1.Comparison of the intraocular pressure value measured with Goldmann applanation tonometer and non-contact tonometer

AIM: To assess the relative agreement of GAT and NCT in IOP measurement by comparing the differences between Goldmann applanation tonometer (GAT) and non-contact tonometer (NCT) in intraocular pressure (IOP) detection.METHODS: IOP of 529 eyes of 265 volunteers were measured with both NCT and GAT, respectively.RESULTS: The measurement results of NCT were lower than that of GAT, there was significant difference between the IOP measured with NCT and GAT (19.13 vs23.43, t=22.644, P<0.05). With the increasing of IOP values, the difference magnitude was greater, especially in IOP group that was more than 30mmHg, but the correlation coefficient became lower.CONCLUSION: The measurement results with NCT are lower than that of GAT. When the IOP with the NCT is in borderline value, it need be corrected with GAT, in order to discover the pathologically elevated IOP and avoid the misdiagnosis and mistreatment of glaucoma.
2.Analysis on envelope gene of type Ⅰ dengue virus isolated from Guangzhou area in 2009

Objective To sequence and analyze the envelope (E) gene of type Ⅰ dengue virus isolated from Guangzhou in 2009 for tracing the infection source. Methods The serum samples were collected from patients diagnosed with dengue fever in Guangzhou area during 2009. Dengue virus was isolated and cultured in C6/36 cells.The whole length of E gene was amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) and then sequenced. The phylogenetic tree was drawn by neighbor-joining method. The bioinformatics analysis was performed by combining the phylogenetic information and the epidemiology data. Results Four strains of type Ⅰ dengue virus were isolated from 19 samples. E gene of these strains was amplified and sequenced. The phylogenetic analysis showed that 09/GZ/9104 strain and 09/GZ/9236 strain had identical nucleotide sequence and fell within the American/African group, 09/GZ/11534 stain and 09/GZ/11562 strain had similar sequence homology and fell within the Asian group. Conclusion The typeⅠdengue viruses in Guangzhou area in 2009 are imported, which belong to two genotypes and may come from two independent origins respectively.
3.Methylation status of TES gene promoter region in 25 patients with leukemia

Objective To study the methylation status of Testin protein (TES) promoter region in bone marrow of leukemia pa-tients ,and to explore the relationship between TES gene and leukemia .Methods The bone marrow specimens of 13 patients with primary acute myeloid leukemia and 12 patients with acute myeloid leukemia were studied .Bone marrow samples from 15 healthy subjects were used as controls ,the methylation level of TES gene promoter in bone marrow was detected by BSP ,the methylation status between healthy subjects and leukemia patients ,and whether there was any difference in methylation status between different types of leukemia were analyzed .Results In 25 cases of leukemia ,there were 21 cases of bone marrow samples ,and the hypermeth-ylation of TES gene promoter region was positive ,with a positive rate of 84 .0% .The methylation of the gene was not detected in bone marrow samples from 15 healthy subjects .The methylation rate of TES gene in bone marrow samples of 12 patients with acute lymphoblastic leukemia was 83 .3% ,the methylation rate of TES gene in bone marrow samples of 13 patients with primary acute myeloid leukemia was 84 .6% ,and there was no significant difference between the two groups (P>0 .05) .Conclusion Methylation of TES gene might be one of the biomarkers in the diagnosis of leukemia .
4.Epidemiological characteristics of novel coronavirus confirmed cases and asymptomatic infected patients in Guangzhou

