1.Analysis of diagnosis and prognosis of MRP8/14 on prostate cancer

Objective To compare the MRP8/14 level in Normal people,benign prostatic hyperplasia(BPH) patient and prostate cancer patient,and explore the relationgship between MRP8/14 and prostate cancer.Methods 150 cases of normal people,150 cases of BPH patients and 150 cases of prostate cancer patients were chose from December 2012 to December 2014 in our hospital.ELISA method was used to detect the MRP8/14 level in each group.ROC cure was used to analyse the prediction value of MRP8/14 for prostate cancer.According to the cut off value of MRP8/14,prostate cancer patients were divided into MRP8/14 low value group (MRP8/14 < cut off value) and MRP8/14 high value group (MRP8/14 ≥cut off value),and the difference of patient's clinical characteristics and survival function between high value group and low value group were explored.Results The MRP8/14 level of normal people was (966.7 ± 152.8) ng/ml,while the BPH patient was (1 207.0 ± 190.6) ng/ml,and the prostate cancer patient was (2 833.3 ± 1 101.5) ng/ml,the difference is statistically significant.ROC analysis result showed that the AUC for the prediction of prostate cancer was O.887 (95% CI 0.841-0.934),with a high statistical significance,indicating that MRP8/14 may possess high prediction value for prostate cancer.In addition,the cut off value was 2 845.682 ng/ml,with the specifity of 76.4% and sensitivity of 85.1%.According to the cut off value of MRP8/14,prostate cancer patients were divided into the low MRP8/14 group (< 2 845.682 ng/ml) and the high MRP8/14 group (≥2 845.682 ng/ml).Among the 150 prostate cancer patients,88 cases were in the low MRP8/14 group and 62 cases were in the high MRP8/14 group.Comparations of the baseline characteristics of the two groups showed that amount of patients belong to ECOG PS =1,Gleason score =8-10,organ involvement > 2 and tumour stage > Ⅲ were much more in MRP8/14 high value group.PSA level,LDH level,AKP level,CRP level and Alkaline Phosphatase level were also significantly increased in MRP8/14 high value group.Besides,prostate cancer patient with an average follow-up of 2 years,a total of 32 cases of patients died,12 cases in the MRP8/14 low value group with mortality of 13.6%,20 cases in MRP8/14 high value group with mortality of 32.3%.Kaplan Meier survival function curve shows 2 years survival rate of patients in high value group was significantly reduced.Cox regression analysis showed that MRP8/14 was possible risk factors associated with mortality,and the independent predictors for all-cause mortality during follow-up.Conclusion MRP8/14 was significantly increased in prostate cancer patients,and it was an independent predictor of 2-year mortality in prostate cancer patients.
2.Alteration of interleukin-17/interferon-γ double positive cells in mice with Coxsackie virus induced myocarditis

Objective To observe the alteration of interleukin-17 and interferon-γ double positive cells(IL-17 +/IFN-γ cells)in mice with coxsackie virus B3(CVB3)induced acute viral myocarditis(VMC).Methods VMC was induced in male Balb/c mice by peritoneal injection of CVB3.Control mice received PBS injection.At 0,1,2,3,4 and 6 weeks after injection,pathological scores were determined on hematoxylin-eosin stained heart sections and flow cytometric analysis was performed to evaluate the percent of IL-17+/IFN-γ+ cells among CD4+ T cells.Results Compared to control mice,the pathological scores of VMC mice were higher on CVB3 infection week 1(1.8 ± 0.5),peaked on week 2(2.8 ± 0.5)and declined thereafter.However,the proportion of IL-17 +/IFN-γ+ cells remained steadily at a low level throughout the observation period and was similar between VMC and control mice.Conclusions Our data shows that IL-17 +/IFN-γ+ cells might not be involved in the inflammation process of CVB3 induced VMC mice model.
3.Study on relativity of hypoxic pulmonary hypertension with levels of endothelin-1 and vascular endothelial growth factor in plasma of pulmonary artery and carotid artery.

