1.Comparison of three types of eye care for preventing exposure keratopathy in the Intensive Care Unit

AIM:To compare the efficacy of three types of eye care (artificial tear, moist chamber and polyethylene covers) for preventing exposure keratopathy in intensive care patients.METHODS: Intensive Care Unit (ICU) patients were randomly divided to three treatment groups, including artificial tear group(29 cases), moist chamber group (27 cases) and polyethylene covers group (28 cases). Patients in artificial tear group received two drops of carboxymethylcellulose dropped to each eye every 2 hours. The moist chamber and polyethylene covers groups were changed every 12 hours or as needed if they became unclean or torn. Corneal fluorescein stain was performed daily.RESULTS:No one in the polyethylene covers group and one case(4%) in the moist chamber group had exposure keratopathy compared to 8 cases (28%) in the artificial tear group. There were statistical significance differences between both artificial tear group and moist chamber group (P=0.02), artificial tear group and polyethylene covers group (P=0.003). Everyday time of eye care in the artificial tear group, the moist chamber group and the polyethylene covers group was respectively 26.69±2.39, 35.33±2.63 and 7.48±0.87 minutes. Everyday time of eye care in the polyethylene covers group was statistically more time-saving than that in the artificial tear group (P<0.01) and the moist chamber group (P<0.01). CONCLUSION: Polyethylene covers are more effective and time-saving in reducing the incidence of corneal damage in intensive care patients.
2.The effect of cinobufacini injection on DNA topoisomerase Ⅰ of human hepatocellular carcinoma HepG-2 cells

Background and purpose:The cinobufacini injection is a traditional antitumor drug.However,its mechanism iS still unclear.The purpose of this study was to observe the effect of cinobufacini injections in DNA TOPO Ⅰ of human hepatocellular carcinoma HcpG-2 cells.Methods:The cells that were proliferated were assessed by MTT assay.Cell cycles were shown through FCM.TOPO Ⅰ mRNA expression was analyzed through RT-PCR.The activity of TOPO Ⅰ was measured by TOPO Ⅰ mediated super coiled PHR322 relaxation.Supercoiled PBR322 was also used to determine the direct DNA breakages.Results:Cinobufacini injections significantly inhibited HepG-2 cells proliferation in ways that were dependent on dosages and time.Induced tumor cells arrest at the S-phase.TOPO ⅠmRNA expression decreased in a manner that was dependent on dosages which inhibited the TOPO Ⅰ mediated DNA relaxations.However,the cinobufacini injections could not directly induce DNA breakage at any concentration.Conclusion:Cinobufacini injections can inhibit human hepatocellular carcinoma HepG-2 cells proliferation.The regulation of topoisomerase Ⅰ activity and mRNA expression may be one of the mechanisms that causes the cinobufacini injection to contribute against tumor.
3.Septal cartilage augmentation for hump nose plasty

Objective To introduce a new method of septal cartilage augmentation for hump nose plasty. Methods The section of septal cartilage was collected and divided into two or three parts. The grafts were sutured by means of a mattress suture, and placed over the dorsum of superior and inferior hump. The dorsum became flat and straight immediately afterward. Results After a short term and long term follow up, 30 patients who underwent this operation were satisfied the operative results. Conclusion Septal cartilage augmentation for hump nose plasty is an efficient new method.
4.Evaluation on dosage-based efficacy-toxicity correlation of Tripterygium wilfordii against immune inflammation in mice.

OBJECTIVETo study the anti-immune inflammation efficacy and toxicity of Tripterygium wilfordii decoction, in order to provide experimental basis for studies on its "efficacy-toxicity" correlation.
METHODThe delayed hypersensitivity model was established by dinitrofluorobenzene in mice. Different doses of T. wilfordii decoction was administered for 5 consecutive days. The ear swelling inhibition ratio and the toxic action were observed. After the final administration, the biochemical indexes of PGE2, TNF-α, IL-2, ALT, AST, PA, TBA, TBIL in serum were detected, and the visceral indexes of heart, liver, spleen and kidney were measured.
RESULTThe DNFB-induced ear swelling could be notably inhibited by multiple oral administration of T. wilfordii decoction, with the ED50 and its 95% confidence limit of 0.34 (0.21-0.42) g x kg(-1). The contents of PGE2, TNF-α, IL-2 in serum decreased in a dose-dependent manner. The activities of serum AST, ALT, TBA, TBIL and the PA content reduced.
CONCLUSIONT. wilfordii decoction shows a significant anti-immune inflammation efficacy within the dosage range between 0.59 and 2.34 g x kg(-1) in a dose-dependent manner. With a certain hepatotoxicity, high dose (2.34-4.68 g x kg(-1)) of T. wilfordii decoction can cause substantial liver injury, with a dose dependence in liver function index. Therefore, the efficacy and toxicity of T. wilfordii is dose dependent, which provides reference for preventing adverse drug reactions in clinic and developing early-warning schemes and ensure the clinical medication safety of T. wilfordii.

5.The role of heat shock protein 70 in regulating neuroinflammation.

