1.Expression of Interleukin-1β in Synovia and Synovium of Rats after High Intensity Exercise

Objective To study the expression of interleukin-1β (IL-1β) in synovia and synovium of rats after high intensity exercise.Methods 24 healthy male Sprague-Dawley rats were randomly divided into control group and high intensity group, and the latter were forced to run 60 min at the speed of 28 m/min for 6 weeks. The histological changes of cartilage from the lesion areas of medial femoral condyle and synovium of knee joint were examined with HE stain and the expression of IL-1β in synovium with immunohistochemistry, in synovial fluid with the enzyme-linked immunosorbent assay (ELISA). Results The articular cartilage of knee were injured and synovitis were found in high intensity group. The expression of IL-1β both in synovial fluid and in synovium of the knee were more in high intensity group than in control group (P<0.05). Conclusion High intensity exercise could cause aticular cartilage injury, synovitis and over expression of IL-1β in synovia and synovium, which may associated with post-traumatic osteoarthritis.
2.Effects of intra-articular hyaluronan injection on the expression of cartilage oligomeric matrix protein in synovium of strenuous running rats

Objective To investigate the effects of intra-articular hyaluronan(HA)injection on the expression of cartilage oligomeric matrix protein(COMP)in synovium of strenuous running rats,and investigate the possibility of predicting the effectiveness of HA based on COMP in synovium.Methods 36 healthy male Wistar rats were randomly divided into control group,strenuous running group and strenuous running group and HA injection group.Strenuous running group and HA injection group were intra- articularly injected with HA once a week for 5 consecutive weeks.The histological changes of synovium of knee joint was examined by H.E.staining and immunohistochemical expression of COMP in three groups after 6 weeks' strenuous running.Results Synovial inflammation was less severe in strenuous running and HA injection group than strenuous running group(t =7.15,P <0.01).The immunohistochemical expression of COMP in rats'synovium of knee joint in strenuous running and HA injection group was significantly lower than that in rats'synovium in strenuous running group(t = 6.30,P < 0.01).Conclusions Intra- articular HA injection suppressed synovitis,and the expression of COMP in synovium could be used to predict the effectiveness of HA.
3.The Expression and Significance of Myeloperoxidase in Acute Lungs Injury of Severe Acute Pancreatitis Associated Ascetic Fluid

Objective To investigate the expression and significance of myeloperoxidase(MPO) in acute lungs injury of severe acute pancreatitis associated ascetic fluid. Methods Forty-five adult wistar rats were randomly assigned into the group of negative control (group C,n=15),the group of severe acute pancreatitis (group S,n=15) and the group of peritoneal injection (group E,n=15). The group C was cut peritoneum and flipped pancreases softly. In group S,3.5% sodium taurocholate was injected retrograde in pancreatic and bile duct to establish the model of severe acute pancreatitis,and the pancreatic homogenate and ascites of the group S was injected into abdominal cavity of group E rats. After animal model established,rats were killed at 3h,6h and 12h point. The blood of inferior vena cava was sucked for determination of amylase.The inferior lobe of left lung was cut for myeloperoxidase detection.And pathology was regularly done about pancreas and lungs. Results Interstitial edema,hemorrhage and infiltration of neutrophilic granulocyte and macrophage were observed in group S and E. At different time point,the amylase levels of blood and myeloperoxidase of lungs in group S and E were significantly higher than those of group C,and the increasing degree of group E was smaller than group S. Conclusion Acute lung injury can be induced by the severe acute pancreatitis associated ascetic fluid. The expression of myeloperoxidase of lungs was increased to induce acute lungs injury.The reason may be concerned with activation of granulocyte by severe acute pancreatitis associated ascetic fluid.

5.Study on Rapid Methods for Quantitative Analysis Rhamnolipid and Its Influence Factors

In order to find an easy and rapid quantitative analytical method to detect rhamnolipid produced by Pseudomonas aeruginosa, three methods, H_ 2 SO_ 4 -anthrone analysis method, L-cysteine-H_ 2 SO_ 4 method and phenol-H_ 2 SO_ 4 method, were compared in the present paper, and the influence factors were also considered.The results showed that H_ 2 SO_ 4 -Anthrone analysis method was better than the others and its optimal reaction condition was obtained.The influence to the quantitative analysis of rhamnolipid from the residual glucose and the top clean liquid layer in the ferment solution could be ignored.But the influence from the bacterial body and the middle layer of the ferment solution reached a certain degree.Thus, the bacterial body should be removed before measuring.However, the influence from the middle layer of the ferment solution could be avoided by making a standard curve which was made by using a rhamnose mixed with the middle layer ferment solution.

7.Dynamic protein-protein interaction subnetworks of lung cancer in cases with smoking history.

