1.Effects of miR-26a on proliferation, apoptosis, migration and invasion of uveal melanoma cell and its mechanism

Objective To analyze the effects of miR-26a on proliferation,apoptosis,migration and invasion of uveal melanoma cell.Methods The expressions level of miR-26a in the uveal melanoma cell lines SP6.5 and M23 and normal cell line ARPE-19 was detected by qRT-PCR.The SP6.5 cell line was divided into miR-26a mimics group (transfect with miR-26a mimics) and NC group (transfect with scramble by Lipofectamine 2000).The proliferation and apoptosis ability were measured by CCK-8 assay and flow cytometry.The cell wound scratch assay and transwell assay were used to detect the migration and invasion ability.The expression level of enhancer of zeste homolog 2 (EZH2) protein was measured by Western blot.Results The expressions level of miR-26a in uveal melanoma cell line SP6.5 was 0.250 ± 0.029,M23 was 0.350 ±0.017,which was significantly lower than 1.0 in normal cell line ARPE-19 (all P <0.001).The OD450 value at 72 hours,96 hours and 120 hours in miR-26a mimics group (0.69 ±--0.09,1.23 ± 0.15,2.12 ± 0.23) were significantly lower than those in NC group (1.39-0.11,2.35 ±0.25,3.53 ±0.27) (all P <0.05).The apoptosis rate of miR-26a mimics group (15.60% ± 2.30%) was significantly higher than that of NC group (5.00% ± 0.70%) (P < 0.01).The wound healing rate of miR-26a mimics group (23.7% ±2.1%) was significantly lower than that of NC group (68.9 ±5.1%) (P <0.01).While the invasive cell number (45.1 ± 3.9) was also significantly less than NC group (115.3 ± 8.9) (P < 0.01).The expression level of EZH2 protein in miR-26a mimics group (0.39 ±0.09) was significantly lower than that of NC group (1.0,P <0.01).Conclusion MiR-26a is low expressed in uveal melanoma cells.Over expression of miR-26a inhibit the proliferation,migration and invasion of uveal melanoma cell,and promote the apoptosis,which may be associated with down-regulated expression of EZH2.
2.Study on the correlation between contrast-enhanced ultrasonographic characteristics and microvessel density in breast cancer

Objective To investigate the relation of contrast-enhanced ultrasound ( CEUS ) characteristics and microvessel density ( MVD ) in the breast cancer .Methods From October 2010 to February 2012, 45 cases of patients with breast cancer were studied in Yantai Yuhuangding Hospital , Affiliated to Qingdao University Medical College .All lesions were examined by CEUS before surgery .The blood perfusion parameters such as rising time (RT),peak intensity(PI),time to peak(TTP),wash-in slope (WIS) and mean transit time ( MTT) were obtained by time-intensity curve ( TIC).Immunohistochemical staining for anti-factor CD34 was performed on surgery specimen and the MVD was evaluated .The CEUS characteristics and blood perfusion parameters between different MVD groups of breast cancer were compared.Results In 45 cases of breast cancer,mean MVD was(47.6 ±14.2)/high power field(HPD). Twenty-one cases(46.7%) were classified as high MVD group(MVD>48/HPD) and 24 cases(53.3%) were classified as lower MVD group ( MVD≤48/HPD) .Besides two cases without contrast agent perfusion in CEUS imaging, blood perfusion was observed in 43 cases (95.6%).Heterogeneous enhancement was observed in 25 cases(55.6%).Local blood perfusion defect was observed in 27 cases(60.0%).Irregular shape was observed in 37 cases(82.2%).Centripetal enhancement was observed in 25 cases(55.6%). Penetrating surrounding vessels was observed in 32 cases(71.1%).Poorly-defined margin was observed in 34 cases(75.6%).Compared with the surrounding normal breast tissue ,RT and TTP of center region of neoplasms was shorter[(9.3 ±3.3)s vs (11.1 ±3.7)s and (25.3 ±5.9)s vs (27.5 ±6.4)s],PI was higher[(12.1 ±4.6)dB vs (9.2 ±2.8)dB],WIS was higher(1.0 ±0.4 vs 0.8 ±0.3) and differences were significant(t =-3.001, -4.785,6.987 and 5.438,all P <0.05).Compared with center region of neoplasms,TTP of periphery region of neoplasms was shorter [(22.2 ±6.0)s vs (25.3 ±5.9)s],PI was higher[(15.4 ±5.1)dB vs (12.1 ±4.6)dB],WIS was larger(1.3 ±0.5 vs 1.0 ±0.4) and differences were significant(t=-2.839,3.194 and 3.151,all P<0.05).The detection rate of blood perfusion defect and heterogeneous enhancement was higher in the high-MVD group than in the low-MVD group(χ2 =4.0179 and 7.2024,both P <0.05).While the enhancement shape,margin and penetrating vessels showed no statistical difference between the two groups .Breast neoplasms in the high-MVD group had higher PI than those in the low-MVD group[(18.2 ±5.6)dB vs (12.9 ±3.1)dB,t=-3.738,P<0.05].While the RT, TTP and WIS showed no statistical difference between the two groups ( t=-0.798,-0.760 and -0.378, all P>0.05).Conclusion CEUS characteristics of breast lesions were associated with MVD ,which may reflect the microvessel distributional characteristics of neoplasm and may be one of bases used to evaluate neoplasm angiogenesis .
3.Long-term follow up of four patients with dopa-responsive dystonia

