1.Investigation of the cognition and self management about osteoporosis and quality of life in elderly osteoporosis patients

Objective To investigate the cognition, self-management and quality of life of osteoporosis in elderly patients, and to provide the basic theory for health education in elderly patients with osteoporosis.Methods 565 elderly patients with osteoporosis in our hospital from May 2012 to Mar.2015 were enrolled.The cognition, health beliefs, self-efficacy and quality of life were investigated by questionnaires and analyzed by Microsoft Office Excel 2007.Results 565 osteoporosis patients, the symptom score was (15.6±2.0), and the score was lower in sporting knowledge (4.3 ± 1.8) than in calcium knowledge (5.7 ± 1.5) (t=2.784, P=0.011).Health beliefs score was (139.0±9.6), containing exercise benefits score of (21.5±3.1), exercise barriers score of (13.8±3.9), susceptible score of (19.4±4.2), serious score of (18.8±4.4), health motivation score of (25.0±3.0), calcium intake benefits score of (20.7±3.5) and calcium intake disorder score of (19.8 ± 4.0).Self efficacy score was (63.7 ± 19.4), containing exercise efficacy score of (58.6 ±18.7) and calcium efficacy score of (69.2± 18.5).Quality of life score included the general health (16.5± 2.9 score), social function (5.8 ± 1.3 score), vitality (14.8 ± 3.2 score), mental health (20.7± 4.2 score), physiological function (20.5 ± 4.1 score), physiological role limit (6.3 ± 1.6 score), emotional role limit (5.1 ± 1.3 score) and body pains (4.3 ± 1.8 score).Conclusions The score in cognitive function is low in elderly osteoporosis patients.It's useful to improve elderly osteoporosis awareness and quality of life by carrying out relative education.
2.Study of the Association of diabetic retinopathy with 2 candidate genes

Objective To study the relationship between diabetic retinopathy(DR) and Landsteiner-Wiener,Indian blood group genes.Methods Peripheral blood samples were collected from 50 DR patients,and 160 unrelated volunteer blood donors(the control).CDNA,including LW gene generated by reverse transcription polymerase chain reaction(RT-PCR),was subjected to sequence analysis,and the genomic DNAs,extracted from each sample,were sequenced directly for LW gene and IN gene.Woolf method was used for calculating relative risk(RR).Results In the 160 control samples,there was A at the nt380 locus in Exon1 of the LW gene,which were genotyped as LWa;and there was G at the nt252 locus in Exon2 of the IN gene,which were genotyped as INb.All the 53 samples from DR patients were LWa homozygous genotypes and all INb homozygous genotypes.Conclusion These two candidate genes are not associated with the incidence of DR.
3.Relationship between HLA class Ⅱ gene polymorphism and schizophrenia

Objective To investigate the genetic association of HLA class II DRB1,DQB1,DQA2 and DQB2 locus with schizophrenia in the Han people of Northern China.Methods The polymerase chain reaction(PCR)and restrictive fragment length polymorphism(RELP) were applied to detect the gene types in the 195 Han family trios including patients with schizophrenia and their healthy parents in the north of China.The chi-square(?2) test,transmission disequilibrium test(TDT) and the haplotype relative risk(HRR) analysis were used to process the genotyping data statistically.Results The genotypic frequency of four genes did not deviate from Hardy-Weinberg equilibrium in both case and control groups;The SNP rs2239800,a G to A base change,presented in the DQA2 locus.Its allelic frequencies showed significant difference between case and control(?2=5.021,P=0.025).And the AA genotype of rs2239800 showed associations with schizophrenia.Conclusion The polymorphism of rs2239800 in the DQA2 gene may be associated with schizophrenia.
4.Role of EGFR mutation status in patients with advanced lung adenocarcinoma treated with pemetrexed

Objective:To assess the role of EGFR mutations on pemetrexed response in patients with advanced lung adenocarcino-ma. Method: Forty pulmonary adenocarcinoma patients with evaluable lesions were retrospectively screened .They had been treated with pemetrexed-included chemotherapy and had EGFR gene test results. The evaluation endpoints were overall response rate,disease control rate and progression free survival. Result:No significant statistical difference was seen in overall response rate(ORR) (44.4%VS 31.8%, respectively) and disease control rate(DCR) (88.9%VS 81.8%, respectively ) between EGFR wild group and EGFR muta-tion group, but patients in EGFR wild group had longer progression free survival(PFS) ( 8.9 months VS 5.3 months;P=0.046). Conclu-sion:EGFR mutation status can influence the efficacy of pemetrexed.
5.Changes of serum cytokines and their clinical significance in infants with cytomegalovirus infection

Objective To investigate the changes of serum cytokines and explore their role in infants with cytomegalovirus (CMV) infection. Methods We recruited 41 positive CMV-IgM plus normal ALT infants (other disease group), 30 positive CMV-IgM plus abnormal ALT infants (hepatitis group) and 30 healthy infants (control group) in the study. The levels of tumor necrosis factor-?(TNF-?), interferon-?(IFN-?) and interleukin-4 (IL-4) in serum were measured with ELISA. The association between TNF-? and ALT was analyzed. Results The levels of TNF-?, IFN-?, IL-4 were higher, and IFN-?/IL-4 was significantly lower in the two CMV infection groups than the control group. Compared with other disease group, the changes of TNF-?, IL-4, and IFN-?/IL-4 showed significance in hepatitis group. 6.Optimization of compound lobelia gel in preparation process

