1.Experimental study on mechanism of Sox2 upregulate EGFR and promote selfrenew of glioma stem cell

Objective To observe the regulation of sry-related high-mobility-group box-containing 2 (Sox2) on epidermal growth factor receptor(EGFR)and on the sphere formation rate of glioma stem cells .Methods Promoter reporter plasmids of epidermal growth factor homologous receptor ErbB2 ,ErbB3 ,ErbB4 were respectively constructed .Sox2 expression plasmid was co-transfected together with the reporter plasmid into U251 cells .Then the luciferase activity was analyzed by dual-luciferase reporter assay sys-tem to test the regulation of Sox2 on ErbB2 ,ErbB3 ,ErbB4 promoters .Sphere formation assay was used to observe selfrenew of the cancer stem cells after transfection of Sox 2 .The expression of EGFR and ErbB2 proteins in the spheres was examined by Western blot .Results Sox2 could dose-dependently increase the ErbB2 ,ErbB3 ,ErbB4 promoter drived luciferase activity .Sox2 promotes the sphere-forming rate of U251 cells and upregulates the expression of EGFR and ErbB2 in the spheres .Conclusion Sox2 could up-regulate the expression of EGFR and promote selfrenew of glioma stem cells .

4.Clinical study of thumb-tack needle therapy for cervical radiculopathy based onmeridian differentiation

Objective: To observe the clinical efficacy and eligibility of thumb-tack needle therapy based on meridian differentiation in treating cervical radiculopathy. Methods: A total of 70 patients with cervical radiculopathy were randomized into an observation group and a control group, with 35 cases in each group. Patients in the control group received thumb-tack needle based on conventional point selection, while those in the observation group received thumb-tack needle according to meridian differentiation. The visual analog scale (VAS) and clinical symptom scores in the two groups were compared before and after treatment, and the clinical efficacy of the two treatments was observed. Results: After treatment, the VAS score in both groups dropped significantly (both P<0.01), and the VAS score in the observation group was lower than that in the control group (P<0.01). The clinical symptoms score in both groups dropped significantly (all P<0.01), and the clinical symptoms score in the observation group was lower than that in the control group (P<0.01). The total effective rate in the observation group was higher than that in the control group (P<0.05). Conclusion: Thumb-tack needle therapy based on meridian differentiation can reduce pain score, improve clinical symptoms in patients with cervical radiculopathy, and produce more significant efficacy compared with conventional thumb-tack needle therapy.

8.Production and Teaching Application of Simulated Pulmonary Air Embolism Demonstration Model

Objective To design simulated pulmonary air embolism demonstration model,so as to solve the problem of pathological anatomy of pulmonary air embolism in experiment teaching. Methods According to the principle of the disturbance of local blood circulation and air embolism, we designed a pulmonary air embolism model. We took 223 school nursing students as the object of this study,and randomly divided them into 2 groups:animal experiment teaching group and model control group,then we compared the teaching effect between the two groups. Result The test scores of students in the animal experiment teaching group were higher than control group (P<0.05) . Conclusion The use of simulated pulmonary air embolism demonstration model teaching can improve the students’experimental test scores,and can be repeatedly used,stimulate students' study interest,reduce the cost of teaching,and improve the teaching quality.

Objective To investigate the effects of ischemic postconditioning (Post) on myocardial cell apoptosis and expres- sion of Bcl-2 and Bax protein during ischemia/reperfusion period in rabbits.Methods Eighteen rabbits were randomly allocated to three groups (6 in each group),sham operation (group S),ischemia/reperfusion group(group IR) and ischemic postconditioning group(group Post).Group IR and group Post were subjected to 15 minutes of left anterior descending coronary artery occlusion followed for 30 minutes of reperfusion.Ischemic postconditioning was achieved by three 30 seconds cycles of reperfusion,each followed by 30 seconds ischemia.Cardiomyocyte apoptosis were determined by in situ TDT-mediated dUTP nick end labeling (TUNEL) and DNA electrophoresis.The expression of Bcl-2 and Bax proteins in apoptotic myocardial cells were detected by immunohistochemistry sepa- rately.Results Compared with group IR,apoptotic index was significantly reduced in group Post [(28.06?2.92) % vs.(55.70? 13.96)%,P
10.Protective effect of rosiglitazone on liver injury in rats with severe acute pancreatitis

Objective To explore the therapeutic effects of peroxisome proliferator activating receptor γagonist-rosiglitazone on HMGB1 expression in liver tissue of rats with SAP. Method A hundred and twenty Wistar rats were randomly (random number) divided into the sham operation group(SO group, n = 20) ,SAP group ( n=80) and ROSI treatment group (n =20). SAP group were randomly further divided into the 3 h, 6h, 12 h and 24h subgroups with 20 rats in each group. SAP model was made by retrograde injection of 5 % sodium deoxycholate into the biliopancreatic duct. The serum amylase, AST and ALT, and pathological scores of pancreas and liver tissue were observed. The expression of NF-κB mRNA and the level of HMGB1 protein were investigated respectively by Reverse transcription polymerase chain reaction (RT-PCR) and Westem blot method, respectively. SPSS 16.0software was used to make one-way ANOVA, q -test and correlation analysis. Results Serum amylase, AST and ALT, and pathological scores of pancreas and liver tissue, and the level of HMGB1 protein were markedly increased in each subgroup of SAP compared with SO group ( P < 0.01). The level of HMGB1 protein was positively correlated with the changes of AST, ALT and pathological scores of pancreas and liver tissue. Correlation was not found between HMGB1 and amylase. Treatment with ROSI could significantly reduce the expression of NF-κB mR-NA and the levels of HMGB1 protein, serum AMY, AST and ALT, and pathological scores of pancreas and liver tissue in comparison with 24 h subgroup of SAP (P <0.01). Conclusions As a late-acting mediator of inflammation, HMGB1 was involved in the pathophysiological process of SAP-related liver injury. ROSI can reduce the liver injury by inhibition of the expression of the HMGB1.
