2.A CYTOCHEMICAL STUDY OF B CELLS IN GERMINAL CENTER OF HUMAN LYMPH NODE AND TONSIL

Several specific monoclonal antibodies for B, T and natural killer (NK) cell were used to investigate the B cell localization and the expression of their phe- notype in lymphoid nodules on frozen and paraffin sections of human tonsil and lymph node by means of an immunocytochemical ABC technic. The results indi- cate that monoclonal antibodies reactive with germinal centers in tonsil and ly- mph node gave a simlar results and the results indicate that transformation and germination of germinal center cells involve phenotype changs but except T-200. For example, in the lower zone of germinal center, the lymphoblasts are weakly stained for IgM andLN-2 antibodies, but not for OKB-2 and BA-1, while in th upper zone the centrocytes are intense staining for IgM, LN-2, OKB-2 and BA-1 antibodies arelight or moderate staining separately Further charaterization of B cells in upperzone is frenquently observed clcavages on their nuclear memb- rane. In the mantle zone, the lymphocytes are strongly reacted with OKB-2 and BA-1, middle staining for LN-2 and light staining for sIgM. Plasma cell is only reactive with T-200 and IgM antibodies.
3.Clinicopathologic analysis of vulnerable plaque in autopsy with acute coronary syndrome

Objective To observe pathohistological features of vulnerable plaques in coronary arteries. Methods Autopsy coronary samples from 67 patients died of acute coronary syndrome (ACS) and 60 patients of non-cardiac death from 1992 to 2006 in Beijing Hospital were examined. Morphological features of vulnerable plaques of ACS cases were evaluated in terms of thrombus, ratio of lipid core, the minimal thickness of fibrous cap and the density of inflammatory infiltration. Results ( 1 ) There are 305 plaques in ACS group and the incidence of big lipid core is 153 (50. 16% ), thin fibrous cap is 187(61.31%),inflammatory infiltration is 263 (86. 23% ), neovasculature conformation is 217 (71. 15% ), severe stenosis is 26 ( 8. 52% ), calcification is 238 ( 78.03% ), superficial calcified nodule is 26 ( 8. 52% ), fissured plaque is 12(3.93% ), endothelial denudation is 3(0. 98% ) and intraplaque hemorrhage is 54( 17. 70% ),which are significantly higher than control samples except endothelial denudation ( P < 0. 01 ). ( 2 ) The incidence of vulnerable plaques in ACS group is significantly higher than in the control group (89. 51% vs.21.98%, P < 0. 01 ). There are 4. 07 sections of vulnerable plaques with high density of inflammatory infiltration out of 4. 55 sections reviewed in ACS patients, while there are 0. 85 sections of vulnerable plaques with mild inflammatory infiltration out of 3. 87 sections reviewed in the control cases. Conclusions Formation of vulnerable plaque was an important pathological factor for the development of ACS. The major morphological characteristics of vulnerable plaque are big lipid core, thin fibrous cap, inflammatory infiltration, neovascularization, severe stenosis, plaque rupture, and endothelial denudation suggesting inflammation performed an important role in the formation of vulnerable plaque.
4.Clinical evidence in concurrence of retinitis pigmentosa and glaucoma.

Glaucoma is rarely complicated by retinitis pigmentosa (RP). To provide clinical evidences for this rare situation, we report the concurrence of these two diseases in two children of a Chinese family. In the present two-child Chinese family without positive history, the older sister presented with bilateral sector RP and coexisting chronic angle-closure glaucoma, and the brother with bilateral whole RP but without coexisting glaucoma. Clinical evidences in concurrence of variants of RP and glaucoma because of possible different gene mutations from the same genetic background represent a rare situation, which may provide clues for future researches in molecular pathogenesis of these rare diseases.

5.Impact of fermentation system initial status on oscillations in very high gravity ethanol continuous fermentation process and analysis of fermentation efficiency improvement.