Neurodegenerative disease is characterized by progressive loss of neurons in specific brain regions that results in neuronal dysfunction of the central nervous system. Although the pathological mechanism is not fully established, the activation of glial cells mediated neuroinflammation appears to be involved. Heat shock protein 70 (HSP70) is originally described as intracellular chaperone, which plays an important role in protein quality control in cells. However, recent study showed that up-regulation of HSP70 had anti-inflammatory effects in the brain. HSP70 protected neurons from damage and improved neurological function by decreasing inflammatory response as indicated by inactivation of glial cells and inhibition of pro-inflammatory cytokine release. So it is of great significance to find new compounds targeting at HSP70 as neuroprotective agents to delay the progress of neurodegenerative disease. This review will focus on the role of HSP70 in neuroinflammation and the recent advances in using HSP70 as a target for the treatment of neurodegenerative disease.

6.Therapeutic effect of a natural squamosamide derivative FLZ on Parkinson's disease model mice induced by LPS plus MPTP.

The aim of this study is to investigate the protective effect of N-[2-(4-hydroxyphenyl)ethyl]-2-(2, 5-dimethoxyphenyl)-3-(3-methoxy-4-hydroxyphenyl)acrylamide (FLZ), a novel synthetic squamosamide cyclic derivative, against Parkinson's disease (PD) model mice induced by the inflammatory bacterial endotoxin, lipopolysaccharides (LPS) and the neurotoxin 1-methy-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP). C57/BL mice were ip injected LPS (5 mg x kg(-1)) once. One week following the LPS injection, mice received a subcutaneous injection of MPTP (25 mg x kg(-1)) once daily for 2 days. Eight weeks later, FLZ (25, 50 and 75 mg x kg(-1)) was orally administered to mice once daily for 60 days. The motor ability of the mice was evaluated by rod climbing test and footprint test. The dopamine (DA) levels in mouse striatum were determined by high performance liquid chromatography system. The tyrosine hydroxylase (TH)-positive cells were showed by immunohistochemical analysis. FLZ treatment significantly improved motor dysfunction of mice challenged by LPS plus MPTP. The increase of TH-positive cell numbers and elevation of DA levels may be contributed to the beneficial effects of FLZ on motor behavior. This study showed FLZ has significant therapeutic effect on LPS plus MPTP induced chronic PD model, which indicates its potential as a new candidate drug to treat PD.
7.Effect of manual massage on relapse rate and quality of life of grand mal epilepsy patients

Objective The aim of the study is to study the effect of manual massage on relapse rate and quality of life of grand mal epilepsy patients.Methods Using prospective research method,clinical data of patients with epileptic seizures and without attack up to 3 years and undergoing withdrawal of drugs for one year were collected.The grand mal epilepsy patients were randomly divided into the experimental group and the control group (76 cases in each group).The experimental group was given manual massage.The control group was not given manual massage.Both groups were given follow-up for 1 year.The relapse rate and quality of life were compared between two groups.Results The relapse rate of the ex perimental group was 17%,and that of the control group was 33%,the difference was significant.The score of life quality in the experimental group was significantly better than that of the control group.Conclusions Manual massage can reduce relapse rate and improve quality of life for patients with epileptic seizures after withdrawal of drugs,which is worthy of wide application.
8.Effect of topical tacrolimus on xenogeneic hair follicle transplantation

Objective To study the effect of topical tacrolimus (FK506) on the survival of the xenogeneic transplanted hair follicles from human scalp to Wistar rats. Methods In our study, Wistar rats were used as recipients and human as donor. The black hair follicles of human scalp were harvested, and then the xenogeneic grafts were transplanted to white Wistar rats on the back. 20 couples of rats were divided in 2 groups: topical tacrolimus group (group A), and blank control group (group B). After operaton, we compared the survival time of hair follicles and their histologic outcomes in order to verify the practicability of xenogeneic transplantion of hair follicles, and the topical application of tacrolimus results.Results The mean survival time of group A was longer [(49. 9 ±7. 1) days] as compared to group B [(13. 1±1. 2) days]. The longest survival time was 65 days in group A and 14 days in group B, respectively. By comparison of the results we found that topical tacrolimus prolonged the survival time of the xenotransplanted hair follicles significantly and that tropical medication could not avoid rejection. Conclusions The immune privilege function dependent on the hair follicle anagen and axillary topical tacrolimus, can prolong the survival time of the xenogeneic transplanted hair follicles in rats significantly.
9.Follow-up and retreatment strategies in the anti-vascular endothelial growth factor-A therapy for neovascularization of age-related macular degeneration

Wet age-related macular degeneration (AMD) is one of major causes of blindness in elder people.Intraocular injection of anti-vascular endothelial growth factor (VEGF) -A regimen has made big breakthrough for the treatment on choroidal neovascularization of wet AM D,while long-term follow-up and necessary retreatments are the key issues to remain obtained visual acuity.Multiple strategies of wet AMD have been used in following-up and retreating based on the visual acuity,optical coherence tomography (OCT),ophthalmoscope and fluorescein fundus angiography (FFA) in abroad.However,there also are major differences in the patient' s composition,treatment habits and distribution of medical sources in China from Western.So we suggest to standardize the follow-up and retreatment strategies about intravitreal injection of VEGF-A for wet AMD as to achieve a better effectiveness.OCT-guided individual follow-up and retreatment strategies should be very helpful for maintaining a long-term efficacy,minimizing the treatment time and reducing medical cost.
