1.Study on the decreasing pattern of activities of daily living in patients with dementia

Objective To investigate the decreasing pattern of activities of daily living (ADL) in patients with dementia. Methods A total of 204 patients with dementia were collected,among which 106 cases were with Alzheimer's diseases(AD),54 cases with vascular dementia(VD),44 cases with other types of dementia.According to the scores of mini-mental state examination(MMSE),mild,moderate and severe dementia were 42 cases,37 cases and 125 cases,respectively.All of them were assessed by Barthel index(BI) and compared among degrees of dementia,between AD and VD,and levels of education. Results The most reserved items were bowel movement (48.5 %),urination (44.1 %) and grooming (40.2%),while the most vulnerable items were climbing stairs (25.5%) and bathing(18.1%).With increasing impairment of cognition,the scores of BI were getting decreasing. There were significant differences among severe [(23.20 ± 29.27)scores],moderate[(65.54 ± 26.35) scores] and mild [(87.38 ± 22.69) scores] groups (x2 =99.686,P =0.000). Basic ADL were more reserved in AD (16.98%-53.77%) compared with VD (1.85%-33.33%) patients.No significant differences were found(x2 =3.653,P=0.161)among patients with education of primary school and below [(40.1 ± 39.0) scores],middle school[(45.3 ± 39.6 ) scores],and high school and above[(53.1 ± 37.7) scores]. Conclusions Among patients with dementia,bowel control,bladder control and grooming are well reserved,climbing stairs and bathing are vulnerable functions.Basic activities of daily living may be affected by cognitive status and the types of dementia but not levels of culture.
2.Status of iodine content of common foods in different geographical areas in Fujian Province

Objective To determine the iodine content of common foods in different geographical areas (coastal city,coastal rural area,mountainous city and rural area) in Fujian Province and provide basic data for evaluation of dietary iodine intake.Methods In 2010,based on the types of food of the total diet study,one food sample(consumption rate is greater than 1%) was collected in coastal city(Taijiang),coastal rural area(Xiang'an),mountainous city (Sanyuan) and rural area(Mingxi).These foods including cereal,beans,potato,meat,eggs,milk,aquatic products,vegetables,fruits,sugar,beverages,liquor and seasoning,and so on 184 kinds of common foods.The iodine content of these food was tested by As (Ⅲ)-Ce4+ catalytic spectrophotometry.Results Among the 184 kinds of food tested,164 were not indicated food iodine content in the Chinese Food Composition Tables (2004).The iodine content of common food in Fujian Province in descending order were salt (30 000 μg/kg),aquatic products(341.4 μg/kg),eggs(255.9 μg/kg),dairy(106.7 μg/kg),meat(103.2 μg/kg),cereals (40.7 μg/kg),vegetables(30.7 μg/kg),beans(12.9 μg/kg),sugar(8.5 μg/kg),fruits(6.7 μg/kg),potatoes(2.4 μg/kg) and alcohol(2.1 μg/kg).The iodine content of kelp and laver was the highest in all the food,which was 314 780.1,176 956.5 μg/kg,respectively.The iodine content of food from animal(241.4 μg/kg) was much higher than that of the food from plant(25.4 μg/kg).The iodine content of common cereals,potatoes,beans,sea algae,meat,eggs and aquatic products was compared in the four areas,and the difference was not statistically significant (M =135.5,20.0,42.0,16.0,54.0,4.0,x2 =1.4,all P > 0.05).The iodine content of vegetables and fruits was compared,and the difference was statistically significant (x2 =8.5,M =204.5,all P < 0.05).Conclusion The iodine content of different foods is different.
3.Expressions and significances of CXCR4 and CD133mRNA in primary lesion of gastric adenocarcinoma

ObjectiveTo investigate the expression of CXCR4 and CD133mRNA in primary lesion of primary gastric adenocarcinoma and the relation with clinicopathological features,and to explore the correlation of CXCR4 and CD133.MethodsThe primary lesion of primary gastric adenocarcinoma and normal tissues adjacent to gastric cancer were obtained from 50 patients.The diction of CXCR4 and CD133 protein expression was detected by the immunohistochemical staining,and the relative gray scale values of CXCR4 and CD133mRNA by semi-quantitative RT-PCR (Fisher' s exact probability method).Their relationship with clinicopathological features was also investigated ( Spearman relation analysis).ResultsThe positive rates of CXCR4 and CD133 protein in gastric cancers were 76.0% and 66.0% respectively,which were significantly higher than that in normal tissues adjacent to gastric cancer ( 16.0% and 10% ; P =0.000,P =0.000).The increment of relative gray scale values of CXCR4mRNA was associated with the larger tumor diameter,the later TNM stage and the occurrence of lymphatic metastasis( P < 0.05 ).And the larger diameter of tumor,the later TNM stage were associated with the higher relative gray scale values of CD133mRNA (P <0.05).The levels of the relative gray scale values of CXCR4 mRNA and CD133mRNA were positively related(r =0.453,P < 0.01 ).ConclusionsThe higher expression of CXCR4 and CD133mRNA correlateswith tumour diameter,TNM stage and lymphatic vessel invasion. The relative gray scale values of CD133mRNA increase with the increment of the relative gray scale values of CXCR4.
4.Reduction of β-catenin expression in hepatocellular carcinoma inhibited the enhanced metastatic potential of hypoxia

