Objective To study the effects of Qi Jia Zhen Wu Decoction (QJZWD), a traditional Chinese medicinal decoction, in combination with corticosteroid on the apoptosis and expression of Fas, FasL in peripheral blood lymphocytes (PBLs) from patients with systemic lupus erythematosus (SLE). Methods QJZWD containing sera were collected from rabbits. PBLs isolated from 19 SLE patients and 10 healthy controls were incubated with QJZWD containing sera (20%, 10%, 5%), dexamethasone (0.001 mol/L), and QJZWD containing sera (20%, 10%, 5%) in combination with dexamethasone. Apoptosis and expression of Fas and FasL of PBL were detected by flow cytometry. Results ①Apoptosis of PBLs cultured in vitro was significantly higher in SLE patients than that in healthy controls (P 0.05). ②Significantly decreased apoptosis was found in PBLs cultured with herb-containing sera (10%, 20%) compared to that without herb-containing sera (P

Objective To investigate the effects of thiopentone and propofol on cerebral ischemia-reperfusion injury during open heart surgery under deep hypothermia circulatory arrest in infants and young children. Methods Twenty patients with VSD and pulmonary hypertension (13 male, 7 female), aged 3-23 months and weighing 4-11 kg were randomly divided into three groups: in group A thiopentone 5mg.kg was added in CPB machine when body temperature was reduced to 30℃ (n = 7); in group B propofol 2 mg.kg-1 was added ( n =8) and group C served as control ( n - 5). When rectal temperature was reduced to 20℃, CPB was stopped and intracardiac manipulation was started. Anesthesia was induced with midazolarn 0. 2mg.kg -1, fentanyl 20 ?g.kg1 and vecuronium 0. 1mg.kg1 . After tracheal intubation the patients were mechanically ventilated and anesthesia was maintained with O2-N2O-isoflurane and intermittent boluses of fentanyl and vecuronium infusion (70?g.kg-1.h1). Radial artery was cannulated and internal jugular vein (IJV) was retrogradely cannulated until bulb, and blood samples were taken simultaneously from artery and IJV before CPB (T1 ), during circulatory arrest (T2 ) at the beginning of reperfusion ( T3 ) and 24h after operation ( T4 ) for blood gas analysis and determination of plasma concentration of lactic acid (LA), creatine kinase-BB (CKBB) activity and neuron-specific endase (NSE). Cerebral arterial-venous O2 content difference (Ca-vO2) and cerebral O2 extraction ratio (CO2ER) were calculated. Results Plasma concentration of lactic acid was increased at T3 in all three groups and was the highest in group A. Cerebral O2 metabolism decreased at T2 in all three groups and was the lowest in group B and resumed at T4 CKBB activity was increased at T3 and NSE level was increased at T3 and T4 in all 3 groups. The increase in CKBB activity and NSE level was slightest in group B. Conclusion Propofol can protect brain from ischemia-reperfusion injury in infants and young children during open heart surgery under deep hypothermic circulatory arrest.
6.Effect of Cold-Stress on Rennin-Angiotensin System in Pregnant Rats
Journal of Environment and Health 1992;0(02):-
Objective To determine whether repeated cold-stress stimulation could activate rennin-angiotensin system in rats and lead to a PIH-like phenomena in pregnant rats. Methods Adult Wistar female rats were randomly divided into 4 groups: non-pregnant control group(NN), non-pregnant cold-stress group(NC), pregnant control group(PN) and pregnant cold-stress group(PC). Rats were kept for 4 hours in (4?2)℃(NC?PC) and in 25℃ (NN?PN) every day respectively, from the 1st day to the 19th day of pregnancy. The AngⅡlevels of plasma?uterus and placentas were determined by radioimmunoassay. The expression of angiotensin Ⅱ type 1 receptor (AT1R) and angiotensin Ⅱ type 2 receptor (AT2R) mRNA were determined in uterus and placentas of rats by RT-PCR. Results The AngⅡlevels of plasma, uterus and placentas were increased significantly in NC and PC compared with those in NN and PN. AT2R was the predominant type in the rat non-pregnant uterus. Downregulation of AT2R and upregulation of AT1R were observed during pregnancy. And at last, AT1R was the predominant type in pregnant uterus. The AngⅡ receptors in the rat placenta are predominantly AT1R. After cold-stress stimulation, the expression of AT1R mRNA in uterus and placentas of rats were higher than that of control groups(P0.05). Conclusions Repeated cold-stress stimulation could activate the renin-angiotensin system(RAS), and lead to a PIH-like phenomena in pregnant rats at last.

Objective To investigate the feasibility of gene delivery to the ischemic myocardium of rabbits by ultrasound-mediated destruction of microbubbles. Methods A ligation model of left circumflex branch coronary artery was established in 48 rabbits. Three days after ligation, the mixture of gene and microbubbles was infused into the vein of rabbits with or without simultaneous ultrasound. Additional controls included ultrasound of microbubbles that did not contain gene, gene alone, gene plus ultrasound, and blank control. Rabbits were killed after two weeks and examined for the expression of vascular endothelial growth factor(VEGF). Results The hearts of five rabbits that underwent ultrasound-mediated destruction of microbubbles containing gene showed VEGF expression in ischemic myocardium.None of the control animals showed VEGF expression. Conclusions Ultrasound-mediated destruction of microbubbles is a promising method for the delivery of bioactive agents to the heart.

Objective To explore the effect of bone marrow stromal cells (BMSC) transferred by Nell-1 gene combined with biomaterial fibrin glue (FG) to enhance segmental bone defect healing in dog mandible. Methods Nell-1 gene vector was reconstructed in retroviral vector and then transfected BMSC. The protein of Nell-1 gene in transferred cells was determined by immunohistochemistry. Segmental defects were created surgically in the dog’s mandible. The defect was repaired with BMSC transfected by Nell-1 retroviral granules in presence of FG,untransfected BMSC in combination with FG,and FG alone. The control group was left untreated. The defect-repairing capability for each treatment were assessed by gross observation, radiography, and histology at 8th week and 16th week. Results Cells transfected by Nell-1 retroviral granules expressed abundant Nell-1 mRNA and protein in the cytoplasm. Positive results were not found in those cells that were not transferred. The use of BMSC transferred by Nell-1 retroviral granules combined with FG materials exhibited the strongest defect-repairing ability. Gross observation, radiographical and histomorphometric analyses revealed a significantly greater total area of bone formation, increased amount of the new bone in the defects than in those treated with the untransfected BMSC. Conclusion Nell-1 gene transfection may be used to promote the osteogenic ability of BMSC.
