Objective:To study the effect of retinoic acid(RA) which is used in malignant tumor therapy on the secretion of C3 and factor B in human lung cancer cell line A549 induced by TNF-? and IL-6.Methods:C3 and factor B in the culture medium were measured by ELISA and Western blot, the mRNA expression of C3 and factor B was analyzed by RT-PCR.Results:When using TNF-?(10 ?g/L),IL-6(50 ?g/L) and RA(1 ?mol/L),the results of ELISA,Western blot and RT-PCR showed that TNF-? and IL-6 promoted A549 cells excreting C3 and factor B and their mRNA expression;RA alone did not affect C3 and factor B;in TNF-? group and TNF-?+RA group, the amounts of C3 were 91.40?12.59 and 133.59?11.25(ng/10~6 cells) respectively(P0.05),and the amounts of factor B were 13.07?2.50 and 32.89?4.22(ng/10~6 cells) respectively(P
3.SITE-DIRECTED MUTAGENESIS OF TSR1 GENE IN YEAST YARROWIA LIPOLYTICA
Microbiology 1992;0(05):-
The TSR1(thermosensitive rescued)gene associated with the secretion of proteins of the yeast Yarrowia lipolytica was site-directed mutagenesis, restricted and spliced to obtain a series of deletion mutants. These mutants would be useful for studing the function of different domains of TSR1 gene in yeast Yarrowia lipolytica

Objective To establish a self-made chiral column for enantiomeric separation of salbutamol. Methods We used different concentrations of acid, alkali additives in the polar phase flow for enantiomers separation of salbutamol by using 3, 4-dichlorophenyl isocyanate vancomycin chiral column, and discussed the chiral recognition mechanism. Results The ratio of acid to alkali additive in the mobile phase was 0.01%:0.01% (V/V), the flow rate was 1ml/min, the column temperature was 25℃, and the best separation of enantiomers of salbutamol was obtained, the selective factor was 1.16, the separation degree reached 1.41. Conclusion Self-made 3, 4-two chlorophenyl isocyanate vancomycin chiral column is effective for salbutamol separation, and it can be as a reference for developing other similar chiral stationary phase.

Objective To develop a method for determination of catechins and alkaloids in brick-tea by high performance liquid chromatography(HPLC) with diode array detection(DAD),and to explore optimum extraction conditions for catechins and alkaloids components in brick-tea.Methods Catechins and alkaloids were separated and detected by HPLC.Orthogonal experiment and paired t test was carried out to compare the effect of water and alcohol as extraction solvents and to obtain optimum extraction conditions for extracting catechins and alkaloids components in brick-tea.Results Six kinds of catechins of catechin, epicatechin, gallocatechin, epigallocatechin, epicatechin gallate, epigallocatechin gallate and two kinds of alkaloids of caffeine and theobromine were separated and detected simultaneously.Calibration curves between peak areas and concentration of each component in bricktea were linear within a suitable concentration range, and coefficients of determination (R2) were between 0.9990-0.9999; spiked recoveries were from 83.78% to 106.35%,and relative standard deviations(RSD) were between 0.50%-1.51%.The optimum extraction condition for catechins and alkaloids was 80% ethanol,solid-liquid ratio of 1:10,temperature 80 ℃ and extraction time 30 min. Conclusion Alcohol as extraction solvents with optimal combination on HPLC,six kinds of catechins and two kinds of alkaloids are separated accurately, qualitatively, rapidly and sensitively.

This study is to investigate the anti-allergic effect of anthocyanidin and to explore its possible mechanism. The experiments of passive cutaneous anaphylaxis reaction (PCA) and colorimetry were used to determine the effect of anthocyanidin on degranulation of mast cells in vivo. For in vitro study, various concentrations of anthocyanidin (100, 50 and 25 micromol x L(-1)) were added to the culture medium of mast cells cultured with 100 microg x L(-1) of dinitrophenyl (DNP) specific IgE overnight. The azelastine (100 micromol x L(-1)) was selected as the positive control. The antigen (DNP-human serum albumin, DNP-HAS)-induced release of degranulation was measured by enzymatic assay, histamine was determined by EIA, and interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha) were measured by Western blotting, separately. In addition, the effects of anthocyanidin on phosphorylation of NF-kappaB, p38MAPK and Akt were observed by Western blotting. The results showed that treatments with anthocyanidin (100 and 50 mg x kg(-1)) were followed by a decrease in PCA of rats. Anthocyanidin (100 and 50 micromol x L(-1)) obviously suppressed the degranulation from mast cells, whereas results from anthocyanidin (100 and 50 micromol x L(-1)) group indicated significant inhibitory effect on histamine, the calcium uptake, TNF-alpha, IL-6, phosphorylation of NF-kappaB, p38MAPK and Akt of mast cells induced by antigen. Anthocyanidin may suppress the anaphylactic reaction by inhibiting the action of mast cells. NF-kappaB, p38MAPK and Akt at least in part contribute to this event.

Objective Research the correlation between expression of MMP-2 in colorectal cancer and patho- logical factors,or biological behaviors in colorectal cancer. Methods The expression of MMP-2 in 68 cases with col- orectal cancer were detected by immunohistochemical staining. Results The expression of MMP-2 in colorectal can- cer was not correlated with tumor region and histologic type, but related with depth of tumor invasion and metasta- sis.The frequency of positive cases in patients with colorectal cancer with of lymph, node metastasis was significantly higher than that in patients without lymph node metastasis(P