Prior research reported the oscillatory behavior characterized by long period and high amplitude during high gravity continuous ethanol fermentations at the dilution rate of 0.027 h(-1). In this paper, high gravity continuous ethanol fermentations using Saccharomyces cerevisia at different dilution rates were carried out. Similar oscillations were observed when the dilution rate was switched to 0.04 h(-1). Both oscillatory and steady processes can be achieved at dilution rates of 0.027 or 0.04 h(-1), which depends on the initial status of the fermentation system. However, compared to steady process at the same dilution rate of 0.04 h(-1), the average residual sugar concentration was lowered by 14.8% for the oscillatory process, while the average ethanol concentration and productivity were increased by 12.6% and 12.3%, respectively. Further investigation revealed that besides the lag time, oscillatory processes were different from steady ones in kinetics because a higher specific growth rate can be achieved at the same residual sugar and ethanol concentrations (increased by 53.8% in average).

6.Fusion expression of Escherichia coli heat-labile enterotoxin B subunit gene and foot-and-mouth disease virus type O VP1 gene and immunogenicity analysis.

LTB gene fragment was amplified by PCR from plasmid pMDTLT, and a recombinant plasmid pETLTBVP1 was constructed by inserting LTB gene fragment into VP1 gene expression plasmid pETVP1 constructed previously. The recombinant plasmids were transformed into E. coli BL21(DE3) and induced to express by IPTG. The recombinant protein existed in the inclusion body and its molecular weight was about 39 kD proved by SDS-PAGE analysis. Western blotting showed that the fusion protein could be reacted with both anti-FMDV and anti-cholera toxin serum demonstrating the immunoactivity of the fusion protein. Strong immune responses can be induced in mice inoculated with the fusion protein intraperitoneally, and the serum antibody level is higher than that of commercial foot-and-mouth disease vaccines.

7.Application of condensed Chinese version of the MOS 36-item short form health survey in assessment of quality of life among adult patients with Kashin-Beck disease in Gansu province

Objective To evaluate the reliability and validity of the application of condensed Chinese version of the MOS 36-item short form health survey (SF-36) in assessment of quality of life among adult patients with Kashin-Beck disease,and to provide a scientific basis in rehabilitation of the patients.Methods Four hundred and twenty seven eases of adult patients with Kashin-Back disease and 419 healthy individuals randomly selected in Kashin-Beck disease endemic areas in 8 counties of Gansu province were surveyed with the SF-36.The reliability of the SF-36 was assessed by split-half reliability and Cronbach's α coefficient and the validity through principal component factor analysis and correlation analysis,etc.The dimension scores of different people were obtained by analysis of variance and univariate t-test.Results The split-half reliability of all the 8 dimensions was greater than 0.6 and the Cronbach's α coefficient was greater than 0.8; the pearson correlate coefficients of all the items to their dimensions were greater than 0.391.SF-36 contained 8 domains and 2 summary scales in the factor analysis.The score differences of quality of life in different ages of the patients,different stages of the disease were statistically significant (all P < 0.05).Conclusion The SF-36 is practical in studying the quality of life among adult patients with Kashin-Beck Disease.
8.Clinical characteristics of glucocorticoid-induced eye adverse reactions in children with primary nephrotic syndrome

