1.Mutations in a Large Pedigree with Y-STR Genetic Markers.

OBJECTIVE:
To explore the mutation of Y-STR loci in meiotic allelic transmission in a large pedigree.
METHODS:
The oral swabs of 163 male individuals were collected from a Lin pedigree. Twenty-two Y-STR genetic markers were typed with AGCU Y24 fluorescent detection kit (AGCU Y24 system), which also contained 16 Y-STR markers included in Yfiler multiple amplification kit (Yfiler system). The genotyping results of Y-STR loci were compared between each two males in the pedigree.
RESULTS:
There were 20 and 30 kinds of haplotypes obtained with Yfiler and AGCU Y24 systems in 163 male individuals from the Lin pedigree, respectively. The rates referred to haplotype differences (RRHD) of these two typing systems between male pairs were 0.910 5 and 0.922 7, respectively. The average number of marker differences were 6.582 1 and 9.824 8, respectively. The RRHD increased along with the incidents of meiosis.
CONCLUSION
Y-STR mutation leads to different Y-STR haplotypes among the male members in a paternal pedigree and the rate of difference increases along with the incidents of meiosis.

3.Effects of intensive insulin treatment on the prognosis of severe multiple trauma patients

16 and the distribution of severe trauma more than 2 anatomic parts.They were randomly divided into two groups:intensive insulin treatment group(n=31)and control group(n=31).Intensive insulin treatment group received insulin with insulin pump in order to maintain blood glucose levels at 4.0-6.1 mol/L,while the control group received routine insulin treatment in order to mmaintain blood glucose levels at 10.0- 11.0 mol/L.Plasma levels of TNF-?,IL-1,IL-6, CRP,APACHEⅡscores and cure rate were analyzed before and after the treatment.Data was expressed as mean?standard deviation.Two- tailed T test and ANOVA were used for comparison in SPSS 10.0,and changes were considered as statistically significant if P value was less than 0.05.Results After the intensive insulin treatment, patient's hemodynamic parameter apparently improved,APACHEⅡscores descended,and the levels of TNF-?, Ib-1,IL-6,CRP all declined,in comparison with control group,there were significant differences. Intensive insulin treatment might improve patient's general condition and decrease complications and mortality of severe multiple trauma.

5.Effects of combined arsenic trioxide and resveratrol on the viability of human acute promyelocytic leukemia cell line NB4 cells

Objective To investigated the effects of combined arsenic trioxide(ATO) and resveratrol(Res)on the viability of NB4 human leukemia cells. Methods NB4 human leukemia cell was used in this experiment.Cells were cultured in ATO (0,0.1875,0.3750,0.7500, 1.1250, 1.5000,2.2500,3.0000,5.0000 μmol/L) and Res (0, 1.5625,3.1250,6.2500, 12.5000, 18.7500,25.0000,37.5000,50.0000 μmol/L). Cell viabilities were measured by MTT in different treatment groups. Half inhibitory concentration(IC50) was calculated. The ratio of concentration of ATO and Res 1.5∶ 18,1.5∶ 25,1.5∶ 35 was added to cells, and the combination index(CI) was calculated. The level of ROS in control, ATO( 1.5000 μmol/L), Res(25.0000 μmol/L) and ATO(0.9000 μmol/L) + Res( 12.5000μmol/L) groups was measured by chemiluminescence assay. Results ①ATO( ≥0.7500 μmol/L) reduced the viability of NB4 cells in a concentration-dependent manner(P < 0.05 ), and IC50 was (1.78 ± 0.11 )μmol/L. ②)Res (≥18.7500 μ mol/L) dose-dependently decreased the viability of NB4 cells (P < 0.05 ), and IC50 was ( 18.71 ±0.18)μ mol/L. ③Combination of ATO and Res showed an antagonistic effect on NB4 cells viability. ④The ROS in Res group( 1670.55 ± 13.97) was significantly lower than that in control group(2345.88 ± 14.48,P < 0.05). The ROS in ATO group (3092.42 ± 94.84) was significantly higher than that in control group(P < 0.05). The ROS in ATO + Res group (1860.27 ± 15.99) was significantly lower than that in ATO group(P < 0.05). Conclusions NB4 cell survival rate can be decreased by ATO and Res. The combination of arsenic trioxide and Res presents an antagonistic effect on NB4 cell viability, in part by reducing intracellular ROS formation.

Based on the literatures published from January 1994 to December 2008 on chronic respiratory diseases treated by the therapy of “Dongbing-Xiazhi” (which means treating diseases occurred in winter but treated in summer),the time,region,specific therapies,herbs,and acupoints of these literature were reviewed end analyzed by bibliometric methods to explore its distribution and trends.The research showed that the therapy was effective and widely used in recurrent chronic respiratory diseases.It also demonstrated that such studies as focusing on the scientific design and the mechanism were needed.
7.Study on the diagnostic test with C-reactive protein in elderly patients with infections.

