1.Messenger RNA expression of IL-6 and IL-8 in allergen-induced late-phase cutaneous reactions (LPR) in Schneiderian membrane subjects

Objective:To detect mRNA expression of IL-6 and IL-8 in allergen-induced late-phase cutaneous reactions in schneiderian membrane subjects. Methods:Cryostat sections from rhinitis biopsies from 24 h allergen-induced late-phase cutaneous reactions (LPR) in 10human atopic subjects were hybridization with 35S-labeled RNA probes for IL-6 and IL-8.Results:mRNA was detected for IL-6 (9/10) and IL8 (10/10).Compared with the control, there were significant increases in the numbers of ce11 expreasing mRNA expression for IL-6 and IL-8(P<0.05, P<0.01). Conclusion: The augmentation of mRNA expression of IL-6 and IL-8 maybe regarded as the mark of rhinits in IL PR.
2.The relationship of peripheral circulating endothelial cell level and the coronary artery lesion in children with Kawasaki disease

ObjectiveTo investigate the relationship between the change of circulating endothelial cell (CEC) level and coronary artery lesion (CAL) of Kawasaki disease (KD),and to further explore the method for early diagnosis of KD.MethodsThirty KD children were recruited for study,including 23 children with complete type of KD and seven children with incomplete KD.According to the results of echocardiography,the KD group was divided into CAL group (9 cases) and non-coronary artery lesion (NCAL)group (21 cases).Ten healthy children were enrolled as control group.Double-blind and controlled trial was conducted,and Hladovec method was applied for CEC counting.Results The CEC level was ( 1.09 ±0.60) × 107/L in KD group,which was higher than that of control group [ (0.38 ±0.14) × 107/L],and the difference was statistically significant ( t =2.85,P < 0.01 ).The CEC level in the CAL group [ ( 1.84 ± 0.24) × 107/L] was higher than that of the NCAL group[ (2.01 ±0.38) × 107/L],and the difference was statistically significant ( t =2.24,P < 0.05 ).The CEC level was ( 1.16 ± 0.63 ) × 107/L in the complete type of KD group and (0.83 ± 0.45 ) × 107/L in the incomplete KD group,which showed no significant difference between the two groups ( t =1.86,P > 0.05 ).CondusionCEC level was elevated significantly in the acute phase of KD.The CEC level in CAL group was higher than that of NCAL group in acute phase.CEC level detection may be helpful for the early diagnosis of KD.
3.Morphological analysis of irradiated dentin by scanning electron microscope under anhydrous condition

Objective To observe the surface morphology of dentin irradiated by Er,Cr∶YSGG laser under anhydrous conditions and discuss the possible clinical applications of the laser.Methods 11 freshly extracted specimens (approximately 1.5 mm thick) taken from the first premolars were selected.One was regarded as the control group without any treatment and the remaining ten were divided into 5 groups whose dentin surface was irradiated by Er,Cr∶ YSGG laser with different energy (1,2,3,4,6 W) under anhydrous condition.The specimens were routinely prepared for electron microscopic scanning observation.Results As the output power increased with Er,Cr∶YSGG laser irradiation,the dentin surface was observed of the constriction of tubular lumen,and part of the tubules closed.The dentin surfaces with 3 ~6 W laser irradiation were observed thermal injury alters such as micro crack and carbonization.Conclusion 1 ~2 W anhydrous Er,Cr:YSGG laser irradiation play a more ideal and safe constriction and sealing role to dentinal tubules,and the laser is expected to be used for the treatment of dentin hypersensitivity.
4.Observation of the morphology and the distribution of mouse thymus dendritic cells

Objective To investigate the morphology and the distributon of thymus dendritic cells (TDCs) in mouse. Methods TDCs in mouse were observed and analyzed by the electron microscopy and the immunohistochemistry with DCs marker (including CD205, CD1a and MIDC 8). Results There were the most TDCs in the boundary area of thymus cortex and medulla, lesser TDCs in the cortex, and the least in the medulla. TDCs made up to 2 per cent of all cells in thymus. On TDCs surface there were many processes which extended into diastema surrounding lymphocytes. The nucleus was irregular in shape and there were intentations. The cytoplasm was electron lucent and there were few organelles without any lysosme. Conclusion It may effectively count TDCs and display their morphological character that TDC be jointly labeled by the diversiform markers and its ultrastructure observed.
5.The coverage of contracture of the first web space by flaps

Objective To introduce our experiences of reconstruct the first web space using flaps.Methods One hundred and twelve patients sustained contracture of the first, summarized the indications of different flaps in coverage of the first webspace and their prognosis of these flaps were retrospectively reviewed.Results The local flaps, such as double Z-plasty or Z-plasty, were suitable to cover minor contracture of the first webspace; the dorsal flap arising from the index finger was suitable for the coverage of the medium contracture, and the posterior interosseous artery flap or the reverse antebrachial island flap based on the dorsal branch of the anterior interosseous artery suitable for the severe cases. Conclusion The usage of flaps is important to resume the cosmetic shape and function of the first webspace after release of the contracture of the first webspace.
6.Effects of Modified Wendan Decoction Combined with Amlodipine Tablets on TCM Constitution and Vascular Endothelial Growth Factor of Phlegm-damp Hypertension Patients

