Ovbective To evaluate whether the deleterious effect of cardiopulmonary bypass (CPB) mediated by oxygen free radicals could be prevented by controlling PaO 2 in cyanotic children. Methods Cyanotic pediatric patients undergoing cardiac surgery (n=38) were randomly allocated into 2 groups. The groups matched in respect to sex,age,body surface area and preoperative ventricular function. Group H (n=18) had CPB initiated at a FiO 2 of 0.8,group L (n=18) had CPB initiated at a FiO 2 of 0.4. SOD and MDA were determined in arterial blood sample before CPB,30 minutes after initiation of CPB and 30 minutes post CPB. Various pulmonary parameters were determined 10 minutes after intubation,30 minutes after CPB,the 2nd hours and the 6th hours in the intensive care units. Results At the 6th hours in ICU,the mean postoperative increase in peak airway pressure and P(A-a)O 2/PaO 2 in group H was significant higher than that in group L(P

Objective To investigate the expressions of p53, bcl-2 and caspase-3 in elderly patients with advanced non-small cell lung cancer (NSCLC), and the clinical significance thereof. Methods The clinical data and paraffin tissue samples of 117 patients with NSCLC confirmed by postoperative pathology were collected. The expressions of p53, bcl-2 and caspase-3 protein in tissues of NSCLC were detected by immunohistochemical method. The correlation between the expres-sions of p53, bcl-2 and caspase-3 protein was analyzed, and the relationship between the expressions of p53, bcl-2 and cas-pase-3 protein and clinicopathological characteristics was explored. Results The positive expression rates of p53, bcl-2 and caspase-3 were 42.74%(50/117), 38.46%(45/117) and 36.75%(43/117) in tissue samples of NSCLC. The positive ex-pression rate of p53 was significantly higher in patients with poor differentiation, central type, lymph node metastasis and squamous carcinoma. The positive expression rate of bcl-2 was significantly higher in patients with high differentiation and lymph node metastasis. The positive expression rate of caspase-3 was significantly higher in patients with high differentia-tion and no lymph node metastasis. There was a positive correlation between expressions of bcl-2 and caspase-3 in NSCLC (r=0.262, P=0.003). Conclusion The expressions of p53, bcl-2 and caspase-3 may play important roles in development and progression of advanced NSCLC in elderly patients, which may serve as predicators for prognosis.

Objective To obtain SGC7901 gastric cancer cell-dendritic cell fusion vaccines by cell fusion technology. The phenotype changes of the fusion cells were detected to provide experimental data for the following research work. Methods The SGC7901 gastric cancer cells and dendritic cells were induced to be fused by PEG and the pure fusion cells were obtained by selective culture with the HAT/HT culture system. The phenotype changes of the fusion cells were detected by flow cytometer. Results The fusion cells had special morphologic characteristic and the phenotypes of the fusion cells were remarkably higher than the parental dendritic cells. Conclusion The SGC7901 gastric cancer cell-dendritic cell fusion cells kept special morphologic characteristic and their phenotypes were significantly higher than the parental dendritic cells, which may be the cellular base for their stronger biological effects.

Objective To study on the cytotoxic effects in vivo and in vitro of the T lymphocytes which were activated by the DC-gastric cancer cell fusion vaccines. Methods The mononuclear cells were separated from the peripheral blood of gastric cancer patients and co-cultured with granulocyte-macrophage colony stimulating factors, interleukin-4 and tumor necrosis factor-? to generate mature dendritic cells. The human gastric cancer SGC7901 cells and the dendritic cells were fusioned by using polyethylene glycol, and the pure fusion cells were screened out by culturing with HAT and HT selective culture systems. The mixture of SGC7901 and DC without fusion and the T lymphocytes served as controls. The ability of fusion cells activated by T lymphocytes to kill SGC7901 gastric cancer cells was investigated by MTT in vitro. The anti-tumor effects were evalutated by detecting the growth of the new planted tumors in the nude mice, the apoptosis and proliferation of the planted cancer cells, the pathological changes of the tumor tissues and the prohibitive rate for planted tumors before and after the application of the fusion cell vaccine. Results The mature dendritic cells were obtained from peripheral blood mononuclear cells of gastric cancer patients successfully. Dendritic cells and SGC7901 cells were fusioned and the pure fusion cells were got. Objective To study the regeneration regularity of neuropeptide Y positive(NPY+) nerves in splenic autotransplantation. Methods Healthy Wistar rats were randomly divided into two groups:(1)splenectomy and splenic autotransplantation group;(2) sham-operation group. Regeneration process and density changes of the NPY+ nerve fibers in the two groups were studied by immunohistochemical methods and computer image analysis qualification on day 7, 14, 30, 60, 90, 120 and 180 postoperatively. Results The NPY+ nerve fibers had no regeneration on day 30 postoperatively, but on day 60 nerve fibers appeared in the peri-region of the autotransplanted spleen, on day 90 nerve fibers extended into the inner-part of splenic autotransplants. Density of the nerve fibers gradually became greater and almost normal on 180 days after operation. Conclusions After splenic autotransplantation, the NPY+ nerve fibers could be regeneration in the autotransplants tissue. The renascent nerve fibers came from greater omentum which wrapes the splenic autografts.