OBJECTIVETo evaluate the value of C-reactive protein (CRP) to diagnostic test in elderly patients with infections.
METHODSC-reactive protein were investigated in 142 elderly patients with infections and 216 elderly patients without. CRP 7 - 20, 21 - 40 and 41 - 60 mg/L were stratified, the index of diagnostic test counted.
RESULTSConcentrations of CRP in patients with different diseases were upper respiratory tract infection 36.9 mg/L +/- 28.9 mg/L, acute bronchitis 30.1 mg/L +/- 28.1 mg/L, pneumonia 55.9 mg/L +/- 32.9 mg/L, urinary infection 49.0 mg/L +/- 27.6 mg/L and enteritis 39.3 mg/L +/- 35.6 mg/L. They were all higher than those in control group (5.2 mg/L +/- 2.9 mg/L, P < 0.001). Stratified analysis disclosed that the specificity of CRP was 83.3% - 99.0% for diagnostic infection disease. The positive likelihood ratio (LR) of 7 - 20, 21 - 40 and 41 - 60 mg/L were 3.6, 27.0 and 128.0, respectively.
CONCLUSIONC-reactive protein was an important marker to diagnose elderly patients with infections.

8.Excretory/secretory antigens from Clonorchis sinensis induces hepatic fibrosis in rats.

OBJECTIVETo investigate the role of excretory/secretory antigens from Clonorchis sinensis (CsESAs) in hepatic fibrosis induced by C. sinensis infection in rats and explore the possible mechanism.
METHODSCsESAs was collected from adult C. sinensis cultured in sterile condition for 12 h and injected intraperitoneally in Wistar rats. Masson staining was used to observe the changes in the hepatic collagen fiber after the injection. HE staining and immunofluorescence staining were performed to detect the expression of alpha-smooth muscle actin (alpha-SMA) to examine the proliferation and the activity of hepatic stellate cells. The specific antibody titer of CsESAs was determined using enzyme-linked immunosorbent assay to investigate the role of the antigen-antibody complex in the development of hepatic fibrosis.
RESULTSAfter intraperitoneal injection of CsESAs, obvious hepatic fibrosis and hepatic stellate cell proliferation and activation were observed in the rat livers. The severity of the hepatic fibrosis was associated with the dose of CsESAs injected, whereas the titer of the specific antibody against CsESAs showed no direct relation to the hepatic fibrosis.
CONCLUSIONIntraperitoneal injection of CsESAs can cause hepatic stellate cell activation and hepatic fibrosis in rats, but the antigen-antibody complex does not seem to play the key role in the activation of the hepatic stellate cells.

9.Visualization of proliferation activity of luminescent bacteria in different physiological media

Objective To investigate the relationship between the number of colonies and the luminous intensity of two kinds of platelets contamination bacteria,Escherichia coli(ECO)and Pseudomonas aeruginosa(PAE)in single platelets, plasma and phosphate buffered salines(PBS), and to explore their growth and proliferation in different biological fluid media.Methods The two luminescent bacteria were constructed before the correlations between the number of colonies and the luminous intensity in different media in vitro were observed with a small animal bioluminescence imaging instrument, and correlation analysis was carried out.By recording the luminous intensity at serial time points in different media, a growth curve was drown to reflect the proliferation of bacteria.Results There was a significant positive correlation between the number of colonies and luminous intensity of ECO and PAE in single platelets,plasma and PBS.The larger the number of colonies was,the stronger the luminous intensity was.The results indicated that single platelets significantly inhibited the proliferation of ECO and PAE,and that plasma also had some inhibitory effect,but not so strong.Conclusion For the two luminous bacteria,luminous intensity can represent the growth and proliferation of bacteria.Single platelets have obvious inhibitory effect on the growth of ECO and PAE, and the plasma also has some inhibitory effect, but the effect is not so strong as that of single platelets.
10.Study of phosphorylated ERK1/2 on nasal polyps.

OBJECTIVE:
To study the expression of phosphorylated extracellular regulator kinase (ERK1/2) on nasal polyps.
METHOD:
The immunohistochemistry and Western-blot were used to study the phosphorylation of ERK1/2 on the normal nasal mucosa and nasal polyps.
RESULT:
The phosphorylated ERK1/2 was mainly expressed on the hyperplastic epithelium, inflammatory cells, glandular epithelium and endothelium of nasal polyps by immunohistochemistry, which was much stronger on nasal polyps than that of normal mucosa. The expression of phosphorylated ERK1/2 was 4.2 times than that of normal mucosa.
CONCLUSION
The study indicated that the phosphorylation of ERK1/2 might play an important role on the occurrence and progression of nasal polyps.
