Progress in Biochemistry and Biophysics 2008;35(1):56-62
Chlamydial infection in human urogenital tract induces inflammation and causes tissue damage and scarring. It is thought that cytokine production by the Chlamydia-infected cells plays a key role in chlamydial disease processes. Although many cytokines have been detected during chlamydial infection, little is known about the molecular mechanisms on how Chlamydia triggers and sustains the inflammatory cytokine cascades. In the current study, chlamydial infection of the human cervical epithelial cell line HeLa cells can induce the production of IL-8, IL-1α, IL-1β and IL-6. Using inhibitors for probing intracellular kinase signaling pathways required for the Chlamydia-induced cytokines, it was found that the Chlamydia-activated MAPK / P38 pathway is required for the chlamydial induction of IL-1α and IL-6 while both the Chlamydia-activated MAPK/ERK and MAPK/P38 pathways contribute to the production of IL-8.

Journal of Forensic Medicine 2016;32(3):204-210
Source identification of human biological materials in crime scene plays an important role in reconstructing the crime process. Searching specific genetic markers to identify the source of different human biological materials is the emphasis and difficulty of the research work of legal medical experts in recent years. This paper reviews the genetic markers which are used for identifying the source of human biological materials and studied widely, such as DNA methylation, mRNA, microRNA, microflora and protein, etc. By comparing the principles and methods of source identification of human biological materials using different kinds of genetic markers, different source of human biological material owns suitable marker types and can be identified by detecting single genetic marker or combined multiple genetic markers. Though there is no uniform standard and method for identifying the source of human biological materials in forensic laboratories at present, the research and development of a series of mature and reliable methods for distinguishing different human biological materials play the role as forensic evi-dence which will be the future development direction.

Objective To study the effects of rehabilitation training on learning and memory ability and the expression of growth-associated protein-43(GAP-43)in rats with vascular dementia. Methods Forty-four female Sprague-Dawley rats were randomly assigned to a rehabilitation group(n=20),an immobilization group(n=20),and a sham-operation group(n=4).The rats in the former 2 groups were operated on to establish the experimental vascular dementia model by repeatedly ischemia/reperfusion injury of brain induced by ligation of bilateral common carotid arteries and lowering of blood pressure induced by intraabdominal injection of sodium nitroprusside. The rats in rehabilitation group were administered with rotating bar and rolling cage exercises for 1 hour once daily, while those in the immobilization group were immobilized and without any exercise:the rats in sham-operation group could move freely in cage.Learning and memory tests were preformed by using step-down avoidance test at the 27th and 28th days after operation.Immunohistochemical staining was used to detect GAP-43 expression in hippocampus CA1 area at different time points after operation. Results The rats in rehabilitation group demonstrated better learning and memory ability than those in immobilization group(P<0.01),and more GAP-43 expression in hippocampus CA1 ar-ea than those in immobilization group and sham-operation(P<0.01).Conclusion Rehabilitation training can im-prove the learning and memory ability of rats with experimental vascular dementia, and the mechanism is probably re-lated to the increase of GAP-43 in hippocampus CA1 area.

OBJECTIVE:To provide reference for the development and improvement of pharmaceutical care in our hospital. METHODS:By questionnaire investigation, habits and understarding situation and demand of knowledge of drug use among pa-tients in our hospital were randomly collected by face-to-face interview or network platform. A statistical analysis was carried out on obtained results. RESULTS:In this questionnaire investigation,289 questionnaires were sent out face-to-face,and 282 valid ques-tionnaires were returned with effective feedback rate of 97.58%. Totally 51 valid questionnaires were returned through network plat-form. A total of 333 valid questionnaires were returned through two ways. The results of questionnaire investigation showed that 44.14% of the surveyed patients would read the drug instructions before taking drugs;41.14% of the surveyed patients would pay more attention to ADR and cautions stated in drug instructions. More than 60% of the surveyed patients had various bad habits of drug use. When getting better,66.07% of the surveyed patients would stop using drugs or reduce the dosage. When having not im-proved,26.73% of them would change drugs or increase dosage.Consulting with the medical staff was the most common source(69.07%)of drug use knowledge, and it was also the most trusted source(84.08%). Among the knowledge of drug use, surveyed patients most expected to understand theADR and side effects(65.46%),and the selectionindication and main us-er,cautions,drug interaction in multiple useandusage and dosagewere also selected frequently, and the choices of some options were significantly affected by the characteristics of people, such as age and education degree(P<0.05). The most expected way to acquire drug use knowledge wasface-to-face consultation with medical staff (72.97%),followed bytelephone consulta-tionandnetwork consultation, and the choices of some options were significantly affected by the characteristics of people, such as age and education (P<0.05). CONCLUSIONS:Patients in our hospital haven't pay enough attention to drug instructions and their content, and bad habits of drug use still exist. Sources of drug use knowledge are diverse, but confidence of other sources is not enough except for source of medical staff. The demands for the knowledge of drug use are different, however, the present form and content of pharmaceutical care in our hospital cannot fully meet the needs of patients.

Objective To investigate the inhibitory effect of octreotide on transforming growth factor-alpha (TGF-?)-induced cell proliferation of human hepatocellular carcinoma cells and its possible mechanism. Methods The effect of octreotide on TGF-?-induced cell proliferation of the liver cancer cells (LCC) was evaluated by immunohistochemistry method. The effect of octreotide on TGF-? secretion and epidermal growth factor receptor(EGFR) expression in the cells was determined by reverse-transcriptase polymerase chain reaction(RT-PCR). The effect of octreotide on extracellular signal-regulated protein kinase (ERK) expression in the cells was measured by Western-blot and immunohistochemistry method. Results The TGF-?-induced expressions of proliferating cell nuclear antigen (PCNA) in nucleus were obviously increased by TGF-?. TGF-?mRNA index of hepatocellular carcinoma cells was decreased by octreotide. Octreotide inhibited significantly the expressions of EGFR mRNA induced by TGF-?. Octreotide inhibited significantly the expressions of ERK protein induced by TGF-?. There was intense staining in the nucleus of the cells by TGF-? and weak staining in the nucleus of the cells treated simultaneously by octreotide and TGF-?.Conclusions Octreotide can inhibit the secretion of TGF-?, the expression of EGFR, and the signal transduction of EGFR of LCC, and consequently exerts an inhibitory effect on TGF-?-induced hepatocellular carcinoma cells proliferation.
