2.Influence of tranilast on expression of OPN and interstitial ED1+ macrophages infiltration in rats with chronic cyclosporine A nephrotoxicity
Chinese Journal of Practical Internal Medicine 2006;0(20):-
Objective To evaluate the effect of tranilast on chronic cyclosporin A(CsA)nephrotoxicity rats and the possible mechanisms of protective effect of it.Methods Rats were on low salt diet and CsA was administered by gastric gavage at a dose of 20 mg/(kg?d)for 28 days, and tranilast was gave to these rats in a dose of 400 mg/(kg?d).The renal function and renal morphology were observed at1,2 and 4 weeks after administration of tranilast. Results CsA induced the corruption of normal condition and weight lost were markedly delayed after administration of tranilast. On histopathology tranilast attenuated tubular cells vacuolations as well as interstitial damage including interstitial ED1+ macrophages infiltration and focal interstitial fibrosis.Meanwhile, tranilast down-regulated the protein of OPN in kidney. Conclusion Tranilast can attenuate the structural changes of kidney in chronic CsA nephrotoxicity rats.Decreased CsA-related ED1+ macrophages,OPN up-regulation expression in the kidney may be related to mechanisms of protective effect of tranilast in the treatment of CsA induced chronic nephrotoxicity.

Objective:To screen the potential characteristic gene spectrums and signal pathways of osteoarthritis based on gene chips. Methods:We analyzed 2 microarrays of human joint synovial tissue (GSE82107 and GSE55235) derived from the Gene Expression Omnibus (GEO) database and included for this study 20 osteoarthritis (OA) samples and 17 healthy control samples. The differentially expressed genes (DEGs) between OA and HC were screened by GEO2R tool. Analyses of Gene Ontology function and Kyoto Encyclopedia of Genes and Genomes pathway enrichment were performed using the Database for Annotation, Visualization and Integrated Discovery to identify the pathways and functional annotations of DEGs (https://david.ncifcrf.gov/). Protein-protein interaction of these DEGs was analyzed based on the Search Tool for the Retrieval of Interacting Genes database and visualized by Cytoscape software (http://www.string-db.org/).Results:191 up-regulated DEGs and 49 down-regulated DEGs were screened out from the 2 microarray databases. Enrichment of DEGs was mainly found in regulation of such biological functions as "inflammation" , "bone cell differentiation" and "positive apoptotic cell regulation" , HTLV-I infection, silk crack on the original amp-activated protein kinase (MAPK) signaling pathway, swine flu, tumor necrosis factor (TNF) signaling pathway, the nf-kappa B signaling pathway, PI3 kinase/Akt pathway, toll-like receptor pathway, legionella, salmonella and other 14 signaling pathways. In 2 modes of MNC and Degree, the top 10 core genes were screened, of which interleukin-6 (IL6), JUN, chemokine 8 (CXCL8), early reaction growth factor (EGR1) and cyclin (CCND1) were identified as valuable biomarkers of OA.Conclusions:Based on GEO chips, 10 characteristic gene profiles such as IL6, JUN, CXCL8, EGR1, CCND and 14 signal pathways such as tumor necrosis factor (TNF) signal pathway, NF-κB signal pathway, PI3 kinase/Akt pathway and Toll-like receptor pathway were screened, which may provide new clues for understanding of the pathogenesis of osteoarthritis.

Objective To obtain an accurate and effective method for thalamus segmentation based on resting-state functional magnetic resonance imaging (fMRI).Methods Based on the fact that resting-state fMRI technique examined spatial synchronization of spontaneous fluctuations in blood oxygen level-dependent (BOLD) signals indirectly reflect the neuronal and synaptic activity, the in-thalamus BOLD signal correlations were calculated, and then the k-means clustering algorithm was applied to obtain functional connectivity-based thalamus segmentation. Results The thalamus was divided into seven regions.Voxels within the same region were highly correlated with each other. The segmentation result was similar to that divided by functional connectivity between thalamus and the cerebral cortex. Conclusions Resting-state fMRI could provide not only the functional connectivity network between cortical and subcortical brain regions, but also the functional characteristics of thalamus.Segmentation algorithm using only internal information of thalamus shows lower computational complexity and higher processing speed than that based on the functional connectivity between thalamus and the cerebral cortex.
