2.Study on recovery and its influencing factors of ferulic acid and tetramethylpyrazine in cerebral microdialysis probe.

To establish a method for detecting microdialysis recovery of tetramethylpyrazine (TMP) and ferulic acid (FA) and investigating the influencing factors, providing the basis for further in vivo microdialysis experiments. The concentration of FA and TMP in dialysates were determined by high pressure liquid chromatography ( HPLC) and probe recovery were calculated respectively. The influence of the flow rates, medium concentration, temperature and in vivo probe stability on the recovery of FA and TMP were investigated by using concentration difference method (incremental method and decrement method). The recovery obtained by incremental method were similar to by decrement method. The in vitro recovery rate of FA and TMP decreased with the increase of 1-2.5 μL min(-1), and increased obviously with the temperature of 25-42 degrees C under the same conditions. The concentration of FA and TMP had no obvious effect on the probe recovery under the same flow rate. In addition, the recovery of TMP and FA remained stable and showed similar trends under the condition of four concentration cycles, indicating that the intra day reproducibility of the concentration difference method was good. The recovery of brain microdialysis probes in vivo 8 h maintained a relatively stable, but certain differences existed between different brain microdialysis probes, demonstrating that each probe was required for recovery correction in vivo experiment. Microdialysis sampling can be used for the local brain pharmacokinetic study of FA and TMP, and retrodialysis method can be used in probe recovery of FA and TMP in vivo.

3.Scalp flaps pedicled with superficial temporal vessel and hair removal for reconstruction of facial skin defects.

OBJECTIVETo investigate the therapeutic effect of scalp flaps pedicled with superficial temporal vessel and hair removal for reconstruction of facial skin defects.
METHODSFrom Jan. 2010 to Dec. 2011, 6 cases with large facial skin defects were treated with scalp flaps pedicled with superficial temporal vessel and hair removal. At the first stage, the skin expanders were implanted subcutaneously at the homolateral side according to the defect size. After the expansion was finished, the expanded flaps pedicled with superficial temporal vessel were used to reconstruct the facial skin defects at the second stage. 2 weeks after operation, hair removal was performed to remove the hair on flaps. 4-6 treatments were needed.
RESULTSSeven flaps in 6 cases were survived completely. The wounds at donor sites were closed directly. The effect of hair removal was reliable. The patients were followed up for 6 months to 1 year. The flap color, texture and thickness had a good match with surrounding skin tissue.
CONCLUSIONSIt is a good option to reconstruct facial skin defects with scalp flaps pedicled with superficial temporal vessel and hair removal.

4.Thymosin beta10 expression and actin filament organization in tumor cell lines with different metastatic potential.

OBJECTIVETo investigate the expression of thymosin beta10 (Tbeta10) and related changes of actin filament organization in human tumor cell lines with different metastatic potential.
METHODSFour groups of nine human tumor cell lines with different metastatic potential were analyzed for the expression of Tbeta10 mRNA detected by northern-blot and its peptide by immunohistochemical staining. The filamentous actin (F-actin) was stained with TRITC-phalloidin to detect changes in actin organization.
RESULTSIn comparison with the non and/or weakly metastatic counterparts, Tbeta10 was upregulated in highly metastatic human lung cancer, malignant melanoma and breast cancer cell lines. TRITC-phalloidin staining revealed less actin bundles and a fuzzy network of shorter filaments in the highly metastatic tumor cells, while in the non and/or weakly metastatic cancer cell lines, there were thick and orderly arranged actin filaments.
CONCLUSIONSTbeta10 levels correlate positively with the metastatic phenotype in human tumors currently examined. The increased metastatic potential of tumor cells is accompanied by the loss of F-actin and poorly organized actin skeleton. There is a consistent correlation between the elevated Tbeta10 expression and the disrupted actin skeleton.

5.Effect of high frequency electrocoagulation by ultrafine-needle on the cutaneous capillary hemangiomas: a preliminary study on cock combs.

