1.Study of probe substrate specificity for human cytochrome P450 isoenzymes

Analysis of metabolic activities of cytochrome P450 isoenzyme is a crucial index to study drug/toxicant metabolism, drug-drug interaction, polymorphism and et al. Due to this practice, it is important to use the proper probe substrate and to conduct the experiment under optimal conditions. The validation information in literatures on the most common and newest in vitro probe substrates have been reviewed.
2.One case of postoperative nursing care for patient undergoing domino liver transplantation with two living donors

Objective To probe into the methods of postoperative nursing care for the patient undergoing domino liver transplantation.Methods The key of postoperative care included:monitoring of vital signs,the condition of cruor and the state of drainage rigorously; keeping the balance of intake and output; taking the immunosuppressive drugs correctly; proper diet; prevention of complication;giving psychological nursing.Results The patients passed the perioperative period successfully and were all cured.Conclusions To aim at the characteristic of this operation,implement holistic nursing care,is an important link to assure the patients of domino liver transplantation to recover smoothly after the operation.
3.Effects of GLP-1 treatment on protection of B cells in Otsuka Long-Evans Tokushima fatty rats

Objective: To investigate the effect of GLP1 on the blood glucose in type 2 diabetic rats,and its protective effects on the islet B cells.Methods: Thirty rats were divided into three groups: spontaneous type 2 diabetes animal model OLETF rats,GLP-1 [from the twelfth week 56 ?g/(kg?d),sc] therapy group and LETO rats as control.In the 14th and 20th weeks,standard OGTT including fast and 2 h-plasma glucose were measured respectively.In the 14th weeks,3 rats from each group were killed randomly,and the rest of the rats were killed until the 20th week.Immunostaining with the marker of PCNA,TUNEL,and insulin assessed metabolic changes in the islet. 4.Establishment of precision-cut fibrotic liver slice technique

AIM: To establish the technique of precision-cut fibrotic liver slice (PCLS) and grope the optimal cultural conditions for researching the liver xenobiotic metabolism in vitro and the drug interaction. METHODS: Complex factors (higher fat diet, alcohol and CCl 4) were used to make the animal model of liver fibrosis. Fibrotic liver slices were prepared and cultivation system was established. Lactate dehydrogenase (LDH) leakage, glutathione S-transferase (GST) activity and 3[4,5-Dimethythiazole-2-yl]-2,5- diphenyltetrazolium bromide (MTT) reduction were chosen as indexes to assess the viability of the slice in different thickness, medium pH and cultural time. RESULTS: Rats were in earlier hepatic fibrosis after administration for 3 weeks. When the thickness of slices was 300 ?m and medium pH was 7.0, the LDH leakage, GST activity and MTT reduction could maintain on a steady level in 6 h. CONCLUSION: A 300 ?m of thickness, 7.0 of medium pH and 6 h of cultural time are the optimal slicing and culturing conditions for fibrotic liver slice.
5.Clinical research on the effects of propranolol on infantile hemangiomas

Objective To study the efficacy and adverse reactions of the treatment on different types of infantile hemangiomas (IH) with different doses of propranolol .Methods One hundred and fifty cases of IH without contraindications were randomly treated with different doses of propranolol (low dose group:1 mg?kg -1 ?d-1 ;high dose group:2 mg?kg -1 ?d-1 ) under ECG monitoring inferior half an hour to breastfeeding taking ,and stay in hospital for observation for 6 to 24 hours after the treatment . The patients returned to the hospital for review every month(ECG and blood RT ;lung function in necessary) .Results The curative effect is better on strawberry IH than that on flat IH ,also better on deeply IH than that on superficial IH .The adverse reactions oc‐curred relatively slight in the low dose group and there was no significantly statistical differences on the treatment effects between the both groups(P>0 .05) .Conclusion It is safe and effective to treat IH with propranolol .
6.Early enteral nutrition in preventing functional delayed gastric emptying after subtotal gastrectomy

Objective:To investigate the role of early enteral nutrition in preventing functional delayed gastric emptying(FDGE)after radical distal subtotal gastrectomy.Methods:A retrospective review was performed in clinical data of patients with gastric cancer undergoing radical distal subtotal gastrectomy in our hospital from March 2004 to March 2008.There were 71 patients receiving early enteral nutrition (group EEN,n=71) after operation and 107 patients receiving parenteral nutrition (group PN,n=107) at the same time.The outcomes of FDGE between of two groups were compared.Results:The incidence of FDGE in group EEN was 1.4%(1/71).And the incidence of FDGE in group PN was 10.3%(11/107).There was significant difference between the group EEN and the group PN (P
7.Isolation,Identification and Characterization of a Methanotrophic Strain