AIMTo study the effects of endothelin-1 (ET-1) and vascular endothelial growth factor (VEGF) on the mechanism of hypoxic pulmonary hypertension (HPH).
METHODSWe studied 4 groups of age-controlled male rats, i.e., normal control for 2 weeks group (N2), normal control for 3 weeks group (N3), exposed to hypoxia for 2 weeks group (H2) and for 3 weeks group (H3). Chronic HPH rat models were established by chronic hypobaric hypoxia [(10.0% +/- 0.5% O2] for 2 and 3 weeks, respectively. The rats were anesthetized and fixed, and the levels of mean pulmonary artery pressure (mPAP) and carotid arterial pressure (CAP) were measured using catheters by a microcomputer via transducers. The weight ratio of right ventricle (RV) and left ventricle and septum (LV + S) [RV/ (LV+S)] were determined. The contents of ET-1 in plasma of pulmonary artery and carotid artery and in homogenates of lung and systemic arteries were determined by radioimmunoassay, and the contents of VEGF in serum of pulmonary artery and carotid artery were determined by ABC-ELISA.
RESULTSHPH rat models were established successfully. Compared with control groups, the values of ET-1 were both enhanced in carotid artery and pulmonary artery plasma in model groups (P < 0.01). In the HPH groups, the level of pulmonary artery plasma ET-1 was significantly lower than that of carotid artery plasma, but just the reverse was ET-1 in control rats. The levels of ET-1 in homogenates of lungs from HPH models were significantly higher than those in homogenates of lungs from control groups (P < 0.01), and markedly higher than those in homogenates of systemic arteries from HPH rats (P < 0.01) SThe values of VEGF in serum of pulmonary artery from H3 group were significantly higher than those from control groups and H2 group (P < 0.01). In serum of carotid artery, the values of VEGF from the HPH models were higher than those from the control groups (P < 0.01).
CONCLUSIONET-1 and VEGF play important roles in the pathogenesis of HPH. The result that ET-1 concentration around pulmonary arteries was significantly higher than that around systemic arteries may be one of the mechanisms accounting for the different reaction of them to hypoxia.

4.Study on quality control of effective fraction in qixue bingzhi decoction.

OBJECTIVETo develop a method for quality control of effective fraction in Qi-Xue-Bing-Zhi decoction, a traditional Chinese medicine (TCM).
METHODPF samples, effective fraction from Qi-Xue-Bing-Zhi decoction, were used as example, and a HPLC assay for chemical fingerprint and quantitative analysis was established.
RESULTThe contents range of Paeoniflorin (PE), Naringin (NG) and Neohesperidin (NH) in effective fractions were changed from 12.5%-16.0%, 8.4%-12.4%, 12.8%-15.3%, and their average contents were (14.7 +/- 1.1)%, (10.6 +/- 1.2)%, (14.2 +/- 0.8)% (n = 10), respectively. The fingerprints of PF samples showed 25 common peaks, and the fingerprint similarity for PF samples were all above 99.00% by comparing with the standard chromatogram.
CONCLUSIONThe method reported could be used effectively for the quality control of effective fraction from TCM.

5.Effect of Huanglian Jiedu Decoction on Monocyte Development in apoE Gene Knockout Mice.