ObjectiveTo explore the role of β-catenin in the proinvasive consequences of hypoxia in hepatocellular carcinoma (HCC).MethodsWe established in vitro and in vivo hypoxic models using the highly metastatic MHCC97 and the stable red fluorescent protein-expressing MHCC97-R cells.The role of β-catenin in hypoxia-mediated aggressiveness was investigated by β-catenin knockdown.ResultsHypoxia caused a pronounced arrest of proliferation in MHCC97 cells,suppressed tumor growth in MHCC97-R xenografts,but promoted in vitro invasiveness and in vivo metastasis.β-Catenin-silencing by short hairpin significantly inhibited the enhanced invasiveness of MHCC97 cells due to hypoxia,reduced the increase in distant metastasis by hepatic arterial ligation,but failed to further restrain cell proliferation.Conclusionβ-Catenin in HCC cells plays an essential role in the hypoxia-induced metastatic potential.A reduction of βcatenin expression inhibited the proinvasive consequences of hypoxia in HCC.
5.The research of p33~(ING1),wt-p53 growth suppressing and collapsing effect toward stomach cancer cell strain

Objective To discuss the growth suppressing, apoptosing effect of new type tumor-supressor gene-p33ING1 in stomach cancer cell strain, and to explore new strategies and methods in tumor therapy. Methods The PCDNA3/p33ING1 nuclear expressing microsome was constructed, p33ING1 and wt-p53 were implanted to human stomach cancer cell both and to evaluate the effect of p33ING1 and p53 toward stomach cancer cell and synergism between them. Results The PCDNA3/p33ING1 nuclear expressing microsome was successfully constructed. The human stomach cancer cell strain SSCG-7901 under implantation of p33ING1 and wt-p53 showed a significant decrease in cell growth, the coupling time was delayed, DNA synthetic phase was shortened and G0/G1 phase prolonged. The cell collapse increased. Conclusions Despite of the tumor-inhibiting effect and biochemical activation of p33ING1, it also plays a role with p53 gene in controling growth of stomach cancer cell, inducing cell collapse and hampering cell proliferation cycle. P33ING1 and p53 are synergistic to each other.
6.Influence of CD133+expression on patients' survival and resistance of CD133+cells to anti-tumor reagents in gastric cancer

Objective: To investigate the influence of CD133+expression on patients' survival and resistance of CD133+cells to anti-tumor agents in gastric cancer (GC).
Methods: Influence of CD133 expression on prognosis was analyzed employing sam-ples from patients with GC. GC cell lines were utilized to separate CD133+and CD133?subpopulations by immunomagnetic separation and to analyze the biological features of two subpopulations in vitro and in vivo, especially in resistant to anti-tumor reagents and its apoptotic mechanism.
Results: The lower CD133+group showed a significantly better survival compared with the higher CD133+group. The highest content of CD133+subpopulations for KATO-III cells had stronger proliferative ability than CD133?subpopulations. A single CD133+cell was capable of generating new cell colony and the tumorigenicity rate in nude mice was 100% for CD133+ clonal spheres or for CD133+ cells, but 0% for CD133? cells. Furthermore, the higher expression levels of Oct-4, Sox-2, Musashi-1 and ABCG2 in CD133+ clonal spheres were identified compared with CD133+ cells or CD133? cells. Under the treatment of anti-tumor reagents, CD133+ cells had lower suppression rates compared with CD133? cells while lower level of Bcl-2 and higher level of Bax were found in CD133+cells compared with CD133?cells.
Conclusions: The patients with lower CD133+expression had a better survival. Enriched CD133+ cells in clonal sphere shared the ability to be self-renewable, proliferative, tumorigenic and resistant to anti-tumor agents as probably regulated by Bcl-2 and Bax.
7.Effect of alprostadil combined with Salvia miltiorrhiza in treatment of hyperlipidemic acute pancreatitis

