1.Effects of HSP70 on hepatic ischemia-reperfusion injury after heat preconditioning in rats

Aim To study the effects of HSP70 on hepatic ischemia-reperfusion injury after heat preconditioning in rats.Method To establish the models of the hepatic ischemia-reperfusion injury. 42 Wistar rats were randomly divided into 6 groups, (each group had 7 rats): normal group(N); quercetin injection group(Q); ischemia-reperfusion group(I); heat preconditioning 16 hours before ischemia-reperfusion group(H+I); quercetin injection before heat preconditioning group(Q+H+I); quercetin injection before ischemia-reperfusion group(Q+I).We detected the activity of serum enzyme of ALT,AST and the pathological changes of the liver;The expressions of HSP70 of the rats were observed by Western blotting. Results The expressions of HSP70 from high to low were:group H+I,group I,group Q+H+I,group Q+I,group Q,group N; The serum levels of ALT and AST from high to low were: group Q+I,group I,group Q+H+I,group H+I,group Q,group N;All groups had visibly hepatic histological changes respectively.Conclusion The protection of heat stress pretreatment from ischemia reperfusion injury was possibly performed by inducing the expression of HSP70.
4.Multislice CT and Clinical Manifestations of Diaphragmatic Hiatus Hernia with Hernial Sac Effusion

PurposeTo explore the multislice CT (MSCT) and clinical manifestations of diaphragmatic hiatus hernia with hernial sac effusion, in order to improve its diagnostic rate.Materials and MethodsMSCT data were retrospectively analyzed for morphologies and clinical manifestations in 32 patients with diaphragmatic hiatus hernia and hernial sac effusion.Results32 patients included 21 males and 11 females (χ2=0.64,P>0.05). 25 patients were older than 60 years and 7 cases younger than 60 years (χ2=13.58,P<0.01). There were 27 cases with non-viscera hiatus hernia including 23 cases of combined fat-water hernia and 4 cases of simple effusion. 5 cases were of viscera type hiatus hernia (χ2=5.47,P<0.05). 28 cases showed ascites including 23 cases with non-viscera hiatus hernia and 1 case with viscera type hiatus hernia (χ2=9.56,P<0.01). The axial images from different levels of non-viscera hiatus hernia with liquid hernial sac demonstrated quasi-circular, meniscus, ringlike and teardrop shapes.Viscera type hiatus hernia and liquid hernial sac were found to be irregular shape . All patient sufferered from dysphagia, chest distress and epigastric discomfort.ConclusionThe increased pressure gradient between thorax and abdomen driving ascites into supradiaphragmatic hernial sac and clamping by hiatus may be the main mechanism. Quasi-circular, meniscus, ringlike andteardrop were the characteristic signs of diaphragmatic hiatus hernia with hernial sac effusion.
5.Preliminary analysis of relative clinical and MSCT measurement of flat inferior vena cava

Objective To investigate the correlation between flat IVC (fIVC)and relative clinical,index of prolapse of kidney(K) and lordosis of abdominal aorta (A)with respect to the lumbar on supine.Methods Abdominal CT image data of 100 cases without sign of low blood volume were selected randomly.The K and A were calculated in the plane of the entrance of the renal vein and fIVC was defined according to the ratio of length to short diameter of IVC (R)up and down about 1.5 cm plane,which large to 3 ︰ 1.The index size between different groups tested by t,the association of fIVC with age and gender tested byχ2 .Results Among 100 cases, male 57 cases,female 43 cases,fIVC was 14 and 20 cases respectively (χ2 =5.26,P <0.05);The old,middle-aged and young groups were 56,38 and 6 cases respectively,the constituent ratio of fIVC was 44.64%(25/56),23.68%(9/38)and 0(0/6)respectively;The mean of K plus A was 2.57±0.25,2.1 5 ±0.14 and 2.05 ±0.1 1 respectively.The two former of old group was all significantly more than that of green group (χ2 =6.40,t=5.06,P <0.05).The mean of K plus A of fIVC and non-fIVC set was 2.67±0.25 and 2.03±0.18 (t=3.89,P <0.05)respectively.Conclusion The increased K and A on supine perhaps as causes for the formation of fIVC and its more located nearby the entrance of the renal vein,and is associated with age and gender.

7.Clinical Significance of Serum Cardiac Troponin in Patients with Dilated Cardiomyopathy

Objective To observe the serum cardiac troponin I (cTnI) level and to analysis the relations between the serum cTnI level and prognosis in patients with dilated cardiomyopathy (DCM) and Cardiac Dysfunction.Methods Serum cardiac cTnI level was measured by enzyme-linked immunosorbant assay. Results The serum cTnI level in DCM patients with class IV cardiac function (0.53 ?0.31) ?g/L was significantly higher than in DCM patients with class Ⅲ cardiac function (0.45?0.27) ?g/L.There was significantly difference in serum cTnI levels between DCM patients with class Ⅲ cardiac function and DCM patients with class Ⅱ cardiac function(0.29?0.27) ?g/L.Conclusion The higher serum cTnI level is correlated with the severity of cardiac function and may be useful for evaluating prognosis in patients with DCM.

9.Protective effects of local heat preconditioning on hepatic ischemia/reperfusion injury.

AIMTo study the protective mechanism of HSP70 induced by the heat stress pretreatment on the hepatic ischemia/ reperfusion (I/R) injury.
METHODSTo establish the models of the hepatic ischemia/ reperfusion injury using pringle's maneuver with or without heat stress pretreatment. The rats were randomly divided into pretreatment group (HP + I/R) and non-pretreatment group (I/R), in which the expression of HSP70, the MDA contents and SOD activity in liver, the activities of serum AST and ALT, and the pathological changes in liver were detected at 0, 4, 8, 12 and 24 h after I/R.
RESULTSAt any time point set after I/R, the SOD activity in the liver in HP + I/R group were higher than those in IR group, HSP70 expression maximum was attained at 12 h after heat pre-treatment. While in HP+ I/R group the levels of liver enzymes and the production of MDA significantly reduced and the pathological changes improved compared with those in I/R group.
CONCLUSIONHSP70 induced by heat pretreated protecting liver against I/R injury may be through increasing the SOD content and then reducing the insults of oxygen free radicals in the liver.

10.Capsazepine in rats′lateral ventricles:Effect of different doses on LPS-induced fever

Aim To observe the effects of the transient receptor potential vanilloid receptor 1 ( TRPV1 ) antago-nist Capsazepine on the LPS-induced fever process. Methods Three doses of Capsazepine were respec-tively injected into rats’ lateral ventricles before the in-traperitoneal injection of LPS caused fever. An emitter was implanted inside the rat’ s abdominal cavity to mo-nitor the body temperature changes during the fever process. Then the changes of calcium ion in the hypo-thalamus were detected by fluorescence and the expres-sions of TRPV1 protein were detected by Western blot. Results The higher dosage of Capsazepine was injec-ted, the higher the level of LPS-induced fever was, and the lower the concentration of calcium ion in the hypothalamus as well as the TRPV1 expressions were. Conclusion Capsazepine could increase the level of fever induced by LPS via acting on the rats’ hypothala-mus. This effect might be related to the functions of TRPV1 .