Objective:To analyze the epidemiological characteristics of novel coronavirus positive cases including confirmed cases with clinical symptoms and asymptomatic infected cases in Guangzhou.Methods:Epidemiological data were collected on the nucleic acid positive cases of COVID-19 in Guangzhou from January to September 2020. The epidemiological characteristics, the distribution of time intervals between the confirmed/isolation date and the date of the first positive detection were analyzed, at last the influencing factors for the confirmed cases and asymptomatic infected persons were discussed.Results:From January 7 to September 4 in 2020, a total of 1 097 nucleic acid positive cases were identified, including 658 confirmed cases (59.98%) and 439 asymptomatic infected cases (40.02%). Among the 658 confirmed cases, the median age was 42 years old, the cases indicated two significant peaks. one of the peaks was related to the imported and associated cases from Hubei province, and the other peak was connected with individuals from overseas. In terms of 439 asymptomatic infected cases, the median age was 32 years old. There were two stages in these cases. The first stage followed the second peak of confirmed cases, and the second stage overlapped with the confirmed cases in Guangzhou when the epidemic was in a period of normal prevention and control, mainly related to imported cases from abroad. The asymptomatic infected persons accounted for 57.32% in all the imported infected cases. In both of asymptomatic and symptomatic cases, the positive rate of pharyngeal swabs was higher than that of nasopharyngeal swabs and anal swabs. There were statistically significant differences in age, source of infection and gender composition between confirmed cases and asymptomatic infected persons ( P<0.05). Older age groups were more likely to have clinical symptoms, with ≥40 years being the risk factor for confirmed cases (OR=2.334, P=0.001), and 20-39 years less likely to have clinical symptoms (OR=0.620, P=0.047), compared with the 0-19 years old group. Compared with those infected in China, those infected abroad were less likely to develop clinical symptoms and became confirmed cases (OR=0.723, P=0.013). Women were more likely to have clinical symptoms than men (OR=1.574, P=0.001). Conclusions:At present, asymptomatic infected persons and confirmed patients with clinical symptoms co-existed, and the number of asymptomatic infected patients was higher than that of confirmed cases in Guangzhou. High age, domestic infection and female may be risk factors for confirmed cases. It was of great value to further explore these underlying mechanisms for the prevention and treatment of the COVID-19.

OBJECTIVETo study the chemical constituents from the herb of Gentianopsis paludosa.
METHODColumn chromatogrophy and spectral analysis were used to isolate and identify the constituents.
RESULTSix compounds were isolated and identified as 1,7-dihydroxy-3,8-dimethoxyxanthone (I), 1-hydroxy-3, 7, 8-trimethoxyxanthone (II), 1, 8-dihydroxy-3, 7-dimethoxyxanthone (III), 1-hydroxy-3, 7-dimethoxyxanthone (IV), beta-sitosterol (V), daucosterol (VI).
CONCLUSIONCompounds III-VI were isolated from the plant for the first time.

6.Clinicopathological analysis of primary intestinal non-Hodgkin's lymphoma: a report of 32 cases.

OBJECTIVETo investigate the clinical and pathological features and prognostic factors of primary intestinal non-Hodgkin's lymphoma (NHL).
METHODSThirty-Two cases of intestinal NHL were studied clinically and pathologically. All cases were reclassified according to the WHO classification of lymphoma in 2001.
RESULTSTwenty-one cases (65.6%) were diagnosed as B-cell lymphomas, including 15 cases of diffuse large B-cell lymphoma, 2 cases of mantle cell lymphoma, 1 case of follicular lymphoma, 2 cases of MALT lymphoma and 1 case of Burkitt's lymphoma. Ten cases (31.2%) were diagnosed as T cell lymphomas, including 2 enteropathy-associated T cell lymphomas (EATCL) and 8 cases non-EATCL. One case (3.1%) was diagnosed as histiocytic lymphoma. Nine cases were at stage I-II and 23 cases were at stage III-IV. Follow-up information was available in all cases ranging from 4 to 168 months (mean 25.6 months). Fifteen cases died of the disease. The mortality rates were 33% for B cell lymphoma and 80% for T cell neoplasms. Based on Cox multivariate analysis, the clinical stage and histological types of the disease were important prognostic factors (P < 0.05). T cell lymphoma had a relatively poor prognosis.
CONCLUSIONSDiffuse large B cell lymphoma is the most common subtype of primary intestinal lymphomas. Colon is the most common site involved, followed by the small intestine. Early stage of the disease and low-grade lymphoma appear to have a better prognosis. T cell lymphoma has a very poor prognosis.

7.Association between calpain-10 gene polymorphism and risk of type 2 diabetes mellitus:a Meta analysis

Objective The purpose of this study was to approach the relation of SNP43,SNP44 locus, main haplotypes and haplotype combinations with type 2 diabetes mellitus(T2DM).Methods According to the theory and principles of systematic review,data from case-control studies regarding the association between calpain-10(CAPN10) gene and T2DM were derived through electronic search of PubMed and Chinese journals databases.To gain a more precise estimation of the relationship,a stratified Meta-analysis with four subgroups was pertbrmed according to the races.Publication bias Was also assessed.Results The association with T2DM in different races was evaluated.In Mongoloid race,SNP43-G allele,G/G genotype and 111/221 haplotype combination showed notable association with T2DM with Ors (95%CI) as 1.368(1.155-1.620),1.437(1.186-1.741) and 2.762 (1.287-5.927) respectively.In Caucasoid race,SNP44-C allele,111/111 hapotype combination showed strong relationship with T2DM with Ors(95%CI) as 1.144(1.023-1.278),1.291(1.050-1.586) respectively.In Hybrid race,only one positive finding Was obtained which Was SNP44-C allele with OR(95%CI)as 1.653(1.025-2.665).Conclusion SNP43-G allele,G/G genotype,111/221 were risk factors to Mongoloid race.And SNP-C allele,111/111 haplotype combination were risk factors to Caucasoid race,and SNP44-C allele to Hybrid race.
8.Study on steroidal saponins from Dioscorea zingiberensis and their platelet aggregation activities.