Smoking is the primary cause of lung cancer and is linked to 85% of lung cancer cases. However, how lung cancer develops in patients with smoking history remains unclear. Systems approaches that combine human protein-protein interaction (PPI) networks and gene expression data are superior to traditional methods. We performed these systems to determine the role that smoking plays in lung cancer development and used the support vector machine (SVM) model to predict PPIs. By defining expression variance (EV), we found 520 dynamic proteins (EV>0.4) using data from the Human Protein Reference Database and Gene Expression Omnibus Database, and built 7 dynamic PPI subnetworks of lung cancer in patients with smoking history. We also determined the primary functions of each subnetwork: signal transduction, apoptosis, and cell migration and adhesion for subnetwork A; cell-sustained angiogenesis for subnetwork B; apoptosis for subnetwork C; and, finally, signal transduction and cell replication and proliferation for subnetworks D-G. The probability distribution of the degree of dynamic protein and static protein differed, clearly showing that the dynamic proteins were not the core proteins which widely connected with their neighbor proteins. There were high correlations among the dynamic proteins, suggesting that the dynamic proteins tend to form specific dynamic modules. We also found that the dynamic proteins were only correlated with the expression of selected proteins but not all neighbor proteins when cancer occurred.

8.Correlation between anxiety among public hospital medical personnel and social support, perceived social support and coping styles

Objective This research aimed to explore the mental status of public hospital medical personnel and social support for them, social support they perceive, and their coping styles. This study was designed to investigate on the relationship between their anxiety and social support, perceived social support and their coping styles. Methods A questionnaire was adopted to collect data, including participants’ background, measurement of anxiety, as well as social support, perceived social support and their coping styles. Results(1)The anxiety level of medical personnel was significantly higher than that of domestic norm 37.23±12.59(t=14.370, P<0.001);(2)Anxiety levels appeared to be significantly different in occupation, educational background and working years;(3)Results shown in the three scales were different among participants with different levels of anxiety in perspectives of every dimension in the scales; (4) With the help of Spearman correlation analysis, it showed that anxiety levels of medical personnel was negatively correlated with objective support (r=-0.098, P=0.051), subjective support (r=-0.189, P<0.001) ,utilization of social support (r=-0.164, P=0.001), friend support (r=-0.356, P<0.001),family support (r=-0.330, P<0.001),other support (r=-0.238, P<0.001) and positive response (r=-0.282, P<0.001), and positively correlated with negative response (r=0.385, P<0.001). Conclusions Mental health of medical personnel should be noticed and enhanced, and social support for public hospital medical personnel should be strengthened. Thus, the performance of the medical team can be developed in order to support further public medical service construction.
9.Antiviral activity of 3-hydroxyphthalic anhydride-modified ovalbumin against herpes simplex virus 2 in vitro.

OBJECTIVETo investigate the antiviral activity of 3-hydroxyphthalic anhydride-modified ovalbumin (HP-OVA) against herpes simplex virus 2 (HSV-2) in vitro.
METHODSBy chemical modification, ovalbumin (OVA) was treated with 3-hydroxyphthalic anhydride (HP) to prepare HP-OVA. The anti-HSV-2 activity against HSV-2 333 virus in vitro and the cytotoxicity of HP-OVA in African green monkey kidney cells (Vero cells) were detected by MTT colorimetric assay. The inhibitory effects of HP-OVA on 17 strains of vaginal lactobacilli were observed by microscopy.
RESULTSAnhydride-modified ovalbumin significantly inhibited the infection by HSV-2 with an IC(50) of 23.56±8.33 µg/ml. HP-OVA showed only low cytotoxicity to the host cells with a CC(50) over 1 mg/ml. HP-OVA did not produce significant inhibitory effect on the 17 strains of vaginal lactobacilli (MIC>1 mg/ml).
CONCLUSIONAnhydride-modified protein HP-OVA exhibits potent anti-HSV-2 activity in vitro and can be a good microbicide candidate for prevention of sexually transmitted diseases.

10.Advances in the study of derivatization of ginsenosides and their anti-tumor structure-activity relationship.

Ginsenosides, belonging to a group of saponins with triterpenoid dammarane skeleton, show a variety of pharmacological effects. Among them, some ginsenoside derivatives, which can be produced by acidic and alkaline hydrolysis, biotransformation and steamed process from the major ginsenosides in ginseng plant, perform stronger activities than the major primeval ginsenosides on inhibiting growth or metastasis of tumor, inducing apoptosis and differentiation of tumor and reversing multidrug resistance of tumor. Therefore ginsenoside derivatives are promising as antitumor active compounds and drugs. In this review, the derivatization methods, ginsenoside derivatives and their anti-tumor structure-activity relationship have been summarized for providing useful information for the research and development of novel antitumor drugs.
