Objective To explore the pattern and quantify the heat shock protein HSP)70 and HSP70 mRNA in Vero-E6 cells after infection with Hantann virus(HTNV).Methods The expres- sion of HSP70 and change of its mRNA level were detected by immunocytochemical staining,nucleic acid hybridization in situ and RT-PCR.Results In situ hybridization and RT-PCR were used to eval- uate the level of HSP70 mRNA during Hantaan 76-118 infection.HSP70 mRNA increased 0.5 h after infection,reached its peak by 12 h and gradually declined to steady state level by 72 h(vs.sham infec- ted group,P＜0.05).The expression of HSP70 protein induced by Hantaan 76-118 infection was e- valuated by quantitative immunocytochemical staining.HSP70 increased 0.5 h after infection,reached its peak by 12 h and decreased at 72 h after infection(vs.sham infected group,P＜0.05).Conclu- sions HSP70 can be induced directly by HTNV infection at both mRNA and protein levels,It pro- vides a basis for the further study of the pathogenesis,prevention and treatment of hemorrhagic fever with renal syndrome(HFRS).

ObjectiveTo compare the diagnostic efficiency of 18F- FDG coincidence SPECT/CT and 99Tcm- MDP whole body bone scan (WBBS) in detecting malignancy.MethodsA total of 71 cases (male 45,female 26,mean age 59.2 ± 15.4 years) with clinically confirmed malignancy underwent both 99TcmMDP WBBS and 18F-FDG coincidence imaging within three weeks.The sensitivity,specificity,accuracy,positive and negative prediction value of these two imaging methods in detecting bone metastases were compared based on the results from pathology or clinical follow-up.x2test was used for data analysis.ResultsA total of 350 lesions (including primary,second malignancy and benign disease) in 71 patients were eval-uated.81.7％ (286/350) malignant lesions were identified by either 99Tcm-MDP WBBS (209/350,59.7％ ) or 18F-FDG coincidence imaging ( 141/350,40.3％ ) (x2 =25.65,P ＜ 0.01 ).The imaging findings of osteoblastic,osteolytic,mixed types of bone metastases by99Tcm-MDP WBBS and 18F-FDG coincidence imaging were significantly different (x2 =20.78,2.89 and 9.94,all P ＜ 0.05 ).The sensitivity,specificity,accuracy,false-positive,false-negative,positive and negative predictive values for detecting bone metastases by 18 F-FDG coincidence study and 99Tcm-MDP WBBS were as follows:11.72％ ( 15/128),91.67％(22/24),24.34％ (37/152),8.33％ (2/24),88.28％ (113/128),88.24％ (15/17),16.30％ (22/135) ; and 53.91％ (69/128),75.00％ ( 18/24),57.24％ (87/152),25.00％ (6/24),46.09％ (59/128),92.00％ (69/75),23.38％ ( 18/77 ).The sensitivity,accuracy,false-negative,positive-predicting value of the two methods had been significant different (x2 =32.70- 46.21,all P ＜ 0.01 ).When two methods were combined,the diagnostic efficiency could been improved.ConclusionThe 99Tcm-MDP WBBS and 18F-FDG coincidence imaging has a complementary role in detecting bone metastases.

