Journal of Huazhong University of Science and Technology (Medical Sciences) 2009;29(1):131-3
To establish a murine carotid artery transplantation model for the study of the chronic rejection, 80 rats were divided into two groups, an allotransplant (ACI-Lewis) group and an isotransplant (Lewis-Lewis) group (control group). The donor carotid artery and the recipient carotid artery were anastomosed by using a polyethylene cuff (internal diameter: 0.7 mm, length: 3 mm).The pathological changes of carotid artery transplant were observed 14, 28 and 56 days after the transplantation. The results showed that the model was successfully established in 95% of the animals. The chronic rejection-associated arteriosclerosis was induced 28 days after the transplantation. The new chronic rejection model of carotid artery by using cuff technique caused fewer traumas and was easy to make. The pathological changes of the transplant mimicked the chronic rejection-associated arteriosclerosis found in human transplant.

Objective To investigate the impact of the modified cast impression technique on distal-extension alve-olar ridge morphology of partially edentulous casts and the restoration outcome of removable partial dentures (PRDs). Meth-ods Fifty-four patients with mandibular free-end dentition defect were selected. Impressions were taken from each patient using both modified cast technique and conventional technique. Coronary cross-sections of the casts were scanned from dis-tal to mesial extension;the images were compared to reveal morphological differences. And the relationship between these differences and location of the cross-sections were analyzed. After the patients were evenly divided into two groups per paired design, the impressions were taken using the respective modified cast and conventional impression techniques to pre-pare RPDs. The subsequent comparisons between the two groups included frequency of adjustments during the adaptive phase, masticatory efficiency of the denture at ending of the adaptive phase and visible movement of the extension base dur-ing mastication. SPSS 16.0 software was used for statistical analysis. Results The number of pixels in the same cross-sec-tion was less in the impression taken with modified cast technique than that with conventional technique. And their differ-ence is of statistical significance (P < 0.01). The difference in the number of pixels (D) was negatively correlated with the number of cutting and grinding, and N (location of the cross-sections). In addition, there were statistically significant differ-ences in frequency of adjustments during the adaptive phase (times:1.12 ± 0.77 vs 3.41 ± 0.82), masticatory efficiency of the denture (0 vs 18.5%) and visible movement of the extension base during mastication (96.3%vs 74.1%) between the modified cast group and conventional group. Conclusion Morphology of distal-extension alveolar ridge of the casts differs signifi-cantly between the impressions obtained with modified cast technique and the impressions obtained with the conventional technique;the difference increases with greater distance to the distal surface of the terminal abutments. The restoration out-come of RPDs is significantly better with the modified cast impression technique than with the conventional impression tech-nique.

Objective To investigate the effect of globular adiponectin on the high expression of monocyte chemotactic protein-1 (MCP-1) induced by high glucose in rat renal tubular epithelial cells (NRK52E),and its relationship with adiponectin receptors and p38MAPK.Methods NRK52E cells were cultured in vitro and divided into six groups:normal glucose group (NG,5.6 mmol/L glucose),high glucose group(HG,25 mmol/L glucose),gAd groupl (HG+gAd 2 mg/L),gAd group2 (HG+gAd 5 mg/L),gAd group3 (HG+gAd 10 mg/L),p38MAPK antagonist group:(SB,HG+SB203580 10 μmol/L).The protein expression of phosphorylated p38MAPK (p-p38MAPK),total p38MAPK (t-p38MAPK),MCP-1 and AdipoR1/AdipoR2 were examined by western blotting. The mRNA expression of MCP-1 and AdipoR1/AdipoR2 were detected by RT-PCR and real-time PCR respectively. Results Compared with NG group, the mRNA and protein expression of MCP-1 increased significantly in HG group (all P< 0.05).The phosphorylation of p38MAPK increased (P< 0.05) with no change in t-p38MAPK protein. The addition of gAd or SB203580 inhibited the unregulation of MCP-1 and p-p38MAPK induced by HG.Two kinds of adipoR,adipoR1 and adipoR2,were all detectable in NG group, and mRNA and protein expression of adipoR1 was higher than that of adipoR2 (P< 0.01).Compared with NG group, the expression of adipoR decreased in HG group, but the difference had no statistical significance(P > 0.05).Compared to HG group, the mRNA and protein expression of adipoR1 increased in gAd groups (all P < 0.01).Conclusion The gAd can dose-dependently attenuate the overexpression of MCP-1 induced by high glucose, and this protective effect may be mediated by adipoR1 and p38MAPK.
