Objective:To develop the representative fingerprint for the quality control of placenta polypeptide injection.Methods:The chromatographic separation was performed using a Phenomenex Gemini C18 column (250 mm × 4.6 mm,5 μm) maintained at 30 ℃.0.1％ aqueous trifluoroacetic acid (Solvent A) and acetonitrile contained 0.1％ TFA (Solvent B) were used as mobile phase with a gradient elution.Detection wavelength was 280 nm with the sample injection volume of 50 μL; the flow rate was 1.0 mL/min.The fingerprints of different samples were investigated by similarity analysis.Results:Nine peaks were identified as the characteristic common peaks.The similarities of the fingerprints of the 10 batches of samples were above 0.992.Conclusion:This method showed high precision and good repeatability,and provided the basis for the improvement of the quality control of placenta polypeptide iniection.

Objective To investigate the association between HbA1c status,arterial stiffness and endothelial function in subjects with type 2 diabetes mellitus(DM).Methods The study group consisted of 338 DM patients,including 74 subjects with normal HbA1c(≤ 6.0％)(Group A),102 subjects with mildly increased HbA1c(HbA1C increased by less than 6.5％)(Group B)and 162 patients with high HbA1c level (HbA1c increased by more than 6.5％)(Group C).All patients were evaluated for glucose,HbA1c,TC and TG.Pulse wave velocity(PWV)and flow-mediated dialation(FMD)were performed simultaneously.Results Compared with Group A,Group B and C had higher baPWV([1734 ± 343]cm/s vs.[1537 ± 313]cm/s and [1853 ± 364]cm/s vs.[1537 ± 313]cm/s).Patients in Group C had a higher baPWV than those in Group B ([1853 ± 364]cm/s vs.[1734 ± 343]cm/s,P =0.006).The patients in group A had a lower level of FMD than that of group B and C([4.20±3.13]％ vs[5.29±3.92]％,[4.09±2.79]％ vs.[5.29±3.92]％,P＜0.01).Leaner association analysis showed a positive correlation between baPWV and HbAlc(r =0.53,P ＜0.01).baPWV was negatively correlated with FMD(r =-0.25,P ＜0.01).Logistic regression analysis showed that HbA1c(OR32.19％,CI 11.26 ～53.12,P＜0.001),age(OR 14.21,95％ CI 11.43 ～ 17.00,P＜0.001),systolic blood pressure(OR 7.36,95％ CI 6.12-8.59)and TC(OR:0.31,95％ CI:9.97-70.64)were independent predictors for the increased arterial stiffness(baPWV).Conclusion Arterial stiffness parameters varied significantly across subgroups of patients with different levels of HbA1c and the arterial stiffness was positively associated with deranged glucose homeostasis.

Objective To investigate and analyze the effect and insulin dosage of continuous subcutaneous insulin infusion(CSII) in type 2 diabetes.Methods 106 hospitalized patients with type 2 diabetes,who had poorly controlled blood glucose,were treated by insulin pump for 8 days,without oral hypoglycemic agents.The treatment days when blood glucose was well controlled and the insulin dosage at that time were observed.The related foctors for the therapeutic effects were analyzed.Results When blood glucose were well controlled,the mean time was 5.4±1.8 days,the(basal) insulin was 0.23±0.07(u/kg,day),the bolus before breakfest was 0.12±0.05(u/kg),the bolus before dinner was 0.14±0.06(u/kg),the bolus before supper was 0.14±0.06(u/kg).The dosage of insulin was decreased after blood glucose were well controlled.The frequency of hypoglycemia was 0.04±0.21(times/per patient),the frequency of biochemical hypoglycemia was 0.07±0.25(times/per patient) and reaction of hypoglycemia was 0.96±1.18(times/per patient).Conclusions The hyperglycemia of type 2 diabetes could be corrected by intensive insulin pump within 5-6 days.Multiple regression showed that fasting blood glucose and age were the factors that affect the time for blood glucose to be targeted.Duration of diabetes,fasting and postprandial blood glucose levels before treatment affect the required insulin dosage.

Object To search for bioactive secondary metabolites from marine organisms, the marine sponge Cinachyrella australiensis was collected from Hainan Island, Southern China Sea to develop the efficient drug for human beings. Methods Silic gel flash chromatography in association with reverse phase semipreparative HPLC was performed for the isolation and purification of the ethanol extracts, and extensive spectroscopy including 2D NMR spectra as well as the comparison of the spectral data with those reported in literatures was applied for the structure elucidation. Results Seven nucleotidoids were isolated and identified. They are named as 2-methyl-6-amino-9-(2-deoxy-?-D-ribofuranosyl)-purine (Ⅰ), 2′-deoxyadenosne (Ⅱ), 6-amino-9-?-D-ribofuranosyl-9H-punne (Ⅲ), uracill (Ⅳ), thymine (Ⅴ), thymidine (Ⅵ), 1-(2-deoxy-?-D-ribofuranosyl) uracil (Ⅶ). Conclusion The sponge C. australiensis is systemically separated, and then two kinds of nucleotidoids compound are extracted.