OBJECTIVETo investigate the injury effect of high frequency electrocoagulation (HFE) by ultrafine-needle on the cutaneous capillary hemangiomas by histopathologic study of cock comb.
METHODS32 male cocks were randomly divided into 4 groups with 8 animals in each group. 3 experimental groups received HFE with different power (6W, 8W, 10W) on the cock comb. Specimens were taken from cock combs for histologic study immediately after treatment and 7, 14, 21, 28 days later.
RESULTSAfter treatment, the cock combs turned pale and followed with scar formation. The shape of combs changed a lot after high power treatment. Immediately after treatment, the capillary number in the experimental groups decreased markedly under light microscope, compared with that in control group (P < 0.05). After that, the capillary number in experimental group increased. 7 days after treatment, the capillary number in experimental groups was still significantly lower than that in control groups (P < 0.01), but not markedly different between the three experiment groups (P > 0.05). Collagen formation also increased after treatment (P < 0.05), which was higher in experimental groups with high power (P < 0.05, correlation coefficient r > 0.5).
CONCLUSIONSHFE with ultrafine needle can effectively decrease the capillary number in cock comb. The 6W treatment is mini-invasive and will not destroy the comb shape with good cosmetic results. The technique may be used for the treatment of cutaneous capillary hemangioma.

6.Effects of myocardial platelet rich plasma injection on rats with acute myocardial infarction:(99)Tc(m)-MIBI gated SPECT imaging evaluation results.

OBJECTIVETo observe the effect of direct myocardial injection of platelet rich plasma (PRP) on cardiac function, ventricular remodeling and myocardial perfusion.
METHODSMyocardial infarction was induced in 30 Fisher rats by left anterior descending coronary artery (LAD) ligation. One week after LAD ligation, rats in control group (n = 15) received 0.5 ml saline myocardial injection and rats in PRP group (n = 11) received 0.5 ml PRP myocardial injection.(99)Tc(m)-methoxyisobutylisonitrile (MIBI) gated single photon-emission computed tomography (SPECT) imaging was applied at 1 week post LAD ligation and prior myocardial saline/PRP injection and repeated at 4 week post LAD ligation to assess myocardial perfusion and ejection fraction (EF), left ventricular end-diastolic volume (LVEDV), left ventricular end-systolic volume (LVESV), and the number of infarct segments.
RESULTSEF remained unchanged between 1 and 4 weeks post LAD ligation in control group and was significantly higher at 4 weeks post LAD ligation than at 1 week post LAD ligation in PRP group (P < 0.05). LVESV was significantly increased in the control group (P < 0.05) while remained unchanged in the PAP group (P < 0.05) at 4 weeks post LAD ligation compared that at 1 week post LAD ligation. LVEDV remained unchanged in the control group (P > 0.05) and significantly increased in the PRP group (P < 0.05) from 1 week to 4 weeks post LAD ligation. Myocardial perfusion remained unchanged in the control group and significantly improved in the PRP group at 4 week post LAD ligations. Histological examination also confirmed that PRP treatment can decrease infarct size [from (47 ± 19)% to (36 ± 11)%], increase ventricular wall thickness [from (3.1 ± 0.9) mm to(4.6 ± 1.8) mm] (P < 0.05).
CONCLUSIONMyocardial PRP injection could improve cardiac function and reperfusion in this rat model of acute myocardial infarction.

7.Investigation of the function of the glass colorant on the machinable infiltrated ceramics color.

OBJECTIVETo investigae the function of the glass colorant on the color of the machinable infiltrated ceramics(MIC).
METHODSFive kinds of glass with different colorant were infiltrated through the aluminous matrix by heating the components to 1 100 degrees C for 2 hours. The specimens surface was polished, and their thickness was 0.5 mm.
RESULTSThe refractive index of the MIC infiltration glass was 1.59691 (587.6 nm, nd) . The most different parameter of the MIC color were L*, then a*, and b* had little difference . The parameters of the color space of MIC were: L*(64.55-71.46), a*(3.35-7.38), b*(10.00-12.41), Ca*b*(11.38-13.95), ha*b*(54.07-73.00). These were almost close to the color parameters of Vita In-ceram.
CONCLUSIONThis experiment proved that the glass colorant was changed the MIC color parameters, and the main function was on L*, then a*. The ceramic color was up to the requirement of clinic.

