytes and osteocytes. Results from flow cytometry showed that 96.8% passage 5 cels expressed CD29 and 97.6% expressed CD44. These findings indicate that high-purity adipose-derived mesenchymal stem cels with high reproductive ability are easy to be isolated from the infrapatelar fat pad, which may be a kind of ideal seed cels for cartilage tissue engineering.