Objective To investigate the clinical characteristics,treatment effect,long-term follow up results,guanosine triphosphate (GTP) cyrclohydrolase Ⅰ (GCH Ⅰ)gene and tyrosine hydroxylase(TH) gene mutations in patients with dopa-responsive dystonia (DRD).Methods The clinical features of 3 families with 4 affected members were analyzed and all of 4 patients were screened for mutations of the GCH Ⅰ gene and TH gene with DNA sequences.Results Four patients were females,average age at onset was (15.3 ± 5.6) years (range:from 9 to 20 years).The initial symptoms were a gait disorder,stiffness or tremor of the lower limbs in all patients presented with diurnal fluctuation.As the increase of disease duration,bilateral hand tremor was found in three patients,systemic torsion was found in one patient and torticollis was found in one patient.All patients' symptoms were in complete remission after administration of low dose of levodopa.Four patients were followed up for 0.5 to 10.0 years,and all were still responsive to the levodopa treatment and effective dosage was decreased as the increase of the disease duration.No longterm side effects of levodopa had occurred after long-term treatment.One patient was found to have c.607G >A(p,Gly203Arg) heterogenetic mutation in GCH I gene.Molecular analysis revealed a compound heterozygous mutation in the TH gene (p.Y447Ter and p.V468M) in one patient.No point mutations in both genes were found in other patients.Conclusions DRD patients have dramatic and sustained response to levodopa and no long-term side effects of levodopa after long-term treatment.The detection of GCH Ⅰ and TH gene mutations is helpful in early diagnosis but the negative results could not exclude the diagnosis of DRD.
4.Effects of culture conditions on biomass and active components of suspension cells of Panax quinquefolium.

OBJECTIVETo study the effects of inoculum, various media, pH value of medium and illumination conditions on the growth of Panax quinquefolium suspension cells and the synthesis of ginsenosides Re, Rb1 and polysaccharides.
METHODThe suspension cells were obtained through tissue culture by manipulation of inoculum, various media, pH value, and illumination conditions. The contents of ginsenosides Re and Rb1 were determined by HPLC, while the contents of polysaccharide were determined by ultraviolet spectrophotometry.
RESULTThe growth rate of suspension cells was greatly increased when inoculum amount was 25 g x L(-1). The effect of media MS, SH and B5 on suspension cells was observed. MS medium was favorable for cells growth, while B5 medium was favorable for the synthesis of ginsenosides and polysaccharides. The polysaccharide content in three media were higher than that of the cultivations. The pH value showed little influence on the cells growth, medium pH 6.0 enhanced the synthesis of Re and polysaccharides. Illumination could significantly enhance secondary metabolite biosynthesis of suspension cells and promoted slightly in polysaccharide synthesis.
CONCLUSIONThe inoculum, various media, pH value of medium and illumination conditions have significant influences on suspension cells growth of P. quinquefolium, secondary metabolite and polysaccharides synthesis.