8.Increased autoantibody production against AT(1)-receptors and alpha(1)-adrenergic receptors in hypertensive patients.

OBJECTIVETo observe autoantibodies production against AT(1)-receptors and alpha(1)-adrenergic receptors and association to risk factors, such as sex, age, family history, course of hypertension and other cardiovascular diseases in hypertensive patients.
METHODSA total of 690 patients with essential hypertension admitted to our hospital were selected and autoantibodies against AT(1)-receptors and alpha(1)-adrenergic receptors were detected by ELISA. Multiple logistic regression analysis was performed based on obtained data.
RESULTSPositive rates for antibody against AT(1)-receptors and alpha(1)-adrenergic receptors were 47.1% (325/690) and 36.4% (251/690) respectively in this group of patients. Duration of hypertension history was significantly longer in the antibody against AT(1)-receptors and alpha(1)-adrenergic receptors positive groups [(9.3 +/- 11.0) year, (9.9 +/- 11.1) year] compared to the negative groups [(7.3 +/- 9.3) year, (7.2 +/- 9.5) year, all P < 0.01]. The ratio of family history with hypertension was also significantly higher in antibody positive groups than negative ones (47.69% vs 39.18%, P < 0.01). Regression analysis demonstrated that 5 risk factors were related to positive production of autoantibody against AT(1)-receptors including female gender, age, family history, duration of hypertension history and diabetes. However, just age, family history, duration of hypertension history were main factors responsible to the production of autoantibody against alpha(1)-adrenergic receptors (all P < 0.05).
CONCLUSIONThe environmental and genetic factors contributed to the autoantibody production in patients with essential hypertension.

9.Association between positive autoantibodies against AT1-receptor and cardiac remodeling in patients with hypertension.

OBJECTIVETo observe the association between positive autoantibodies against AT(1)-receptor and cardiac remodeling in primary hypertensive patients.
METHODSEchocardiography was performed and serum autoantibodies against AT(1)-receptor were detected by enzyme linked immunosorbent assay (ELISA) in 592 patients with primary hypertension. The differences on blood pressure level, course of hypertension, vasoactive substance and echocardiography parameters between the positive group and negative group were compared. Factors related to left ventricular enlargement were analyzed by multiple logistic regressions.
RESULTSThe positive percentage of autoantibodies against AT(1)-receptor was 38.0% (225/592). End-diastolic right atrial and left ventricular diameters in positive group were significantly larger than that in negative group (P = 0.049 and P = 0.044, respectively). Regression analysis demonstrated that positive autoantibodies against AT(1)-receptor, male gender, diastolic blood pressure and course of hypertension were related to left ventricular enlargement (all P < 0.05).
CONCLUSIONThe autoantibodies against AT(1)-receptor is associated with left ventricular and right atrial enlargement in hypertensive patients.

10.Concurrent chemoradiotherapy with sodium glycididazole and cisplatin for local advanced nasopharyngeal carcinoma.

OBJECTIVETo evaluate the radiosensitivity and toxicity of sodium glycididazole and cisplatin in concurrent chemoradiotherapy for local advanced nasopharyngeal carcinoma (NPC).
METHODSSixty patients with local advanced NPC (T3-4N2-3M0) were randomly divided into chemoradiotherapy group (n=30) and chemoradiotherapy plus sodium glycididazole group (n=30). All the patients received radiotherapy with (60)Co or 6-8 MV linear accelerator and concurrent injection of cisplatin at a weekly dose of 20 mg/m square. In sodium glycididazole group, the patients received injections of sodium glycididazole at 800 mg/m square prior to the radiotherapy 3 times a week.
RESULTSAt the end of the therapy and 3 month after the radiotherapy, a response rate of 100% was achieved in both of the groups. But at the end of the therapy, the chemoradiotherapy plus sodium glycididazole group showed a significantly higher rate of complete tumor remission than the chemoradiotherapy group (93.3% vs 73.33%, chi(2)=4.32, P=0.038). The patients in the two groups showed similar tolerance of the therapy during the observation.
CONCLUSIONSodium glycididazole plus cisplatin can accelerate the tumor remission and improve the complete remission rate in patients with local advanced NPC without causing severe toxicity.
