Objective To investigate the minimal inhibitory concentration (MIC) of Pseudomonas aeruginosa to antibacterial agents which were induced by disinfectants. Methods The susceptibility test of 68 strains from clinic was determined by VITEK-2 system. The MIC of povidone iodine, glutaraldehyde and chlorhexidine was determined by agar dilution method.68 strains were induced by different concentrations of three kinds of disinfectants for eight times. The concentration of three kinds of disinfectants was lower than their MIC.The MIC before and after inducement was determined by micro dilution. In the mean-while,strains of their MIC raised to antibiotics were inoculated on plate agar containing the PAβN which was the efflux pump inhibitor,in order to determine the phenotype of efflux pump. Results Six strains,MIC in 68 strains significantly changed after induction of disinfectants. The MIC of these six strains was higher than pre-induced to some antibacterial agents such as FQNs,cephems and carbapenem. The induction of povidone iodine and gilutaraldehyde were more obvious, and five strains, phenotype of efflux pump were positive after being induced by these two disinfectants. Conclusions The antimicrobial susceptibility of some Pseu-domonas aeruginosa will change after being induced by some disinfectants and the efflux pump will play an important role in this process. So the reasonable application of disinfectants were indispensable,in order to decrease the resistance and the opportunity of nosocomial infection.

By using the pharmacophore model of mineralocorticoid receptor antagonists as a starting point, the experiment stud- ies the method of traditional Chinese medicine formula design for anti-hypertensive. Pharmacophore models were generated by 3D-QSAR pharmacophore (Hypogen) program of the DS3.5, based on the training set composed of 33 mineralocorticoid receptor antagonists. The best pharmacophore model consisted of two Hydrogen-bond acceptors, three Hydrophobic and four excluded volumes. Its correlation coefficient of training set and test set, N, and CAI value were 0.9534, 0.6748, 2.878, and 1.119. According to the database screening, 1700 active compounds from 86 source plant were obtained. Because of lacking of available anti-hypertensive medi cation strategy in traditional theory, this article takes advantage of patent retrieval in world traditional medicine patent database, in order to design drug formula. Finally, two formulae was obtained for antihypertensive.

Objective To analyze the expression of glial fibrillary acidic protein(GFAP) in hippocampal of rats with hypoxic-ischemic brain damage(HIBD) after injection of bone marrow stromal cells(BMSCs) supernatant into the lateral ventricle of the neonate rats, assess the ability of learning and memory, and explore the mechanism. Methods 7-day-old SD rats were randomly divided into sham operation group, model group, cells and supernatant group, with 20 rats in each group. The HIBD model was established via the ligation of left carotid arteries followed by 2-hour hypoxia. One week later, rats in control group were injected with 0.01 mol/L PBS 2 μl via the left ventricle.Meanwhile, the cell and the supernatant groups were injected with BMSCs and supernatant harvested from BMSC culture, respectively, via the same route. In the sham operation group, the left carotid arteries were separated but not ligated, and no hypoxia treatment was imposed on this group. They also received 0.01 mol/L PBS injection 2 μl per weak post surgery.8 weeks later,Morris water maze test was performed to assess the learning and memory, and the expression of GFAP in the dentate gyrus of rats was measured by immunohistochemistry. Results Morris water maze showed that the searching time of the probe trail(T1:(15.40±2.80) s) and reversal probe trail (T2:(16.45± 1.16) s) of the model group was shorter than that of the sham operation group (T1:(19.96±2.57) s,T2:(2:25.32±2.54)s,P<0.05),while the searching time of the cell group(T1:(17.54± 1.80)s,T2:(18.99± 1.47) s) and supernatant group (T1:(17.40±2.37) s,T2:(17.96± 1.09) s) was prolonged compared with that of the model group (P<0.05).No significant difference between the cell group and the supernatant group (P>0.05).The integral optical density (IOD) value of GFAP positive cells was higher in the model group than that in the sham operation group(15.26±1.49 vs 12.82±2.56,P<0.05),while the IOD of cell group(18.13±1.97) and that of the supernatant group(17.38± 1.64) were higher than that of the model group (P<0.05).However,there was no significant difference between the cell group and the supernatant group (9>0.05).Conclusion BMSC supernatant injected through lateral ventricle can improve the HIBD rats' abilities of learning and memory. The mechanism might be that MSCs secrete some cytokines to promote central nervous system repair.

Objective To compare real-time fluorescence quantitative PCR with general PCR in detecting bovine and porcine derived materials in hydrolysate samples. Methods DNA were extracted from hydrolysate samples which prepared by different steps by real-time fluorescence quantitative PCR and general PCR.Results DNA of bovine and porcine could be detected by real-time fluorescence quantitative PCR and general PCR in samples prepared in the processes before enzymolysis solution, but not detected in samples from supermatant to the fourth ultrafiltrate. Conclusion Both real-time fluorescence quantitative PCR and general PCR can be applied to detect the fragments in hydrolysate samples. And real-time fluorescence quantitative PCR has the advantage such as rapid, convenient, non-environment-polluted, good repeatability, which improves the quality and efficiency.

Objective To primarily investigate the clinical value of analgesia/nociception index (ANI) in evaluating the analgesic effect during lobectomy performed via video-assisted thoracoscope. Methods Forty ASA physical status Ⅰ or Ⅱ patients, aged 25-64 yr,weighing 45-80 kg,undergoing elective lobectomy performed via video-assisted thoracoscope, were enrolled in this study. After induction of anesthesia with propofol, sufentanil and cisatracurium, patients received double lumen endotracheal intubation.Anesthesia was maintained with targetcontrolled infusion of propofol, and iv infusion of remifentanil and cisatracurium. The concentration of propofol was adjusted to maintain the bispectral index (BIS) value in the range of 40-60.ANI,HR,systolic blood pressure (SBP),diastolic blood pressure (DBP) and BIS value were recorded within 5 min before and after the predefined time points including posture change between lateral and supine position, ventilatory pattern change between onelung and double-lung ventilation, skin incision and trocars insertion, lymph node dissection and pleural lavage. At skin incision and during trocars insertion, lymph node dissection and pleural lavage, the development of hemodynamic responses (increase in HR and SBP > 20% of baseline value) were recorded. Results The incidence of hemodynamic responses was 100% at skin incision and trocars insertion, and 84 % during No.4,7,10 groups of lymph node dissection and after pleural lavage and difference was found in ANI during these stimuli.ANI was significantly decreased within 5 min after skin incision, trocars insertion,No.4,7,10 groups of lymph node dissection and pleural lavage than that before the procedures (P < 0.05).The BIS value was maintained at 40-60,and no significant changes were found between before and after the procedures (P > 0.05).No significant changes were found in ANI,HR,SBP,and DBP between before and after the changes of posture and respiratory pattern (P > 0.05).Conclusion ANI can be used to evaluate the analgesic effect during lobectomy performed via video-assisted thoracoscope in patients and is unaffected by the changes of posture and ventilatory pattern.