Objective:To investigate the clinical features and side effects,with regard to glucocorticoid-induced ocular hypertension,glaucoma or cataract in children with primary nephrotic syndrome.Methods:Clinical data were collected and analyzed from 71 cases of primary nephrotic syndrome with glucocorticoid-induced ocular hypertension,glaucoma or cataract from Jun.2014 to Jun.2016.These children were hospitalized in Peking University First Hospital.Results:Totally 1 580 children with primary nephrotic syndrome were collected,glucocorticoid-induced complications in eyes were found in 71 cases,and the incidence was 4.5%.There were 66 cases with ocular hypertension,2 cases with glucocorticoid glaucoma,2 cases with glucocorticoid glaucoma combined with cataract,1 case with high intraocular pressure combined with cataract.There were 41 boys and 30 girls with eye-related side effects caused by glucocorticoid.The average age of onset of glucocorticoid-induced eye adverse reactions in children with primary nephrotic syndrome in our research were 8 (2,16) years.The average duration or interval time from glucocorticoid medication use to eye adverse effects was 157 (6,420) days.No statistical significance was found in intraocular pressure between different genders,types of glucocorticoid,different route of glucocorticoid and whether methylprednisolone pulse treatment (P > 0.05).There was no significant correlation between age,body mass index,blood pressure,cumulative dosage,duration time of glucocorticoid,mean daily dosage and glucocorticoid-induced ocular hypertension (P > 0.05).The ocular hypertension was controlled after treatment.Conclusion:Children with nephrotic syndrome after treatment of glucocorticoid are susceptible to ocular complications,and the occurrence of ocular hypertension is closely related to glucocorticoid susceptibility of the nephrotic children.Regular eye monitor is indispensable for the children suffering from primary nephrotic syndrome.
9.Prognostic value of tumor-infiltrating lymphocyte subtypes in residual tumors of patients with triple-negative breast cancer after neoadjuvant chemotherapy

Background::After neoadjuvant chemotherapy (NAC), non-pathological complete response of breast cancer patients can benefit from tailored adjuvant chemotherapy. However, it is difficult to select patients with poorer prognosis for additional adjuvant chemotherapy to maximize the benefits. Our study aimed to explore whether the subtypes of tumor-infiltrating lymphocytes (TILs) in residual tumors (RT) is related to the prognosis of triple-negative breast cancer (TNBC) after NAC.Methods::Data from patients with primary TNBC consecutively diagnosed at the Breast Disease Center of Peking University First Hospital from 2008 to 2014 were retrieved, and the cases with RT in the breast after NAC were enrolled. TILs subtypes in RT were observed by double-staining immunohistochemistry, and counted with the median TILs value per square millimeter as the cut-off to define high versus low TILs density in each subtype. The relationships between the TIL density of each subgroup and the clinicopathological characteristics of the RT after NAC patients were analyzed by Fisher exact test. Disease-free survival (DFS) and overall survival (OS) were analyzed by the Kaplan-Meier method and log-rank statistics.Results::A total of 37 eligible patients were included in this study, and the median follow-up period was 50 months (range 17–106 months). There was no significant correlation between the infiltrate density of CD4 +, CD8 +, CD20 +, and CD68 + lymphocytes and clinic-pathological characteristics. Significantly better prognosis was observed in patients with high CD4 +-TILs (DFS: P = 0.005, OS: P = 0.021) and high CD8 +-TILs (DFS: P = 0.018) and low CD20 +-TILs (OS: P = 0.042). Further analysis showed that patients with CD4 +/CD20 + ratio greater than 1 (DFS: P = 0.001, OS: P = 0.002) or CD8 +/CD20 + ratio greater than 1 (DFS: P = 0.009, OS: P = 0.022) had a better prognosis. Conclusions::Subtypes of TILs in RT is a potential predictive biomarker of survival in TNBC patients after NAC.
10.Results of functional management of condylar fracture in 3 to 16 yeats old children

Objective To investigate the effect of soft occlusal splint in the treatment of condylar fracture in children and adolescents.Methods Twenty-three children with condylar fracture aged from 3 to 16 were included in this study.Impressions of both jaws were taken and stone working models poured.After occlusion was recovered by mounting on a bionic articulator,a soft occlusal splint was fabricated.The occlusal splint was worn for 1 to 3 months accompanied with functional exercise.Follow-up was carried out by clinical observation and panoramic image.Results Clinical satisfactory results were obtained in all the patients with good occlusion,unimpaired function and normal growth and development of the mandibles.Panoramic image showed reconstruction of the fractured condyles,which were flattened and short.Conclusions Soft occlasal splint is a promising approach for treating condylar fracture in children.