5.Protective effects of local heat preconditioning on hepatic ischemia/reperfusion injury.

AIMTo study the protective mechanism of HSP70 induced by the heat stress pretreatment on the hepatic ischemia/ reperfusion (I/R) injury.
METHODSTo establish the models of the hepatic ischemia/ reperfusion injury using pringle's maneuver with or without heat stress pretreatment. The rats were randomly divided into pretreatment group (HP + I/R) and non-pretreatment group (I/R), in which the expression of HSP70, the MDA contents and SOD activity in liver, the activities of serum AST and ALT, and the pathological changes in liver were detected at 0, 4, 8, 12 and 24 h after I/R.
RESULTSAt any time point set after I/R, the SOD activity in the liver in HP + I/R group were higher than those in IR group, HSP70 expression maximum was attained at 12 h after heat pre-treatment. While in HP+ I/R group the levels of liver enzymes and the production of MDA significantly reduced and the pathological changes improved compared with those in I/R group.
CONCLUSIONHSP70 induced by heat pretreated protecting liver against I/R injury may be through increasing the SOD content and then reducing the insults of oxygen free radicals in the liver.

6.Analysis of 105 Incarcerated Inmate's Death.

OBJECTIVE:
To analyze the characteristics in the incarcerated inmate's death, investigate the main cause of death of the incarcerated inmate and provide some information for forensic investigation.
METHODS:
The cases from the forensic medical center of Shanxi Medical University from 2005 to 2013 were selected. The statistical analysis was performed by using the incarcerated inmate's gender, age, cause of death, manner of death, and disease as the markers.
RESULTS:
There were 100 men, 5 women in the 105 incarcerated inmates; the age range was from 16 to 65 years; Inmates were mostly died of natural diseases, mainly in the respiratory and cardiovascular diseases; the main unnatural death was suicide with a rate of 54.5%.
CONCLUSION
At present, most incarcerated inmate's death are due to natural diseases. The prison should improve incarcerated inmate's lives, work and health care conditions, and strengthen supervision of law enforcement.

7.Bacterial drug resistance situation and clinical features of urinary tract infection in children

Objective To analyze the basic clinical characteristics,auxiliary diagnostic indexes,bacterial infection spectrum and drug resistance of child urinary tract infection to provide the basis for the prevention,diagnosis and treatment of child UTI.Methods The clinical data in the inpatients with UTI or complicating UTI in our hospital from January 2012 to March 2014 were collected. The repeated strains were excluded.The differences in the pathogens between the patients with complicated UTI and the patients with non-complicated UTI were comprehensively analyzed.Results The onset peak of child UTI for the first time was 0 -0.5 years old.Escherichia coli ,Klebsiella and Pseudomonas aeruginosa are the major pathogens of child UTI,in which 110 strains were Escherichia coli,accounting for 52.9%.The enterobacteriaceae pathogens of UTI had higher sensitivity to carbapenems antibacterial drugs,with the resistance rate of less than 10%;the resistance rate of others detected antibacterial drugs was more than 20%.Con-clusion UTI in the children inpatients of this area has higher drug resistance rate,in the treatment of child UTI,the antibacterial drugs should be rationally used by combining the bacterial drug resistance situation in the local place and the disease severity in or-der to avoid the aggravation of bacterial drug resistance.
8.Conjugated effects of fluoride and aluminium on rat whole blood zinc, iron, calcium, magnesium and copper level

