Objective To study the changes of serum liver functions in recurrent hepatitis C ~after liver transplantation. Methods The clinical data of 50 cases of recurrent hepatitis C (~simultaneously negative for serum HBV DNA) after liver transplantation were collected. The relationship between liver function parameters with the HCV RNA genotype, inflammation histological activity, fibrosis stage, steatosis grade and rejection was evaluated. Results ALT, AST and ?-GT were increased in recurrent hepatitis C. Among them, ?-GT was obviously elevated along with the ~aggravation of histological activity (P

Objective To investigate the changes in cyclophilin A (CyPA) signal pathway in rat myocardial tissue after cardiopulmonary resuscitation (CPR).Methods Sixty-eight healthy male Sprague-Dawley (SD) rats were randomly divided into sham operation group (n =8),instantancous CPR after cardiac arrest (CA),immediate CPR after CA (CRP),and 15,30,60 and 120 minutes after CPR groups, respectively, with 10 rats in each group.Asphyxia was simulated by occlusion of the tracheal tube at the end of exhalation.Mechanical ventilation, compression and epinephrine injection were given for restoration of spontaneous circulation (ROSC) in order to reproduce cardiopulmonary resuscitation after cardiac arrest (CA-CPR) models in rats.Hemodynamic changes were recorded at different time points. The blood was collected from abdominal aorta, and myocardial tissue was also harvested. The serum CyPA and CD147 levels were determined by enzyme-linked immunosorbent assay (ELISA).The protein positive cells and mRNA expression of CyPA and CD147 in myocardial tissue of the rats were determined by immunohistochemical and real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR).The protein expressions of matrix metalloproteinases (MMP-2,MMP-9) and myeloperoxidase (MPO) in rat myocardial tissue were determined by Western Blot. The neutrophil infiltration in rat myocardial tissue was assessed by hematoxylin and eosin (HE) staining. Results The heart rate of rat was lowered to 0 with arterial pressure lowered immediately after CA.Arterial pressure was elevated to normal range immediately after CPR.Heart rate was restored at about 30 minutes later, and the dose of epinephrine was 50-60 μg,and ROSC time was 1-4 minutes.Compared with those of the sham group, serum CyPA and CD147 levels were gradually increased along with elongation of ROCS time within 120 minutes, and peaked at 120 minutes,CyPA was increased from (786.11 ± 3.93) μg/L to (2 001.80 ± 10.61) μg/L,and CD147 was increased from (2.94±0.02) μg/L to (5.99±0.023) μg/L (P < 0.05 or P < 0.01).CyPA and CD147 mRNA expressions (A value) in rat myocardial tissue were gradually increased, and peaked at 120 minutes.Relative expression of CyPA at 120 minutes was 2.42 ± 0.05 when it was 1 in the control, and relative expression of CD147 at 120 minutes was 1.88 ±0.10 (both P < 0.01).The immunohistochemical results under light microscope showed that the brown positive cells were gradually increased, which indicated that the expressions of CyPA and CD147 were increased.Expressions of MMP-2,MMP-9 and MPO (gray value) in myocardial tissue were also gradually increased, peaking at 120 minutes, and MMP-2 was increased from 0.396 ± 0.021 to 0.879 ± 0.020,MMP-9 was increased from 0.372 ± 0.009 to 0.819±0.012,and MPO was increased from 0.176±0.005 to 0.829±0.018 (P < 0.05 or P < 0.01).No obvious neutrophil infiltration in myocardial tissue was found with HE staining. Conclusion Expressions of CyPA and CD 147 were up-regulated in serum and myocardial tissue after CPR in rats, which may be the markers of inflammatory reaction.

Objective: To study the relationship between the microvascular density, cell nuclear DNA content and the prognosis in larynge al cancer patient. Methods: In 31 cases, microvascular density was obs erved with anti-factor Ⅷ antibody and SABC method, and the proliferating nuclea r DNA content was examined with image cytometry. Results: The microva scular dencity and DNA content were not related to histologic grade and type of laryngeal cancer, but the differences of microvascular density were significant ( P<0.05) between the patients with metastasis (10.23±0.78) and these withou t metastasis (6.38±0.33) after two years. The measurement by the image cytometry showed that the metastasis groups had a mean nuclear peak area value of (61.03 ±0.28)μm2 and the mean stage cell value of (38.11±0.37)%,and those of the g roups without metastasis were (40.97±0.41)μm2 and (22.97±0.29)%,respectivel y.There were significant differences (P<0.05) between them. Conclusion: The angiogenesis and proliferating nuclear DNA content o f laryngeal cancer are correlated with the lymph node metastasis and prognosis.

Objective To investigate the effects of electroacupuncture (EA) on the implementation of blind nasojejunal (NJ) tube placement and enteral nutrition (EN) in neurosurgical severe coma patients in intensive care unit (ICU).Methods Seventy-nine neurosurgical severe coma patients admitted to ICU were randomly divided into conventional group (blind NJ tube placement,n =40) and EA group (NJ placement and EA,n =39).EA was performed after NJ tube placement at bilateral acupoints Zusanli (ST36) and Hegu (L14) points using EA treatment instrument. The impelling distance of NJ tube were measured and the success rate of NJ tube placement were calculated. The postoperative complications were observed. Results The difference of NJ tube impelling distances at the 24th,48th,and 72th hours after surgery in EA group were significant longer than that in conventional group (P < 0.05).The success rates of NJ tube placement at the 24th and 72nd hours after surgery in EA group were significantly better than that in conventional group (P < 0.05).Their EN calories qualifiedness rate in 72 hours also increased significantly compared with conventional group and the proportion of patients assisted with parenteral nutrition decreased (P <0.05).The postoperative complications including alimentary tract hemorrhage, vomiting, and abdominal distension decreased remarkably in EA group compared with conventional group (P < 0.05).Conclusions EA stimulation at acupoints could promote the gastrointestinal peristalsis of neurosurgical severe coma patients and elevate the success rate of blind NJ tube placement,so it is beneficial for the implementation of early enteral nutrition (EEN).

Objective·To prepare chitosan-gelatin porous microspheres by high voltage electrostatic method combined with freeze-drying and ionic cross-linking method and investigate the factors that influence the formation of porous medium. Methods·Porous chitosan microspheres and chitosangelatin porous microspheres were prepared using high voltage electrostatic method combined with freeze-drying and ionic cross-linking method, with sodium tripolyphosphate (STPP) as crosslinking agent.Factors that affect the porous structure and pore size of porous microspheres were compared, such as different chitosan-gelatin ratio, freezing temperature, curing time with saturated STPP in 85% ethanol solution. The morphology, surface and internal structure, particle size of the porous chitosan microspheres and chitosan-gelatin porous microspheres were observe by using light microscope, scanning electron microscope and hematoxylin-eosin staining. Results·Microspheres prepared by freeze-drying with an electrostatic and ionic cross-linking method have open, interconnected and highly macroporous, with good spherical surface.Saturated STPP ethanol solution (85% ethanol) was chosen as the crosslinking agent to prevent destruction of the porous structure. The order of freeze and crosslinking,cross-linking time and the second freezing temperature, can influence the pore size of porous microspheres.Gelatin and chitosan can form polyelectrolyte complexes, and can also be used as porogen in porous structure. Conclusion·The preparation of porous chitosan-gelatin microspheres via this method has a large pore size (diameter 100-200 μm),suitable for cell growth and the migration.