Objective To review and analyze the prenatal diagnosis and treatment of fetal bronchopulmonary sequestration (BPS).Methods Data of 68 BPS fetuses in the Department of Fetal Medicine,Guangdong Women and Children Hospital and Health Institute from January 20,2009 to January 31,2013 were reviewed.All of the 68 fetuses were diagnosed by ultrasound and volume to head circumference ratio (CVR) was measured.Postnatal surgeries at neonatal or infantile period were performed and babies were followed-up.Results With antenatal ultrasonography,fetal chest solid or hyper-echoic mass was found in all 68 cases at an average of 24 weeks of gestation (18-30 weeks) and were diagnosed as BPS after at least twice ultrasound scans.CVR was measured in these 68 cases,among them 56 cases (82.4％) were ＜1.6,and 12 cases (17.6％) were ≥1.6.Among the 12 cases with CVR≥1.6,seven developed fetal hydrops.Except for two terminated pregnancies because of CVR≥ 1.6 with fetal hydrops,all of the rest 66 cases continued their pregnancies till birth and 58 (87.9％) neonates were asymptomatic and eight (12.1％) were symptomatic.The mass was undetectable with ultrasonography in 18 fetuses during the third trimester,however,the mass was identifed again under CT scan after birth in 14 cases (another four cases unexamined).There were totally 45 cases fully recovered after lobectomy or BPS resection after birth,among which five cases underwent emergency surgery in neonatal period.Pathology results showed that 12 cases were intra-lobar and 33 cases were extra-lobar type.Totally,six cases were diagnosed as BPS under diaphragm.Sixty cases were followed up at three months to two years after birth,and all developed well and CT scan indicated normal pulmonary function.Conclusions Most BPS fetuses without hydrops have a good outcome.Fetus with BPS could wait for full term delivery following professional evaluation and standard protocol.However,the prognosis of BPS with fetal hydrops depends on prenatal evaluation and early surgical intervention.And the effect of early surgical intervention in BPS neonates is satisfied.CVR is the most important index for prenatal evaluation of BPS and CT scan after birth is critical for those babies with undetectable mass by prenatal ultrasound.

Objective To explore the treatment of hemangioma associated with thrombopenia(Kasabach-Merritte syndrome,KMS) in infant.Methods The clinical manifestation and the therapy of 14 cases patients with KMS in hospital from 2003 to 2006 were collected ang analyzed,the 400 g/L urea(manufactured by ourself)combined with methylprednisolone local injection were used and followed 0.5 to 1.0 year.Results Two in 14 cases were emergency exairesis,7 cases were recurred for several times,12 cases were cured,1 case was improved,1 case was loss the connection.Conclusions It is an ideal therapy method to use the urea combined with glucocorticoid to treat infancy KMS,little trauma,definite effective,low side effect,high cure rate.

Objective To study the effect mechanism of momordica charantia total saponin ( MCTS)′s ameliorating insulin resistance through discussing MCTS′s influence of protein kinase B ( Akt -2 ) and adenosine monophosphate activated protein kinase ( AMPK ) in type 2 diabetes ( T2DM ) rats.Methods T2DM models were built with the method of higher lipid and sugar diets and injection of a low dose of streptozotocin.Rats are divided into normal group, model group, control group and three dosage test groups.The positive control group and three dosage test groups rats were administrated with 2 mL metformin (50 mg· kg-1) and MCTS (100, 200, 400 mg· kg -1) per day for 8 weeks.Fasting serum glucose and insulin were detected with biochemical analyzer and ELISA method.The Akt-2 and AMPK′s mRNA and the activated protein were measured by RT-PCR and Western blot method.Results The homeostasis model assessment of insulin resistance ( HOMA-IR) reduced and the mRNA and protein expression amount of Akt-2 and AMPK increased obviously in all test groups compared to the model group.Conclusion The ameliorating insulin resistance mecha-nism of MCTS maybe caused by up-regulating the mRNA and protein of Akt-2 and AMPK.

Objective To evaluate the method of detecting antibodies to Bacillus anthracis by enzymelinked immunosorbent assay(ELISA)using crude antigen.Methods The anti-Bacillus anthracis antibody levels in sera of 42 healthy people and 42 patients were detected by indirect ELISA.Standard curve was plotted using the data from positive controls,based on which the relative content of each serum was calculated and compared with the result of rLF.Results The median of antibody's relative content in patient group and healthy people group are 1.19 and 0.24,the differences being statistically significant(uc=7.643,P＜0.05).The result of crude antigen is in concordance with rLF(but not parallel absolutely).Conclusions Crude antigen can distinguish most of patients with healthy population effectively.The results suggested that crude antigen is applicable in anti-Bacillus anthracis antibody surveillance.

