ObjectiveTo analyze the changes of plasma concentration of calcitonin gene-related peptide (CGRP) and substance P (SP) after electrical stimulation on dura mater adjacent to sinus sagittalis superior (SSS) in rats.Methods13 rats were randomly divided into control group(n=6) and stimulation group(n=7) in which the dura matter adjacent to SSS was stimulated electrically with square wave of 20 Hz, 4 mA and 250 μs. The plasma concentrations of CGRP and SP in both groups were compared by means of radioimmunity method.ResultsThere was significant difference in the plasma concentration of CGRP between control and stimulation groups (50.60±7.16 pg/ml vs 42.20±4.82 pg/ml, P=0.029). However, there was no significant difference in the plasma concentration of SP between two groups (308.59±43.07 pg/ml vs 264.60±60.28 pg/ml, P=0.165).ConclusionPlasma concentration of CGRP decreased after electrical stimulation on dura mater in rats but the concentrations of SP unchanged.

Using MS as basic medium, supplemented with 1.0 mg · L(-1) IBA, the adventitious roots of Tripterygium wilfordii were induced, and the good adventitious root culture system was established by leaves or callus induced by leaves as explants. The adventitious roots were also induced with 2.0-4.0 mg · L(-1) NAA and the good adventitious root culture system established by using suspension cells from callus as materials to induce adventitious root. The content of triptolide of three adventitious roots culture system were exceeded in the natural root bark. The content of triptolide of AR3 adventitious roots was the highest about 5.3 times as that in the natural root bark. By using 5 L stirred fermentor during pilot enlarge cultivation, compared with 250 mL flask cultivation, the adventitious roots increment and secondary metabolites content per liter medium showed no significant difference. The accomplishment of this analysis laid a foundation by tissue culture production of the secondary metabolites of T. wilfordii.

Chinese Journal of Orthopaedics 2013;33(10):997-1003
Objective To investigate anterior migration of spinal cord after anterior cervical decompression and reconstruction for cervical spondylosis and its mechanism. Methods Eighty-one patients with cervical spondylosis who underwent anterior decompression and reconstruction were reviewed and evaluated retrospectively. There were 44 males and 37 females, with an average age of 52.3 years (range,37-72 years). The distance from anterior margin of the spinal cord to posterior wall of the spinal canal (Da),and medium of the spinal cord to posterior wall of the spinal canal (Dm) in magnetic resonance imaging were measured preoperatively and postoperatively. The recovery rate was calculated comparing the Japanese Orthopaedic Association (JOA) scores before and after operation. Results The average following up was 20.7 months (9-48 months).According to the measure on MRI,preoperative Da and Dm were 11.11±1.59 mm and 6.30±0.84 mm respectively, which were significant different from postoperative Da 11.63±-1.47 mm and Dm 6.62±0.80 mm.The postoperative Da and Dm at C3-4,C4-5,C5-6,C6-7 levels had statistically significant differences compared to preoperative Da and Dm,except for ones at C2-3 level. The significant migration rate was 43.2%(35/81),which reached △Da >2 mm or △Dm >1.3 mm.Preoperative cervical curvature index (CCI) was 10.0%±6.3%,which was significant different from postoperative CCI 13.5%±6.5%.JOA score increased from 9.7±1.5 preoperatively to 13.7±1.1 at one week postoperatively, and 13.9±1.3 at 6 months postoperatively. But the JOA score decreased at 6 months postoperatively compared with that at 1 week postoperatively in 15 cases.Among them,14 cases got the standard of spinal cord migration based on MRI measurement. No other severe complications were found in these 81 cases. Conclusion Anterior migration of the spinal cord commonly occurred after cervical decompression and reconstruction via anterior approach, and some patients might suffer from the secondary compression of the cervical spinal cord.

Objective:In this study,a prokaryotic expression of the 3-ketosteroid-Delta (1)-dehydrogenase (KSDD) which came from Arthrobacter simplex was built. Moreover,in order to investigate the catalytic mechanism of KSDD and improve its stability, the structure of KSDD was predicted by computer and the critical sites were confirmed by site-directed mutations. Methods:The recombinant plasmid was constructed by eukaryotic expression vector pET-22b and the recombinant strain was constructed and expressed in Escherichia coli BL21 (DE3).High-performance liquid chromatography was used to determine the transformation rate of 4-AD to ADD.The KSDD structure and key sites were predicted by SWISS-MODEL.Site-directed mutations for the amino acid residues of key sites were constructed and activities of the mutations were detected. Results:The recombinant strain E.coli pET-22-ksdd was successfully constructed. It was induced to express the dehydrogenase by IPTG and the conversion rate of 4-AD to ADD was 27% at 21 ℃.The structure of 3-ketosteroid-Delta (1)-dehydrogenase and the four key sites was analyzed by SWISS-MODEL.Four mutants,Y120R,Y320L,Y488F and G492Y were constructed.Mutants Y120R and Y488F were inactivated,so they were proved to be the key active sites of KSDD.The conversion rate of mutant Y320L was consistent with that of wild type, but the stability at 37 ℃ was improved. The conversion rate of mutant G492Y was 1.2 times of the wild type and the stability has been improved at 37 ℃.Conclusions:At present, there are few studies about the structure and catalytic mechanism of dehydrogenase. The active sites of the enzyme were verified by this study, which laid the foundation for the further study of the properties of the enzyme KSDD.
