3.Preliminary study of odor change mechanism in Crataegi fructus stir-fried process based on correlation analysis.

In order to investigate the mechanism, the correlation between the odor change in Crataegi Fructus stir-fried process and 5-HMF were studied. Required samples were retrieved from Crataegi Fructus stir-fried process. Statistical quality control (SQC) was used to analyze the response values acquired by the electronic nose. At the same time, the content of 5-HMF was detected by high performance liquid chromatography (HPLC). Correlation analysis was used to analyze the relationship between the above two. Experimental results showed that SQC model established by response values of all samples could show the change law of odor in Crataegi Fructus stir-fried process and changes of 5-HMF content was dropped after the first increase. Correlation analysis showed that the odor change in Crataegi Fructus stir-fried process and 5-HMF were significantly correlated (P < 0.05). Sugar degradation reaction and the Maillard reaction may be one of the mechanisms of the odor change in Crataegi Fructus stir-fried process.

5.The expression of chicken interleukin 2 in Pichia yeast

Objective To express recombinant chicken interleukin 2 (IL-2) in Pichia yeast eukaryotic cell expression system for a massive preparation of chicken IL-2. Methods Spleen lymphocytes taken from Xiaoshan chicken were activated by ConA in vitro. Chicken IL-2 gene was amplified and cloned into pPIC9. The pPIC9-IL-2 was transformed into Pichia yeast by electroporation. The recombinants were induced by methanol for the expression of recombinant protein. Results The protein was examined with SDS-PAGE and Western bloting. A specific molecular weight band at about 30 ku appeared. Conclusion The recombinant protein of chicken IL-2 has been expressed in Pichia yeast.
6.Effect of Warm Needling Moxibustion on Bone Density, BGP and IL-6 in Osteoporotic Compression Fracture

Objective To investigate the therapeutic effect of medication-based acupuncture on osteoporotic compression fracture and explore clinical effective intervention in the patients. Method Fifty-six patients were randomized to acupuncture-medicine and medicine groups, 28 cases each. The clinical therapeutic effects were evaluated after treatment. Bone density, BGP and IL-6 were measured before and after treatment. Result BGP and IL-6 decreased significantly in the two groups after treatment compared with before (P<0.01) and decreased more in the acupuncture-medicine group than in the medicine group (P<0.05). BMD increased significantly in the acupuncture-medicine group after treatment compared with before (P<0.05) and did not increased significantly in the medicine group after treatment (P>0.05). Conclusion Warm needling moxibustion plus medication is effective than medication alone in treating osteoporotic compression fracture. It can decrease serum BGP and IL-6 contents and relieve the clinical symptoms in the patients. Acupuncture plus medicine not only produces a marked analgesic effect but also has a positive role in preventing the aggravation of osteoporosis and avoiding second fracture.
7.The reverse genetics systems for human and animal RNA viruses.

The recovery of the virus from genetic materials in in vitro culture systems or sensitive animals is called virus rescue. A functional infectious clone of RNA virus provides unlimited possibility for genetic studies and the related reverse genetics system that allows directed genetic manipulation of an RNA virus is an extremely powerful research tool. In the past twenty years, especially since the first infectious clone of a negative-stranded RNA virus was reported in the mid-1990's, the reverse genetics systems have been available for nearly all the major human and animal RNA virus groups. The article reviews the progress of this technology, highlighting the obstacles in the construction of reverse genetics systems for major groups of human as well as animal RNA viruses and how the virologists overcame them. There are mainly four external expression systems for construction of the RNA virus reverse genetics systems basing on the kind of RNA viruses. These systems include in vitro RNA transcripts, RNA polymerase I-driven expression plasmids, RNA polymerase II-driven expression plasmids and modified vaccinia virus/T7 RNA polymerase-driven expression system. In particular, the viral nucleoprotein and polymerase proteins are required to assemble the viral ribonucleoprotein (RNP) complexes for the rescue of the negative-stranded RNA viruses. Relevant topics about the rescue of the typical viruses are discussed, including poliovirus with the de novo synthesis, Coronaviridae with the largest size of genome, Flaviviridae with the instable clones, HCV with the quasispecies nature, nodaviruses with the virus-host interaction, influenza virus with the RNA pol I transcription system, Arenavirdae with the ambisense coding strategies etc.

8.Expression of hypoxia inducible factor-1alpha in long bone development.