This study was aimed to establish the method of identifying bupleurum cultivated germplasm using simple sequence repeat (SSR) molecular markers and to initially establish dataset of characteristic SSR bands to the bred cultivars or strains. From the bupleurum SSR primer pairs which were designed in previous work, 50 primer pairs were selected. Two bred strains and 4 other bupleurum cultivated germplasms were used as test materials. Primers pairs were screened with effective PCR amplification and high polymorphism. Meanwhile, conditions for PCR amplification and electrophoresis were optimized. Then, obtained SSR bands were analyzed and a clustering tree on the basis of genetic distance was constructed. The results showed that 9 SSR primer pairs can be used for identification. The suitable assay conditions were established and characteristic SSR bands were obtained for tested materials. The tested samples can be divided into 4 categories in the genetic similarity coefficient of 0.73. TheB. scorzonerifolium cultivated inHeilongjiang andChuanhongchaiNo. 1 strains were clustered as one category. ChuanbeichaiNo. 1 strain andZhongchai No. 1 cultivar clustered as another category. Cultivated germplasms fromSichuan Fengshunand Rongxian clustered as a unique category. It was concluded that the primer pairs and assay method established in the present study can be used as reference in identification of bupleurum cultivars or cultivated germplasms.

Objective To detect the global level of histone 4 lysine 20 (H4K20) methylation and its relation with DNA damage-repair in peripheral blood cell of arsenic-exposed residents in the coal-contaminated arsenism areas in Guizhou,in order to provide a basis to deepen the interpretation of the role of arsenic in inhibiting DNA damage-repair.Methods Jiaole village in coal-burning-borne arsenism areas in Xingren County of Guizhou was selected as the survey point,and 115 cases of arsenic-exposed residents were selected as the arsenic exposed group on the basis of physical examination.Moreover,53 residents from one village of non-epidemic area neighboring the diseased area were selected as controls.Hair and peripheral blood samples of these subjects were collected,and the histone protein was extracted from the lymphocytes separated from blood samples.The hair samples were digested with microwave digestion instrument,and the hair arsenic content was tested via the inductively coupled plasma-mass spectrometry (ICP-MS) method;the level of H4K20 1,2,3 methylation (H4K20me2,me2,me3) in peripheral blood cell was tested by enzyme-linked immunosorbent assay (ELISA);DNA damage of peripheral blood cell was measured by single cell gel electrophoresis (SCGE).Results The testing results of hair arsenic contents showed that the arsenic levels of hair in arsenic exposed group [0.30 (0.19-0.46)μg/g] were significantly higher than those of control group [0.12 (0.08-0.18) μg/g,F=11.968,P ＜ 0.05].Compared with the control (0.44 ± 0.14,0.99 ± 0.41,1.06 ± 0.33),the level of H4K20me1 (0.60 ± 0.29) in arsenic exposed group was higher (F =2.513,P ＜ 0.05),H4K20me2 (0.75 ± 0.26) was lower (F =4.707,P ＜ 0.05),and H4K20me3 (1.20 ± 0.62) was of no significant difference (F =0.582,P ＞ 0.05).The detecting results of DNA damage of the lymphocytes separated from peripheral blood showed a statistically significant increase (F =9.307,9.457,all P ＜ 0.05) in TailDNA％ and Olivetailmoment in arsenic exposed group [Median (M):10.75,11.69]compared with those of control group (M:2.12,1.16).The correlation analysis indicated that the arsenic levels of hair of subjects were positively correlated with H4K20me1 (r =0.214,P ＜ 0.05) and inversely with H4K20me2 (r =-0.224,P ＜ 0.05);H4K20me1 was positively associated with TailDNA％ (r =0.383,P ＜ 0.05) and Olivetailmoment (r =0.380,P ＜ 0.05);H4K20me2 was inversely associated with TailDNA％ (r =-0.290,P ＜ 0.05) and Olivetailmoment (r =-0.298,P ＜ 0.05).Conclusion H4K20 methylated modification makes a response to arsenic exposure of human body,the alteration of H4K20mel/me2 may participate in regulating DNA damage-repair induced by arsenic in vivo.
