1.Development of hepatitis virus C therapeutic vaccine with hepatitis virus B core antigen us the carrier

Objective To invesgate hepatitis B c antigen(HBeAg)as the carrier to construct mixed hepatitis C virus(HCV)therapeutic vaccine.Methods Fused the pTrc-core gene with two synthetic T-epitope antigen gene of HCV,expressed the plasmids pTrc-core-T1 and pTrc-core-T2, applied sucrose density gradient centrifugation to get the fusion protein HBcAg-T1 and HBcAg-T2, dialysised and concentrated the protein,mixed and immunized them in mice using the protein HBcAg (expressed by pTrc-core)as control.The tumor regression trial in mice was evaluated at appropriate time.After immunized four times,got the blood and spleen of mice.Interleukin(IL)-12 in serum were detected by enzyme-linked immunosorbent assay(ELISA),nonspecific T lymphocytes prolifera- tion response of splenic lymphocytes was respectively examined by thiazolyl blue(MTT)assay.T cell subset of splenic lymphoeytes,IL-5 in serum,IL-4 and interferon(IFN)-?in lymphocyte were evalu- ated by FACS.Results Tumor regression trial showed the experimental group formed only one tumor(diameter=0.1 cm),smaller than the T_1T_2 peptides group(diameter=0.9 cm)and blank group(diameter=1.3 cm).FACS indicated that CD8~+ T cell percentage of spleen cells from HBcAg- T_1T_2 group(20.21?2.01)% was higher than T_1T_2 peptides group(15.33?1.45)% and blank group(5.09?1.66)%,the percentage of IFN-?positive cells in these three groups were(1.58?0.05)%,(0.88?0.02)% and(0.53?0.03)%.The ELISA discovered that the level of IL-12 in the experimental group was the highest.Different from above,the IL-4 and IL-5 were lower in the exper- imental group.The detection of eytotoxic T lymphocyte(CTL)activity showed that the quantity of Hela cells infected by HCV in HBcAg-T1T2 stimulated group was different obviously from T1T2 peptides group. Conclusion The mixed protein HBcAg-T1 and HBcAg-T2 can induce stronger cellular immunity and it is able to serve as a therapeutic vaccines candidate specific for HCV.

4.Expression of hTERT in benign and malignant gastric diseases and its clinical significance

Objective: To investigate the expression of human telomerase reverse transcriptase (hTERT)in benign and malignant gastric diseases and its clinical significance. Methods: The expression of hTERT in 126 gastric tissue specimens obtained by surgery and endoscopy were studied by SP immunohistochemical method. The specimens included 10 cases of nomal mucosa,26 of chronic gastritis, 22 of benign ulcer,5 of leiomyoma, 3 of hyperplastic polyps, 50 of gastric cancer, 2 of maligmant lymphoma and 8 of sarcoma. In precancerous lesions,there was 54 cases of atypical hyperplasia,including 18 low grade, 25 of intermediate and 11 high grade. The positive expression rates and relations of clinical characters were analyzed. Results: hTERT was not or seldom expressed in normal and benign gastric diseases, while it was highly expressed in malignant lesions.There was a significant difference between them(P
5.Lignan constituents and activities of Linum usitatissimum L. above ground

Objective To investigate the chemical constituents and activities of Linum usitatissimum L.aboveground. Meth-ods The chemical constituents were separated through silica gel,ODS,Sephadex LH-20,and semi-preparative RP-HPLC chroma-tography and identified by optical rotation and spectroscopic analysis. All of the isolates were evaluated for their inhibitory activities by the luciferase assay. Results Eight dibenzylbutyrolactone lignans were separated from L. usitatissimum and identified as(-)-hinoki-nin(1),(-)-bursehernin(2),(-)-dimethylmatairesinol(3),(-)-yatein(4),(-)-thujaplicatin trimethyl ether(5),nemerosin (6),(+)-E-7,8-dehydromatairesinol dimethyl ether(7),and E-7,8-dehydrothujaplicatin trimethyl ether(8),respectively. Conclu-sion Compounds 7 and 8 were isolated from L. usitatissimum for the first time,and NMR spectral data of compound 8 were reported for the first time. Compounds 1 and 3 showed moderate inhibitory activities on IL-6/STAT3 signaling pathway with IC50 values of 42.12 and 43.43μmol/L,respectively.

8.The protective effect of rosiglitazone on renal injury of severe acute pancreatitis

ObjectiveTo investigate the effects of rosiglitazone, the agent of highly selective peroxisome proliferator-activated receptor-γ agonist, on the renal injury of rats with severe acute pancreatitis. MethodsFifty-four male Wistar rats were randomly (random number) divided into three groups : sham operation group ( SO group), severe acute pancreatitis group ( SAP group ) and rosiglitazone pretreatment group (ROSI group) . Severe acute pancreatitis model was induced by retrograde infusion of 5% sodium taurocholate into the biliopancreatic duct. Rosiglitazone (6 mg/kg) dissolved in 10% DMSO were injected into the femoral vein 30 minutes prior to the modeling. The solution of 10% DMSO was given to rats of SO group and SAP group. Rats were sacrificed 3, 12 and 24 h after modeling. The levels of serum amylase, serum creatinine, urea nitrogen, urinary albumin, urinary lgG and αl-microglobulin were measured and analyzed statistically. Kidney tissue samples were stained respectively with hematoxylin and eosin for histopathological evaluation.Results The levels of serum amylase, serum creatinine, urea nitrogen, urinary albumin, urinary IgG and αl-microglobulin were significantly increased (P < 0. 05 )after modeling, while lesser increases were found in ROSI group 12 h and 24 h after modeling (P <0. 05)compared with those in SAP group. ConclusionsRenal injury can be induced by severe acute pancreatitis,while Rosiglitazone protects rats from renal injury in the setting of severe acute panereatitis.