Objective To investigate the effect of excessive fluoride,aluminum on Zn,Fe,Ca,Mg,Cuin rat blood.Methods Forty eight SD rats were randomly divided into 4 groups matched with their weights:control group,high aluminum group,high fluorine group and high fluorine-aluminum group.Aluminum content in their drinking water was 0,90,0,90 mg/L respectively.Fluorine content of their feed was 5.2,5.2,106.0,106.0 mg/kg and aluminum Was 6.8,6.8,19.7,19.7 mg/kg respectively.90 days later,the level of blood Zn,Fe,Ca,Mg, Cu Was detected by the atomic absorption spectrometry.Results Compared among these groups,Zn,Fe,Mg and Cu content of the whole blood had significant difierences(F=46.25,14.74,6.10,2.93,P<0.05),while Ca content of the whole blood did not significantly change(F=2.81.P>0.05).Factorial analysis showed that excessive intake of aluminum could significantly decreased Zn,Fe,Mg content of the blood(F=42.66,5.41,7.04,P<0.05)and excessive intake of fluorine could significantly decreased Zn,Fe,Mg,Cu content of the blood(F=64.50,37.90,9.75,6.74, P<0.05).The coexistence offluorine and Muminum had interaction to the level of Zn(F=31.59,P<0.05)and did not obviously interact with other elements(F=0.91,1.63,1.51.0.00,P>0.05).Compared with the control group [(131.30 ±13.86)μmol/L,(10.24 ±1.02),(1.71 ±0.19)mmol/L,(20.43 ±4.42)μmol/L],Zn content in the high aluminum group[(90.84±9.98)μmol/L]decreased significantly(P<0.05),so did Zn,Fe,Mg content in tlle high fluorine group[(85.85 ±10.92)μmol/L,(8.49 ±0.68),(1.52 ±0.13)mmol/L],the difference being statistically significant(P<0.05)0Zn,Fe,Mg,Cu content in the high fluorine-aluminum group,being(82.82 ±11.00)μmol/L, (8.16±0.45),(1.46±0.09)mmoL/L,(15.69±2.38)μmol/L,respectively,all decreased signitlcarIdy(P<0.05). Compared with the high aluminum group[(9.43±1.09)mmol/L],Fe content of the high fluorine aluminum group[(8.16±0.45)mmol/L]decreased significantly(P<0.05).Conclusions Excessive fluoride can cause blood zn, Fe,Mg,Cu decline,so can excessive aluminum.Combination of excessive fluofine and aiuminum has 8ignificant synergic effect on the level of Zn but have rio influence on Ca.
9.Point-selection Rules Analysis in Acupuncture-moxibustion Treatment of Asthma Based on the Evaluation of Pulmonary Function

Objective To discuss and summarize the point-selection rules in acupuncture-moxibustion treatment of asthma,by arranging and analyzing the relevant clinical studies with pulmonary function as the evaluation index.Method A total of 264 eligible articles of clinical trials about acupuncture-moxibustion treatment of asthma published in the last 10 years were analyzed by using frequency statistical method.Result and Conclusion In acupuncturemoxibustion treatment of asthma,acupoints frequently used majorly included Feishu (BL 13),Dingchuan (EX-B 1),Danzhong (CV 17),Pishu (BL 20),and Shenshu (BL 23),mainly distributed in the Bladder Meridian,Conception Vessel,Governor Vessel,Stomach Meridian,and extra points;syndrome differentiation,symptomatic analysis,and empiric selection were predominant in the point-selection rules.Acupuncture-moxibustion at the above points can effectively improve pulmonary function indexes and attenuate asthma symptoms,thus worth promoting in clinic.
10.Effects of culture conditions on biomass and active components of adventitious roots culture in Panax ginseng.

OBJECTIVETo optimize the culture condition of adventitious roots of Panax ginseng.
METHODThe adventitious roots were obtained through tissue culture by manipulation of inoculum, various sucrose concentrations and salt strength. The contents of ginsenosides Re, Rb1 and Rg1 were determined by HPLC while the contents of polysaccharides were determined by ultraviolet spectrophotometry.
RESULTThe multiplication of adventitious roots reached the peak when the inoculum was 20 g x L(-1). The effects of sucrose concentration and salt strength on adventitious roots were observed. The contents of polysaccharides were higher when the medium contained more sucrose. 40 g x L(-1) sucrose was favorable for roots growth and biosynthesis of Re, while 30 g x L(-1) was favorable for the biosynthesis of Rb1 and Rg1. 3/4MS medium was benefit for the growth of adventitious roots and the biosynthesis of ginsenosides. The contents of polysaccharides were decreased with the increase of salt strength.
CONCLUSIONThe results showed that inoculum, various sucrose concentrations and salt strength have significant influences on adventitious roots growth, secondary metabolite and polysaccharide synthesis in P. ginseng.