OBJECTIVETo observe monocyte (Mo) development in wild type C57BL/6 mice and apoE gene knockout (apoE(-/-)) mice, and to evaluate the immuno-regulatory effect of Huanglian Jiedu Decoction (HJD) on peripheral Mo development in apoE(-/-) mice.
METHODSFour, 8, 12, and 16 weeks old female C57BL/6 mice were set up as control groups of different ages, while 4, 8, 12, and 16 weeks old female apoE(-/-) mice were set up as hyperlipidemia groups of different ages. Four-week old female C57BL/6 mice were recruited as a blank group. Four-week old female apoE(-/-) mice were randomly divided into the control group, the Western medicine group, and the Chinese medicine group by paired comparison, 5 in each group. Equivalent clinical dose was administered to mice according to body weight. Mice in the Western medicine group were administered with Atrovastatin at the daily dose of 10 mg/kg by gastrogavage, while those in the Chinese medicine group were administered with HJD at the daily dose of 5 g/kg by gastrogavage. Body weight was detected each week. After 4 weeks blood lipids levels (such as TG, TC, LDL-C, and HDL-C), and the proportions of Mo and Ly6c(hi) were detected.
RESULTSCompared with 4-week-old homogenic mice, the proportion of Mo decreased in 16-week-old C57BL/6 mice (P < 0.05). Levels of TC and TG, and the proportion of Ly6c(hi) subtype increased, but the proportion of Mo de- creased in 8-week-old apoE(-/-) mice (P <0. 05). Levels of TC, TG, and LDL-C increased in 12-week-old apoE(-/-) mice (P < 0.05). Levels of TC, TG, LDL-C, and HDL-C increased in 16-week-old apoE(-/-) mice (P < 0.05, P < 0.01). Compared with 8-week-old homogenic mice, the proportion of Mo decreased in 16-week-old C57BL/6 mice (P < 0.05); levels of TC and LDL-C increased in 12-week-old apoE(-/-) mice (P < 0.05); levels of TC and HDL-C increased in 16-week-old apoE(-/-) mice (P < 0.05, P < 0.01). Compared with C57BL/6 mice of the same age, TC and TG increased, HDL-C decreased (P < 0.01) in 4-and 8-week-old apoE(-/-) mice (P < 0.01); levels of TC, TG, LDL-C increased, and HDL-C level decreased in 12- and 16-week-old apoE(-/-) mice (P < 0.05, P < 0.01); the proportion of Mo increased in 4-week-old apoE(-/-) mice (P < 0.05); proportions of Mo and Ly6c(hi) increased in 8-week-old apoE(-/-) mice (P < 0.05). Compared with the blank control group, levels of TC, TG, and LDL-C, proportions of Mo and Ly6c(hi) increased (P < 0.01, P < 0.05), but HDL-C level decreased (P <0. 01) in the control group after intervention. Compared with the control group, body weight gained less in the Western medicine group and the Chinese medicine group (P < 0.05); the proportion of Ly6c(hi) subtype decreased in the Chinese medicine group (P < 0.05).
CONCLUSIONSIn development process blood lipids levels in apoE(-/-) mice are not only associated with age. Blood lipids levels induced growth changes in natural immune system are also correlated with age. In early stage of lipids development HJD intervention could correct this special immune disorder in apoE(-/-) mice.

6.Clinical analysis of 10 patients with de novo CD5 positive diffuse large B cell lymphoma.

To explore the clinical characteristics, diagnosis, treatment outcome and prognosis of de novo CD5 positive diffuse large B cell lymphoma (CD5(+)DLBCL), clinical data of 10 patients with pathologically confirmed CD5(+)DLBCL were retrospectively analyzed. The results indicated that 9 out of 10 patients were older than 60 years. All cases were in III/IV stages according to Ann-Arbor Staging System. Bone marrow biopsy with immunohistochemistry showed lymphoma involvement in 5 cases. Nine patients received chemotherapy with anti-CD20 monoclonal antibody (Rituximab) except one. Five cases achieved CR, two cases achieved PR, two cases achieved SD, one case achieved PD. Eight cases died within 2 years because of relapse or disease progression, in which 3 cases developed central nervous system lymphoma. The median survival time was 16 (1-23) months, 2-year survival rate was 20.40%. It is concluded that de novo CD5(+) DLBCL is rare in clinic, but it is a kind of highly aggressive lymphoma with poor prognosis. So, new treatment strategy should be explored.

7.Observation of combination anesthesia with low-dose ketamine and mid-dose pentobarbital for open heart surgeryin juvenile pigs

Objective To investigate combination anesthesia with low-dose ketamine and mid-dose pentobarbital for open heart surgery in ju-venile pigs.Methods Thirty experimental juvenile pigs received ket-amine(3 mg·kg~(-1))and pentobarbital(20 mg·kg~(-1))intramuscularly for induction and intubation,then were given ketamine(5 mg·kg~(-1)·h~(-1)),pentobarbital(6-8 mg·kg~(-1)·h~(-1)),midazolam(0.1-0.2mg kg~(-1)·h~(-1))and pipecuronium(0.1 mg·kg~(-1)·h~(-1))intravenously for maintenance.The infusion of pentobarbital was withdrawn after car-diopulmonary bypass started.During the experimental treatment,vital signs were monitored;artery gas analysis and hemodynamie parameters were recorded.Results Twenty-eight juvenile pigs got stable hemody-namic parameters in the perioperative period.The time of anesthetic in-duction and maintenance is(9±2)and(179±15)min,respectively.The respiratory and cardiac arrest rates were 6.7% in induction and 3.3% in maintenance.respectively.Two cases got respiratory and cardi-ac arrest.on in induction and the OtIler after extubation.In addition,class I anesthetic effectiveness was achieved 73.3% in induction and 80.0% in maintenance,respectively.Conclusion This study demon-strated that lOW-dose ketamine combined with mid-dose pentobarbihal has little inhibition on respiration and circulation.The combination can achieve both hypnosis and analgesia effects with good surgical anesthetic effectiveness in juvenile pigs with open heart surgery.
8.A study on engineering protection measures and risk assessment of an oil terminal project