Objective To investigate the effect of alprostadil combined with Salvia miltiorrhiza in treatment of hyperlipidemic acute pancreatitis.Methods Patients with high cholesterol of hyperlipidemic acute pancreatitis were randomly divided into control group and treatment group,32 cases in each group.The control group were treated with conventional therapy combined with alprostadil 10μg intravenous injection,the treatment group was given Salvia miltiorrhiza injection 20mL vein infusion on the basis of the control group.The abdominal pain relief time,APACHE -II,C reactive protein levels,prognosis,hospitalization time and cost were compared between the two groups.Results The abdominal pain relief time,APACHE -II score,C reactive protein level in the treatment group were (5.31 ± 1.09)d,(2.34 ±1.18),(48.41 ±22.64)mg/L,which were better than those in the control group (8.16 ±1.39)d, (4,47 ±1.68)and (65.34 ±18.02)mg/L,the differences were statistically significant (t =9.08,0.14,5.84,-0.49,3.31,all P <0.05).There was no difference in mortality between the two groups (P >0.05).The operation rate of control group was 31.25% (10 /32),which was significantly higher than that of the treatment group [6.25%(2 /10)],the total hospitalization time,total cost of the treatment group were (14.50 ±1.55 )d and (4.97 ± 1.00)ten thousand yuan,which were significantly lower than those of the control group (16.78 ±1.83)d and (5.72 ± 1.71)ten thousand yuan,the differences were statistically significant (χ2 =6.65,t =1.00,t =5.39,all P <0.05). Conclusion Alprostadil combined with Salvia miltiorrhiza injection in the treatment of hyperlipidemia hyperlipidemia acute pancreatitis is safe,has good treatment effect,and can shorten the course of disease,reduce patients'hospitaliza-tion time and cost,and effectively improve the cure rate.

9.Interference of RNAi to CD133 gene and the comparison of the interferential effects in KATO-Ⅲ cells of human gastric cancer

Objective To compare the inhibition effects of three synthesized fragments used in small interfering RNA(siRNA) against CD133 gene in KATO-Ⅲ gastric cancer cells,and to study effects of suppressed CD133 on the proliferating ability of intervened cells.Methods Three fragments of siRNA were designed and synthesized targeted at the mRNA of CD133.Cell fluorescence counting under confocal laser scanning microscope was used to determine the transfection efficiency after transfection with the CD133FITC-siRNA.The knock-down effect of the CD133 gene was detected by RT-PCR and Western blotting.CCK-8 (cell counting kit-8 assay) was performed to measure the variation of the cell proliferative viability after the above-mentioned treatment.Results The transfection efficiency of siRNA was (85 ± 8) % in KATO Ⅲ Gastric cacer cell.All these three fragments of CD133 siRNA effectively inhibited the expression of CD133 gene,the inhibition rate being (11 ± 2) %,(19 ± 2) %,(24 ± 3) %respectively.Compared with the control group,the cell proliferation viability was restrained (42 ± 4)% in CD133siRNA-3 group (P <0.05).Conclusions CD133siRNAs were successfully transfected into KATO Ⅲ Gastric cacer cells and repressed the expression of CD133.Meanwhile,the CD133siRNA fragment 3 was screened from three CD133 siRNA,which has the best inhibition effect.The results provide preliminary evidence for the intereference of CD133+ gastric cancer cells subsequently.
10.Transforming growth factor-β1 generates epithelial-to-mesenchymal transition and promote CD44 expression in SGC7901 cells

Objective To investigate the biological effect of epithelial-to-mesenchymal transition (EMT) under the treatment of transforming growth factor (TGF)-β1 on the human gastric cancer cell line SGC7901 in vitro,and to observe whether the TGF-β1 can generate the tumor initiating cells ability in SGC7901 or not.Methods SGC7901 cells were cultured with TGF-β1.The morphological change was observed.The effect on proliferation of SGC7901 cells was detected by CCK-8.The invasion assay was used to investigate the motility and the invasion ability of SGC7901 cells.Immuofluorescence was used to detect the expression of E-cadherin and N-cadherin.The mRNA and protein's expression levels of EMT-related factors and CD44 were analyzed by RT-PCR and Western blotting respectively.Results TGF-β1 induced morphological alterations from epithelial to mesenchymal cells.The proliferation of SGC7901 cells was inhibited,and the ability of motility and invasion of SGC7901 cells were greatly enhanced after being treated with TGF-β1.RT-PCR and Western blotting showed that the expression of Snail (P < 0.05),N-cadherin (P < 0.05) and CD44 (P < 0.05) were significantly increased while the expression of E-cadherin was decreased (P < 0.05).Conclusions TGF-β1 can generate the EMT.The CD44 expression was up-regulated.TGF-β1 can inhibit the proliferation and promote the motility and invasion ability of SGC7901 cells.