Strain QJ16,one of some methanotrophic bacteria isolated from soil samples utilizing the separate device designed by our laboratory;was studied in detail and identified as one of the Genus methylomonas through 16S rDNA sequencing and comparison. The culturing and reaction conditions for methane utilization of this strain were investigated. The results indicated that the optimal conditions for the growth of strain QJ16 were methane gas as carbon source and the coexistence of NH4Cl and KNO3 as nitrogen source,the cultivation temperature of 30℃ and medium initial pH 6~7,and the optimal Cu2+ concentration of 15 ?mol/L.
8.Data Mining Technique and Its Application in Management Against Hospital Infection in Operating Room

OBJECTIVE To explore the application of data mining(DM) technique in the management against hospital infection in operating room.METHODS The data in hospital information system(HIS) were comprehended,prepared and then excavated by the base of DM technique,which were used to evaluate the management against hospital infection in operating room.RESULTS DM could be applied in variety fields against hospital infection in operating room,such as,intelligent preparing system of sterile and disposable articles in operating room,prediction of incisional wound infection after operation to arrange operation reasonably and to prevent abuse of antibiotic,analysis of the application of antibiotics during peri-operation and the rules of their drug resistance happened,and mangement of visitors and trainees,in solving the difficulty and avoiding the error using HIS,and in dynamic monitoring of routine work of hospital infection in operating room.CONCLUSIONS The new technique of DM can be applied to distinguish those unknown,effective,novel and potential information or knowledge from database,and can be effectively applied on the management of hospital infection in operating room.
9.Integrons and Multi-drug Resistance among Pseudomonas aeruginosa Strains Isolated from Blood Samples

OBJECTIVE To investigate the incidence of integrons in Pseudomonas aeruginosa strains isolated from blood samples in Peking University People′s Hospital and to analyze the correlation between integrons and drug resistance of P.aeruginosa.METHODS Forty-two strains of clinically isolated P.aeruginosa were collected.The antibiotics susceptibility was tested by K-B methods.Integrase gene of integron was amplified by PCR using degenerate primers.The integrons were classified by restriction fragment length polymorphism(RFLP) analysis of positive PCR products with Hinf Ⅰ restriction enzyme.RESULTS The drug-resistance rates of 42 strains of P.aeruginosa against 20 kinds of antibiotics ranged from 9.5% to 100%.Twenty-three strains were resistant to 12 kinds of antibiotics.Nineteen of the 42 isolates(45.2%) contained integrons,all of which were revealed as class Ⅰ of integrons by RFLP analysis.Neither class Ⅱ nor class Ⅲ of integron was detected.The positive percentage of integrons was increased by years.CONCLUSIONS Class Ⅰ integrons are widespread in isolates from blood samples in our hospital.The presence of integrons is closely associated with multi-drug resistance of P.aeruginosa.
10.Modified Brisson technique for concealed penis in children

Objective To investigate the corrective effect of modified Brisson technique to treat concealed penis in children.Methods From July 2014 to July 2016,selected 12 patients (3-10 years old,mean 7.2 years old) with concealed penis were surgically treated with the modified Brisson technique in the Second Affiliated Hospital of Nanjing Medical University.Their clinical manifestations included the penis buried below the surface of prepubic skin which can be palpated and visualized through pushing the skin around the penis to the pubis and the short foreskin which was not attached to the penile corpus cavernosum.Patients with other conditions such as obesity and penial agenesis that caused buried penis were excluded from the study.The modified Brisson technique consisted of 4 steps.Ventral incision was used to gradually slit the narrow ring of the prepuce and to expose the glans followed by circumfe-rential inner foreskin incision made below the coronary sulcus of penis then the foreskin stripping released the corpus cavernosum.Finally the skin coverage was accomplished by posthioplasty.The operation time,blood loss,complications,postoperative treatments and followed-ups were retrospectively analyzed.Results For all the 12 patients,the operations were successful,lasting 40 to 60 minutes.The bleeding was about 5 to 10 mL and there was no operation-related complications like bleeding and infection of incision.Satisfactory cosmetic outcomes were obtained without retraction and the scars were similar to those of circumcision.All patients had penile lymphedema postoperatively which disappeared within 6 months after operation.Follow-up ranged from 6 months to 1 year,and there were no complications like skin necrosis and contracture or voiding difficulty.Conclusions The modified Brisson technique for concealed penis repair is technically easy and safe.In addition,it provides a good cosmetic appearance,functional outcomes and excellent postoperative satisfaction grades.