Using the absorbent resin, silica gel and ODS column chromatography as well as semi-preparative HPLC, ten compounds were isolated from 70% ethanol extract of tubers of Dioscorea zingiberensis C. H. Wright, and their structures were elucidated as trigoneoside XIIIa (1), parvifloside (2), trigoneoside IVa (3), deltoside (4), protobioside (5), lilioglycoside k (6), zingiberensis newsaponin I (7), deltonin (8), prosapogenin A of dioscin (9), and trillin (10) on the basis of NMR and MS spectral data analysis. Among these compounds, 1, 3, 5 and 6 were isolated from this plant for the first time. In the screening test on platelet aggregation, compounds 7 and 8 exhibited induction effect on platelet aggregation, while compound 9 exhibited significant inhibitory effect on platelet aggregation in vitro.

9.Effects of ulinastatin slow flow and no reflow phenomenon during emergency percutaneous coronary artery intervention

Objective To study the effect of ulinastatin on no reflow or slow flow in the infarct related artery in patient with acute ST-elevation myocardial infarction (STEMI) undergoing emergency percutaneous coronary interventional therapy (PCI).Methods 180 STEMI patients were divided into the control group (n=100) and the ulinastatin treatment group (n=80).The control group received conventional PCI treatment and the treatment group received conventional PCI treatment plus ulinastatin. The level of serum inflammatory factors IL-6,IL-10,superoxide converting enzyme,the infarct related coronary artery reperfusion TIMI flow grade (TFG) and myocardial perfusion grade (TMPG),the results of postoperative cardiac ultrasound examination and clinical outcomes were compared between the two groups.Results Compared with the control group,the level of IL-6 was decreased,while the levels of IL-10 and superoxide converting enzyme were increased significantly in the ulinastatin treatment group(P<0.05).The TFG and TMPG of the infarct related vessels were increased significantly in the ulinastatin treatment group. The left ventricular end diastolic diameter[(54.6 ± 5.2 mm vs. (50.4±4.6) mm,P=0.046)]and left ventricular ejection fraction [(58.4±10.2) % vs. (62.2±9.8) % P=0.048] showed statistical difference between the two groups.Compared to the control,the major cardiovascular event rate of the treatment group during hospitalization (1% vs. 5%, P=0.038), after one month (1.2% vs. 3%,P=0.046) and 6 months (3% vs 12%,P=0.018) were all significantly lower .There was no significant difference in mortality between the 2 groups.Conclusion Ulinastatin may lower the incidence of no flow and slow flow after emergency PCI,improve heart function and the lower the rates of MACE.
10.Molecular epidemiologic analysis on new emerged type 3 dengue virus in Guangzhou in 2009

Objective To analyze and trace the infection source the envelope(E) gene of the new emerged type 3 dengue virus in Guangzhou in 2009. Methods Sera were collected from patients infected with local dengue fever. Dengue virus was cultured and isolated by C6/36 cells. The whole length E gene was amplified from the positive specimen by RT-PCR, thereby sequenced and phylogenetic tree drawn by neighbor-joining method. Both data on epidemiologic and molecular studies were processed and analysed. Results 7 strains of type 3 dengue virus were isolated from samples of the 19 patients. E gene of these strains was amplified. The complete E genes of 7 strains belonged to 1479 nucleotides in length, encoding a polyprotein of 493 amino acids. Data from the phylogenetic analysis showed that 09/GZ/1081, 09/GZ/1483 and 09/GZ/10806 strains fell within the Southeast Asia/South Pacific group. 09/GZ/10616, 09/GZ/11144, 09/GZ/11194 while 09/GZ/13105 strains fell within the India group. Conclusion The type 3 dengue virus identified in Guangzhou area in 2009 was imported and could be devided into two genotypes.