Objective To analyze and evaluate the occupational hazards,engineering protection measures and risk levels in an oil terminal project.Methods Engineering analysis,occupational health survey,inspecting -testing and semi -qualitative semi-quantitative evaluation method (MES method)were used to analyze the production process,the type of occupational hazards,characteristics and concentrated/strong degrees and engineering protection measures of the project. Also,the occupational health risk and the effect of engineering protection were assessed.Results The main occupational hazards of this project were dust,solvent naphtha,benzene,toluene,xylene,hexane,non -methane hydrocarbons, carbon monoxide,carbon dioxide,manganese and its inorganic compounds,hydrochloric acid,hydrogen sulfide,noise, high temperature,power frequency electric field,welding electric arc and so on.Approximately,16.67%of noise samples and 66.67% of high temperature samples exceeded the Chinese National Occupational Health Standards.Except for noise of the fuel pump shed and high temperature of 100000 tons pier and fuel tank,the occupational hazards of other jobs were in line with the national occupational health standards.In normal conditions,the occupational risk levels of all positions were 5 (slightly hazardous),while in special conditions,the occupational risk level of repair welders was 5 (slightly hazardous),of pier operators and tank farm operators were 1 (extremely dangerous),of sewage treatment workers,boiler room operators and electricians were 3 (significant risk ).Conclusion This project can take effective engineering protective measures to control occupational hazards and risk management of special conditions need to be strengthened.
9.A study on application of semi-quantitative risk assessment models in furniture industry

Objective To explore the suitable risk assessment model for printing industry, we evaluated the risk of furniture industry using models provided by Singapore Ministry of Manpower (MOM) . Methods According to the occupational-disease-prevention laws and regulations, we improved the MOM and increased the risk assessment of physical agents. Then, we evaluate the risk of furniture industry using the improved MOM. Results The range of wood dust was 1.9-8.9mg/m3, and the percent of pass was 50%. The range of formaldehyde, benzene, methylbenzene and dimethylbenzene was 0.24-0.36 mg/m3, <0.6mg/m3, 3.8-6.7mg/m3 and 6.1-16.1mg/m3 respectively, and their percent of pass was 100%. Furthermore, the noise intensity was 80.8 dB (A), which was under the occupational exposure limit. The risk level of wood dust, formaldehyde, benzene and noise was high, while the risk level of methylbenzene and dimethylbenzene was moderate. Conclusion The improved semi-quantitative risk assessment was appropriate for furniture industry, but its applicability in industrial enterprise needs to be studied further.
10.Treatment of an infant with severe neonatal hypoxic-ischemic encephalopathy sequelae with transplantation of human neural stem cells into cerebral ventricle.

OBJECTIVESevere newborn hypoxic-ischemic encephalopathy (HIE) has a very high rate of disability and no effective treatment is available. The present study aimed to preliminarily evaluate the effects of human neural stem cell transplantation in treatment of severe neonatal HIE.
METHODSThe patient was a 75-day old male infant with sequelae of severe HIE who had highly delayed development of intelligence and movement and myotonia. MRI showed multiple cerebromalacia and encephalatrophy. Cells obtained from the forebrain of an 11-week old fetus were cultured and amplified for 15 days. And then the human fetal neural stem cells were injected into cerebral ventricle of this infant.
RESULTSTwenty eight days after transplantation, remarkable improvement occurred not only in his myotonia but also in his intelligence and movement, which became similar to those of the normal infants of the same age. Positron emission tomography (PET) showed significantly increased radioactivity at temporal and occipital lobes which suggested that the cellular metabolism had increased greatly.
CONCLUSIONThe short-term effect of NSCs transplantation on the infant with severe HIE sequelae was significant. PET suggested that the implanted NSCs survived. Many more studies are needed to evaluate long-term effects of NSC transplantation in treatment of HIE.