Objective To observe the clinical efficacy of modified Wendan Decoction combined with amlodipine tablets in treating hypertensive patients with phlegm-damp constitution, and the impact on patients' constitution and vascular endothelial growth factor (VEGF). Methods Totally 126 patients were randomly divided into two groups, 63 cases in each group. The treatment group was given modified Wendan Decoction combined with amlodipine tablets, while the control group received amlodipine tablets, for 12 weeks. The blood pressures, TCM phlegm-damp scores, and serum VEGF levels in both groups were observed before and after treatment. Antihypertensive effect between the two groups was compared. Results TCM phlegm-damp scores decreased significantly in the treatment group after treatment (P<0.01), while that was not significant in the control group (P>0.05). The treatment group was superior to the control group (P<0.01). The systolic blood pressure, diastolic blood pressure, and serum VEGF level significantly decreased after treatment (P<0.01), and the indicators of treatment group were better than those of the control group (P<0.05). The total efficiency of treatment group and control group were 92.1%(58/63) and 76.2%(48/63), respectively, the difference was significant (P<0.05). Conclusion Modified Wendan Decoction combined with amlodipine tablets can significantly improve biased constitution in hypertension patients with phlegm-damp constitution, lower serum VEGF level, and improve endothelial function, with good antihypertensive effect.
7.Clinical and pathological characteristics of multiple system atrophy

Objective To study the clinical,pathological characteristics and diagnosis of multiple system atrophy(MSA).Methods Among 4 cases of MSA,2 of them were from clinical data,the other 2were frum pathological data.Two cases verified by autopsy were investigated using Hematoxylin-eosi,Kltiver.Barrera and Gallvas-Braak silver staining and confirmed by immunohistochemistry using ubiquitin,α-synuclein staining.Results All 4 patients were male.aged 51-76.Case No.1 wag diagnosed as Parkinson's svndrome.Case No.2 was diagnosed as spinal ataxia cerebella,and the other two eases were diagnosed as MSA.The following changes were found by pathological studies.Macroscopic atrophies were presented in pens and cerebella.as well as putamen,globus pallidus and substantia nigra.Cerebral ventricles were dilated.Neuronal lOSS and gliosis could be seen at cerebral cortex.nigrostriatal.globus pallidus,pontine nuclei,subslantia innominata,inferior olives,doral motor nucleus of vagus,cerebellum antl intermediolateral column of the spinal cord.In the white matter of these regions cytoplasmic inclusions bodies were extensively present in oligodendrocytes.Conclusions Olivopontocerebellar atrophy mainly shows the,clinical symptoms of pens,cerebellum,and autonomic nerves damage;Shy-Drager syndrome presents mainly with the erecting hypotenstion symptom,while striatonigral degeneration mainly involves extrapyramidal system.As these three diseases share the common basic pathological changes,they are preferred to be classified as the subtype of MSA.
8.Cortical activations for covert and overt picture naming on fMRI

Objective To investigate cortical activation patterns for covert and overt picture naming with functional magnetic resonance imaging (fMRI). Methods fMRI data were collected on 24-27 years old volunteers during performance of covert and overt picture naming. After statistical postprocessing analysis, head movement data were compared across tasks and average neural activation maps were available for both tasks. Results Mean and maximal translations of head movement in covert picture naming were less than those in overt picture naming, but the difference has no statistical significance (P=0.23). It was shown that covert picture naming involved an orchestration of bilateral occipital gyri and cerebellums, bilateral supplementary motor area, postcentral gyri, inferior frontal gyrus and anterior cingulate cortex. Activations in overt picture naming included those in covert naming (but more intensive), bilateral precentral gyri and posterior superior temporal gyri, left anterior superior temporal gyrus, bilateral thalamus, basal ganglia, and left insula. Conclusion Covert and overt picture naming are two different tasks involving different neural processing networks and levels. They can not be taken as substitutes for each other.
9.HPLC Fingerprint Differentiation of Herba Selaginellae moellendorfii and Amentoflavone Determination

Objective To establish the HPLC fingerprint of Herba Selaginellae moellendorfii for identification and comparison with other species of the same genus and to determine the content of amentoflavone.Method Separation was performed on ZORBAX SB-C18 chromatographic column,acetonitrile(A)-0.5 %solution of acetic acid in water(B)as mobile phase with gradient elution and the flow rate was 1.0 mL?min-1.The detection wavelength was at 270 nm.The content determination of amentoflavone carried out synchronously.Results The characteristic eighteen peaks in the chromatogram consisted of the common pattern of Herba Selaginellae moellendorfii.The fingerprint coupling with similarity and Principal Component Analysis differentiated and classified the various species of selaginella.Nine species could be divided into 3 classes.The content of amentoflavone in Herba Selaginellae moellendorfii was 0.8~1.0 %.Conclusions The weighted similarity coefficient was calculated from 13 batches of Herba Selaginellae moellendorfii samples as high as more than 0.98.Among 9 species of Selaginella,5 species(S.biformis,S.involvens,S.doederileinii,S.trachyphylla,S.tamariscina)were sorted out by means of Principal Component Analysis in the same class with S.moellendorfii.Which indicated the possibility of bio-equivalence among the herbs of these six species,while S.picta,S.uncinata and S.delicatula are considered as adulterants of S.moellendorfii as their dissimilar fingerprints.
10.Antibody dependent cell-mediated cytotoxicity on hepatocytes during immune response in halothane induced hepatitis

Objective: To study antibody dependent cell-mediated cytotoxicity (ADCC) on hepatocytes during immune response in halothane hepatitis of guinea pigs. Methods: The model of halothane hepatitis of guinea pigs was used. Mixed lymphocyte hepatocytes were cultured and supernatants containing specific antigens and cytokines were added. The changes of hepatic function in immune response of halothane hepatitis were analyzed. Results: The cytotoxicity on hepatocytes induced by halothane 3 times included ADCC mediated by specific antigens. ADCC on hepatocytes induced by halothane once required the second induction. There was no cytotoxicity on hepatocytes of specific antigens and cytokines produced by proliferative lymphocytes induced by Kupffer cells. Conclusion: Humoral immunity is mainly ADCC. Specific antibodies and cytokines can not separately kill hepatocytes. The extent of immune response is related to induction by halothane.