Objective To investigate the exercise performance in maintenance dialysis patients,and analyze its correlative factors.Methods Maintenance dialysis patients admitted in Tongji Hospital of Shanghai from December 2014 to March 2015 were enrolled,with their baseline data and biochemical measurement being collected.The anthropometric indexes including arm circumference,triceps skinfold,waist circumference and hip circumference were detected.The exercise activity was assessed by hand grip test,timed up and go test (3mTUG) and five times sit-to-stand test (FTSST).Patients were divided into fast group (3mTUG≤12 s) and slow group (3mTUG ＞ 12 s).Univariate and multivariable analyses were used to evaluate the factors influencing exercise performance in maintenance dialysis.Results There were 121 patients enrolled:62 on peritoneal dialysis and 59 on hemodialysis,76 men and 45 women.Patients' average age was (61.6± 13.0) years and median dialysis age was 31.7(12.3,69.0) months.There was no statistical difference between fast group (n=80) and slow group (n=41) in gender,dialysis method,dialysis age,body mass index (BMI),arm muscle area (AMA),waist-hip ratio (WHR),hemoglobin (Hb) and total cholesterol (TC).Patients in fast group were younger,had higher serum albumin,prealbumin,serum phosphate and iPTH,and less prevalence of diabetes than those in slow group.In exercise activity,patients in fast group had better performance in handgrip,3mTUG and FTSST (all P ＜ 0.05).Univariate analysis showed that,handgrip was correlated with sex (male),AMA,BMI,age,diabetes,serum phosphorus and TC;scores in FTSST was correlated with age,BMI,diabetes,WHR,dialysis method,dialysis age,prealbumin and serum phosphorus;scores in 3mTUG was correlated with age,diabetes,WHR,dialysis method and dialysis age,prealbumin,serum phosphorus and iPTH (all P ＜ 0.05).Multiple stepwise regression analysis showed that sex (male),age,AMA and diabetes were independently correlated with handgrip in dialysis patients (all P ＜ 0.05);age,dialysis method,BMI and diabetes were independently correlated with scores in FTSST (all P ＜ 0.05);age,dialysis method,diabetes and WHR were independently correlated with scores in 3mTUG (all P ＜ 0.05).Conclusions The exercise performances of patients on maintenance dialysis are impaired.Age and diabetes are independent factor associated with the exercise performances of patients on maintenance dialysis.AMA is independently associated with upper limb movement,and dialysis method,BMI and WHR are independent factors associated with lower limb movement in dialysis patients.

Objective To analyse the morphological and phenotypic characteristics of dendritic cells obtained by induction of peripheral blood monocytes of gastric cancer patients and provide an experimental basis of (dendritic) cells for the biological treatment of gastric cancer. Methods Peripheral blood monocytes of gastric cancer patients were co-cultured with GM-CSF, IL-4 and TNF-? to obtain the dendritic cells. The (morphological) characteristics of the dendritic cells were observed by light and electron microscopes and the (phenotypes) were studied by flow cytometry. Results Mature dendritic cells were oblained seven days after (induction) culture. These cells showed numerous typical dendritic protuberances on their surface.The (phenotypes) of the cells increased gradually with their maturation and reached stable levels seven days later. Conclusions Mature dendritic cells could be obtained from peripheral blood monocytes of gastric cancer (patients) after induction culture with GM-CSF,IL-4 and TNF-?. The obtained cells had typical morphological and phenotypic characteristics. These cells could be used for further research in the biological treatment of (gastric) cancer.

Objective To provide experimental basis for the study of SGC7901 gastric cancer cell-dendritic cell fusion vaccine. Methods The fusion of SGC7901 gastric cancer cells and dendritic cells was induced by PEG. We obtained pure fusion cells by selective culture with HAT/HT culture systems. The growth curve characters, clone abilities, abilities to irritate T lymphocytes and the ability to grow were detected. Results The fusion cells had weaker proliferative abilities and clone abilities with increased abilities to irritate T lymphocytes. The CPM value of T lymphocytes were 15083?231、 9608?83、 4214?135、 3020?28 respectively when they were co-cultured with the fusion cells on different proportions. Conclusion SGC7901 gastric cancer cell-dendritic cell fusion cells keep strong abilities to irritate T lymphocytes while inable to grow into new planted tumors in immunodeficiency animals.

Objective:To investigate the endothelialization and intermediate and long-term patency rate of prostheses seeded by CD34+ stem cells.Methods:sixteen crossbred dogs were randomized into 2 groups.Prostheses covered with ePTFE or Dacron were implanted into the abdominal aorta artery(AAA) and inferior vena cava(IVC).Twelve dogs were implanted by prostheses seeded by CD34+ stem cells,4 dogs were implanted by autogenous blood only as control.The prostheses were explanted at thirty or sixty or one hundred days.Light and electron microscopy were applied to examine endothelialization of prostheses.CD34 factor stain was used to identify endothelial cells.Results:All venous prostheses implanted by autogenous blood were blocked.Others were patent.Confluent endothelial cells appeared on the neointima of seeded prostheses.There were no endothelial cells in the no-implanted prostheses.Conclusion:Endothelialization and higher patency rate might be achieved in the prostheses covered with ePTFE or Dacron that were implanted by CD34+ stem cells.