Objective To study the factors which affect the preparation of compound lobelia gel and establish the optimal preparation process. Methods Based on single factor test, formability, spread performance, stability were used as comprehensive evaluation indicators to select the preparation process by orthogonal design, taking the dosage of carbopol-940, drug loading, 5% hydroxy ethyl benzene ethanol solution and triethanolamine as factors.The content of scutellarin in gel was determined by HPLC.Results The best prescription of compound lobelia gel was as follows: substrat of carbopol-940 was 0.25 g, 10% Azone for promoting the permeability was 2 g, pH regulator of triethanolamine was 1.0 g, moisturizer of glycerol was 0.4 g; preservatives of 5% ethylparaben ethanol solution was 0.3 g; the drug loading was 1 g processed herbs per 1 g gel. Conclusion The preparation process of compound lobelia gel is simple, the product is texture and delicate stable.
7.Resistance mechanism and molecular epidemiology of the New Delhi metallo-β-lactamase-1 possessing Klebsiella pneumoniae

Objective To investigate the molecule phenotype, epidemiology, and resistance genes of the New Delhi metallo- β-lactamase-1 ( NDM-1 ) producing Klebsiella pneumoniae ( K. pneumoniae ) . Methods Retrospective study was made on one hundred and ten non-repetitive carbepenem-resistant K. pneumoniae clinical isolated strains, which were collected from January 2011 to December 2012 in our hospital. The minimal inhibitory concentrations ( MICs ) of antibiotics were tested by the GN13 cards of BioMerieux Company. Modified Hodge test were used for the detection of carbapenemases. The blaNDM-1 encoding gene and linkage of ISAba125-NDM were detected by PCR method. The purified PCR products were cloned and sequenced. The homology of the K. pneumoniae were analyzed by the multilocus sequence typing ( MLST ) . Plasmid conjugation experiment and curing method were used to study the transfer of bacterial resistance. The Fisher′s exact probability test was used to compare the data. Results 13% NDM-1-producing K. pneumoniae were detected and confirmed as blaNDM-1 by sequencing (14/110). The resistance rates of the 14 NDM-1-producing K. pneumoniae strains to imipenem, meropenem, ertapenem, ciprofloxacin, levofloxacin, amikacin, and aztreonam were 14/14, 14/14, 13/14, 10/14, 9/14, 5/14, and 11/14. Meanwhile, the positive rate of ISAba125-NDM linkage of those 14 NDM-1-producing K. pneumoniae strains was 14/14. The E. coli J53 transconjugants, whose MICs of imipenem, meropenem, and ertapenem were increased by 4 to 64 times, were blaNDM-1 gene and ISAba125-NDM linkage positive. In addition, it was showed that the blaNDM-1 gene and ISAba125-NDM linkage were located on a plasmid with a size of approximately 65 000 bp. Conclusions The NDM-1 producing K. pneumoniae strains in this study were resistant to many commonly used antibiotics, however, the resistance rate to aminoglycoside and aztreonam were relatively low. The carbapenemase-resistant genotype spread by blaNDM-1 carried plasmid. Attention should be paid to its easily transmissible feature among the strains in clinic. The insertion sequence ISAba125 may be involved in the blaNDM-1 gene mediated carbapenemase-resistant genotype.

9.Cardiopulmonary Quantitatively Assessment for Spinal Cord Injury during Aerobic Exercise

Objective To quantitatively assess the cardiopulmonary exercise function of spinal cord injury (SCI) patients and observe the effect of aerobic exercise on cardiopulmonary function, motor function and activities of daily living. Methods From December, 2014 to June, 2016, 34 incomplete SCI patients (ASIA C and D) and 23 healthy controls received cardiopulmonary exercise test (CPET). SCI pa-tients were randomly divided into conventional rehabilitation group (n=17) and aerobic exercise group (n=17). The aerobic exercise group received aerobic exercise for four weeks. They were assessed with CPET, motor and sensory function, walking index for spinal cord injury II (WISCI II) and spinal cord independence measure (SCIM) before and four weeks after training. Results Oxygen uptake (VO2)peak, anaerobic threshold (AT), metabolic equivalent of energy (METpeak), VO2/heart rate (HR)peak, respiratory exchange rate (RER)peak, minute ventilation (VE)peak, work rate (WR)peak and systolic blood pressure (SBP)peak were lower in the patients than in the controls (t>2.714, P<0.05). VO2peak、AT、METpeak、VO2/HRpeak、WRpeak increased in the aerobic exercise group after training (t>2.431, P<0.05). METpeak and WRpeak improved in the conventional rehabilitation group after training (t>3.282, P<0.01). The scores of motor in ASIA and SCIM improved in both groups after training (t>2.985, P<0.05). Conclusion The cardiopulmonary function decreased in incomplete SCI patients, which could be improved by moderate intensity aerobic exercise.
10.MiR-503 sensitizes human hepatocellular carcinoma cells to cisplatin by targeting bcl-2

Objective:To investigate effects of miR-503 on cisplatin sensitivity in BEL-7402 cells by targeting of bcl-2.Methods:MiR-503 and bcl-2 mRNA expression levels in hepatocellular carcinoma cells were measured by real-time quantitative (qRT)-PCR;Bcl-protein level was detected by Western blot;miR-503 mimics were transiently transfected to the BEL-7402 cells by liposome transfection;potential target genes of miR-503 were predicted by Bioinformatics software;miR-503 potential targets were validated by dual luciferase activity;and the cell viability was measured by MTT assay.Results:MiR-503 level was down-regulated and Bcl-2 protein expression level was up-regulated in BEL-7402 cells compared with HL-7702 cells.MiR-503 could interact with bcl-2 and inhibit its expression.Cell vitality with miR-503 transfection was significantly reduced compared to that in the negative control.Conclusion:MiR-503 may enhance the sensitivity of BEL-7402 cells to cisplatin and inhibit the cell proliferation by targeting bcl-2.