OBJECTIVETo observe dynamically the development of fetal long bone and detect the expression and distribution of HIF-1alpha,to investigate the expression pattern and possible effects of hypoxia inducible factor-1alpha (HIF-1alpha) in fetal long bone development of mouse.
METHODSE12.5, E13.5, E14.5, E15.5, E16.5 and E17.5 pregnant C57BL6 mice were sacrificed. After sacrifice, the embryos were delivered by caesarean section. The development of fetal long bone was dynamically observed by stereoscopic microscope, and the distributional expression of HIF-1alpha protein was detected by using method of immunohistochemistry. The expression of HIF-1alpha mRNA and osteoblast marker gene at various stage were also detected by using methods of reverse transcription-polymerase chain reaction (RT-PCR).
RESULTSThe cartilaginous long bone began to form and joints outline arised at E13.5, then the primary ossification center was observed at E14.5, showing opaque ossification under stereoscopic microscope,and then the osteogenesis expanded and extended to both sides. Immunohistochemistry demonstrated lots of HIF-1alpha protein positive chondrcytes in the center of primary ossification at E14.5, then they decreased dramatically. HIF-1alpha mRNA expressed at high level from E13.5 to E15.5, and then decreased to low level.
CONCLUSIONFetal long bone development pattern appeared to be endochondral osteogenisis process, existing hypoxia microenviroment may increase HIF-1alpha mRNA expression and thus initiate the cascade of endochondral osteogenisis.

9.Applications of chitosan and its derivatives in pharmaceutical industry of Chinese medicine.

Recently, chitosan has been widely used as a clarifier in clarification procedure for many kinds of TCM, which is better than alcohol in retaining active substances, such as polysaccharids, as well as the removal of heavy metals. Because it is a biocompatible polymer with low toxicity and can be biodegradable in vivo, chitosan with its derivatives is one of the ideal materials for drug controlled release systems to enhance the efficacy and reliability of TCM drug therapy. On the other hand, chitosan, the only natural alkalic polysaccharid, also has several therapeutic effects such as anti-bacteria, antipyrotic and ulcer healing. Chitosan and its derivatives wide application prosperity in pharmaceutical industry of Chinese medicine.

10.Comparison of strain of mental workers by characteristics.

OBJECTIVETo compare of strain of mental workers by characteristics.
METHOD851 participants were sampled with stratified cluster random methods from various kinds of occupations, including teachers in primary and middle schools, doctors, bankers and environmental protection officers. The psychological strain was measured with SCL-90 and General Well-being Schedule (GWB). The physiological strain were assessed by Plasma sugar, TG, HDL, VLDL, LDL, CHOL, cortisol and ACTH level, neurobehavioral reaction function.
RESULTSThe subjects at age of 40-years exhibited the highest scores of SCL-90 significantly among three groups. Likewise, 20-years service group were higher than 10- and under 10 years groups. However females' GWB scores were higher than males'. It tended to be higher in postgraduate and undergraduate teachers than others groups, indicating the greater psychological strain increased, the higher age and service were. Plasma sugar, LDL, TG and CHOL level were compared; Subjects 40- years group were higher than 30- and 20 age groups, as well as senior versus minor. Female teachers were lower than males. Meanwhile single subjects were lower than married. As HDL level, 30-age group were lower decreased than 40-years group; 10-service group were responsible for lower HDL level than 20-years group; Divorced and spouse-lost teachers were higher concentration than single and married. VLDL level in under 10 years service were lower than 10-years group; The senior was the highest level of VLDL among three groups and the minor was the lowest. The tendency was equal to increased professional title; Married and spouse-lost groups were higher than single groups. It had no individual difference in plasma cortisol and ACTH. But reaction sensitivity in 40- and 30-age groups were lower than 20-age group. Likewise, 20- years service teachers had lower reaction and action sensitivity than 10- and under 10 years groups; Senior group was lowest among three groups; However 40- years old group were lower than 20- years group, postgraduate and undergraduate teachers were higher than others groups in muscle endurance. It indicated that reaction and action sensitivity had descent tendency with increased age and service length.
CONCLUSIONPsychological strain is higher with increased age, length of service and education career. The trend is the same as plasma sugar, TG, CHOL, HDL, LDL and VLDL, but neurobehavioral reaction function is decreased. In addition general well-being feeling of females is lower than males.
