1.Studies on blood compatibility and toxicology of modified femoral arterial and venous cannulae for children in thoracoscope-assisted cardiac surgery

Objective To evaluate clinical feasibility of using a modified bio-material of femoral arterial and bi-caval femoral venous cannulae for children by investigating the blood compatibility and toxicological characteristics of the bio-material of the cannula.Methods The evaluation tests including hemolysis test,skin irritative test were performed in rabbits,and systemic acute toxic test was performed in mice.First,the haemolysis rate of this modified cannula bio-material was calculated by determination of absorptance conducted in vitro;second,the extracts of this material were prepared according to the national evaluation standards of medical bio-materials,and then the extracts were injected subcutaneously in rabbits to observe the histological responses of skin 72h after injection;finally the prepared extracts were injected intravenously into mice to observe the systemic responses 72h after injection.Results Hemolysis test showed that the hemolysis rate of the novel cannula bio-material was 1.58%,which was much lower than the international standard(5%).The blood compatibility of this material was also satisfactory.Primary skin test in rabbits showed no erythema,necrosis or edema in skin.Furthermore,acute systemic toxic test in mice showed no death and no loss of body weight,and no obvious abnormal response was observed 72h after injection.Conclusion The novel bio-material for femoral arterial venous cannulae for children meets the requirements of medical material property,and it can be clinically used.
2.Effects of advanced glycosylation end products on interactions between cis-elements and trans-acting factors of hepatic insulin receptor gene

Objective To explore the effects of advanced glyeosylation end products (AGEs) on the binding of hepatic nuclear factors to the human insulin receptor (hlR) gene promoter. Methods The oligonueleotides with hlR gene promoter activity, 42 bp (spanning -1 441 to -1 400, US1) or 146 bp (spanning -638 to -493), were artificially synthesized, with point mutations at 5 key G residues in 42 bp US1 m5 oligonucleotides. US1 and rat hepatic nuclear extracts (HNE) were incubated with glucose 6-phosphate, prior to non-competition and competition gel retardation electrophoresis. Results Competition gel retardation electrophoresis showed that the binding capacity of 32p-labeled US1 probe to HNE could be signifficantly decreased with increased US1. US1-AGEs and US1m5 decreased the binding of probe to HNE as well, but only partly affected the electrophoretie bands [1,5,10 ng US1-AGEs: (41.08±2.86)%, (27.64±2.92)%, (15.35±1.81%) vs (52.05±1.79)%]; 5,10 ng US1m5: (5.20± 1.03)%, (1.81±0.21)% vs (52.05±1.79)%]. AGEs formed on HNE could increase the binding of HNE to probe, along with nonspecifie binding increasing. Conclusion The impact of AGEs on hlR gene expression seems to be related to the effects on cis-elements and trans-acting factors.

7.THE OBSERVATION OF IMMUNOMORPHOLOGICAL CHANGES IN THE DRAINING LYMPH NODES OF MICE AFTER IMPLANTATION OF TUMOR CELLS INTO FOOTPAD

The response of murine lymphoreticular tissue with Special reference to regional lymph node after implantation of Ehrlich ascites carcinoma cells was investigated histologically and ultrastructurally. The tumor induced a vigorous histological response in both regional and nonregional lymph nodes that was proliferation of immunoblasts with enlargement of paracortical area in early stage and enlargement, fusion of germinal centers with an increased number of plasma cells in the medullary cords during the late stage of tumor growth.The results suggested that the response of lymph nodes might be mainly related to cell-mediated immune mechanism in the early stage and humoral immunity during the late stage of tumor growth. Regional lymph node gave reaction earlier and more vigorously in participating cell-mediated immunity, nonregional lymph node might be more significantly involved in humoral immunity. It was observed ultrastructurally that sinus histiocyte engulfed tumor cell in which the cooperation with lymphocyte might be necessary. The experimental results indicated that the increased number of immunoblast in paracortex may be considered as an indicator for antitumor cellmediated immunity with an important prognostic significance for host bearing tumor.
8.Efficacy of hyperosmotic glucose in treatment of corneal edema after cataract surgery

AlM: To explore clinical efficacy of hyperosmotic glucose after cataract surgery.METHODS: Eighty eyes ( 80 cases ) with ll level and above grades corneal edema after small incision cataract operation were enrolled. They were randomly divided into treatment group and control group. Control group used TobraDex eye drops and other conventional treatment and the treatment group received hyperosmotic glucose(500g/L) with the bases of the control group. The regression of corneal edema was observed by slit lamp microscope. SPSS 17. 0 statistical software was used for statistical analysis.RESULTS: The level of corneal edema at the first day after surgery, age, sexuality and nuclear hardness in the treated group were compared with those in the control group, the differences were without significances ( P>0. 05). Disappearance time of corneal edema was 4. 95 ± 3. 62d of the treatment group, and was 6. 80±3. 33d of the control group, the treatment group was better than control group (t=8. 55, P=0. 00).CONCLUSlON: Hyperosmotic glucose has exact and obvious efficacy for early corneal edema after cataract surgery.
9.Practical digitalized patient records control

Objective The objective of digitalized patient records control is to meet some new demands set on patient records control under the new circumstances. Methods A computer system for digitalized patient records control and supporting processing, management and applications setups were established. Results The system put the contents of the patient records into various categories with difierent standards and designed relatively strict means of authorization. It rendered patient records easier to use and their control more strict, making it possible to make general use of the records without access to the original documents. As a result, data loss was avoided, authority over the data was put under control, data sharing was provided, and full and convenient use of data was realized. Conclusion Digitalized patient records control, a form of shaping computer-based patient records before the latter fully comes into being, enhances the efficiency of patient records application and reduces the pressure of paper records storage. It is a practical and effective way of controlling patient records.
10.Structural feature and biological function of PPP2R5C gene.

PPP2R5C is one of the members of regulatory subunits of protein phosphatase 2A (PP2A), which plays a critical role in cell proliferation, differentiation and transformation, based on its induction of dephosphorylation of P53 at various residues. Recently, it was characterized that the alteration of expression pattern of PPP2R5C is associated with cell malignant transformation, thus PPP2R5C was thought as a marker for progressive disease in B-CLL. In this article the gene structure and biological function of PPP2R5C as well as relation of PPP2R5C with genesis and development of cancer were discussed.