Objective:To study the expression of 11 cancer-testis (CT) genes and autologous humoral immune response to NY-ESO-1 in gastric carcinoma in China to provide some data for developing a vaccine against this cancer. Methods: Pathology tests were conducted in 101 gastric carcinoma samples,including tumor tissue and paired adjacent normal non-tumor tissue. Reverse transcription-polymerase chain reaction (RT-PCR) was used to test for 11 CT antigen gene expression and ELISA for autologous humoral immune response to NY-ESO-1.The antigen of NY-ESO-1 was also detected in tumor tissues by mAb E978 in IHC.Results: About 74.3% of the samples expressed at least one CT antigen,and the most frequently expressed one was MAGE-3 (41.6%). Twelve out of the 101 samples (11.9%) were found to be NY-ESO-1 mRNA positive. Simultaneously, autologous humoral immune response to NY-ESO-1 was detected in 6 of 12 NY-ESO-1 mRNA positive patients. Conclusion: Multiple CT genes were expressed in gastric cancer samples. MAGE-3,SSX-1,SSX-4, NY-ESO-1/LAGE-1, and MAGE-1 can be chosen as candidate antigens for cancer vaccines due to their higher expression rate. The potential of a multivalent CT antigen vaccine, including NY-ESO-1 and MAGE-3 along with other CT antigens, should be evaluated in patients with gastric cancer.

Objective To investigate the expression and pathological diagnostic utility of ?-methylacyl-coenzyme A racemase(P504S)in prostate cancer(PC). Methods The specimens of 42 cases of PC,18 cases of prostatic intraepithelial neoplasia(PIN)and 25 cases of benign prostatic hyperplasia(BPH)tissues were immunohistochemical stained with P504S,34?E 12 and vascular endothelial growth factor(VEGF)antibody,respectively. Their staining extent and intensity were analyzed and compared. Results Of the 42 PC cases,33(79%)showed positive P504S staining with moderate to strong staining intensity, which was significantly stronger than that of PIN and BPH cases,while adjacent normal glands were all negative. Weakly positive P504S staining was found in 6(33%)of 18 PIN cases and 2(8%)of 25 BPH cases,while it was negative in others.The specimens of BPH cases had a complete basal cell layer and were positive for 34?E 12 ;while 40 cases of PC had no basal cell layer and only 2 cases had discontinuous basal cell layers with weak staining.The basal cell layers of PIN cases were positive for 34?E 12 and the staining was a little weaker than that of BPH cases.All the specimens of BPH,PIN and PC cases showed positive staining for VEGF,the extent and staining intensity had no significant difference among these groups. Conclusions P504S has high sensitivity and good specificity in the diagnosis of PC,and is helpful for differentiating the suspected cases when combined with 34?E 12.

Objective To investigate the expression of programmed cell death-1(PD-1)ligands on HepG2 cell and 2.2.15 cell.Methods Culture HepG2 cell and 2.2.15 cell in vitro and stain the cells with fluorescence-labeled monoclonal antibodies specifically against PD-L1 and PD-L2,respectively.Isolate total RNA of HepG2 cell and 2.2.15 cell and detect the mRNA expressions of PD-L1 and PD-L2 by RT-PCR.Results The expression of PD-L1 on 2.2.15 cell was upregulated on the levels of both proteins and mRNA.The expression of PD-L2 in 2.2.15 cell was not detected.And neither the PD-L1 nor PD-L2 was detected in HepG2 cell.Conclusion HBV infection can upregulate the expression of PD-L1 on hepatocyte,which might contribute to the immune evasion of HBV.

Objective: Fetal monitoring is extremely important in perinatal care. Analysis of cardiotocogragh(CTG) can suppose if the fetus is lack of oxygen, distressed, acidosis, etc. in order to take measures timely. Methods: We have designed Meticare analysis software package to analyze 867 pieces of CTG data, and researched the relationship between short term variation (STV) and basal heart rate(BHR) by statistics, and short time monitoring analysis based on STV. Results: The amplitude of fetal heart rate curve variation(long term variation) tended to decrease when BHR increasing, with the negative correlation and regression equation STV = 9.866-0.02×BHR. Positive correlation exists between 5 and 10 mimute-STV, with the regression equation STV_(10min)=2.861-0.596×STV_(5min).Conclusion: STV calculated by 5 minutes CTG data can forecast the one by 10 minutes data as a reference demanded by Dawes & Redman criterion. If fetal doppler contains STV analysis function, out-patient fetal heart auscultation will be more significant. After 30 weeks pregnancy, with the STV decreasing, fetal distress, asphyxia, low birth weight, and even mortality increasing. BHR increasing should cause attention that STV might decrease, when monitoring with no analysis functional devices or fetal dopplars.

Allergic diseases such as allergic asthma, allergic der-matitis, allergic rhinitis, are polygenic diseases, involving the interaction between the environment, genes and immunity. In the past few decades, the incidence rate of allergic diseases in-creased predominantly and influenced the quality of people's lives seriously, so looking for new targets for the prevention and treat-ment of allergic diseases and drugs with less adverse reaction be-comes a hot topic for researchers. MicroRNAs(miRNAs)are a class of endogenous non-coding small RNAs that mediate nega-tively posttranscriptional regulation of gene expression by targe-ting specific mRNA sequences to inhibit the translation of mR-NAs. They are widely involved in the biological processes of cell differentiation, immune response and tumor development. The study shows that miRNAs can control the occurrence and devel-opment of allergic diseases. Studying the regulatory role of miR-NAs in allergic diseases has important implications for exploring the immunopathological mechanisms and discovering new thera-peutic targets of drugs.
