1.Effect of paeonol on expression and function of drug transporters BCRP and SLCO1 B1 in HepG2 cell

Objective To investigate influences of paeonol on mRNA expression and function of drug transporters BCRP and SLCO1B1 in HepG2 cell.Methods Cell counting Kit-8 assay was used to detect the viability of HepG2 cells;Real-time fluorescent quantitative PCR (qPCR) was performed to measure the expressions of BCRP and SLCO1B1 mRNAs; flow cytometry was applied to determine the transport functions of BCRP and SLCO1B1. Results Paeonol (2-8μg/mL) did not decrease HepG2 cell survival rate, but 16 μg/mL paeonol significantly reduced cell survival rate (P<0.05). Paeonol(2-8μg/mL)significantly induced the mRNA expression and function of drug transporters BCRP and SLCO1B1(P <0.05).Compared with control group, transcription level of paeonol group’s BCRP and SLCO1B1 drug transporters obviously up-regulated, the of translocation efficiency of substrate specificity increased significantly (P <0.05).Conclusion Paeonol can induce drug hepatocellular transporters BCRP and SLCO1B1 gene expression, thereby promote the substrate transport the transmembrane.It is indicated that the drug combination of paeonol and BCRP and SLCO1B1 transporters, there may be a risk of drug interactions.

CXC chemokine ligand 4 (CXCL4) is a platelet-derived chemokine,and it belongs to CXC subfamily.Its receptor is CXC chemokine receptor 3 (CXCR3),which belongs to the seven transmebrane G-protein-coupled receptor family,and its coreceptor is glycosaminoglycan (GAG).CXCL4 and its receptors take part in the regulation of many biological processes,such as immune regulation,inflammatory response,endothelial cell proliferation and migration and angiogenesis.Recent studies show that CXCL4 and its receptor have significant influence on the cell growth,angiogenesis,invasion and metastasis of colorectal cancer,which offers a new perspective to explore the mechanism of colorectal cancer and its targeted therapy.
3.Therapeutic Observation of Acupuncture at Guangming (GB37) and Diwuhui (GB42) in Treating Dry Eye Due to Different Syndromes

Objective To observe the clinical efficacy of acupuncture at Guangming (GB37) and Diwuhui (GB42) in treating dry eye due to different syndromes. Method Thirty-nine dry eye patients (78 eyes) conforming to syndrome of up-flaming liver fire or liver-kidney yin deficiency were randomized into a distant group and a periocular group, to receive acupuncture at distant acupoints [Guangming (GB37) and Diwuhui (GB42)] and periocular acupoints [Cuanzhu (BL2), Tongziliao (GB1), and Taiyang (EX-HN5)], respectively. Result In the distant group, the total effective rate was 60.0% after 2-week acupuncture and 75.0% after 4-week acupuncture; in the periocular group, the total effective rate was 53.6% after 2-week acupuncture and 65.8% after 4-week acupuncture. There were no significant differences between the two groups (P>0.05). The tear production and tear film break-up time (BUT) were significantly improved in both groups after the intervention (P<0.05). The inter-group difference in the improvement of up-flaming liver fire syndrome score was statistically insignificant (P>0.05), while the inter-group difference in the improvement of liver-kidney yin deficiency syndrome score was statistically significant (P<0.05). Conclusion Acupuncture at Guangming and Diwuhui can boost the improvement of dry eye symptoms and tear production, and prolong BUT; dry eye due to up-flaming liver fire achieved a satisfactory therapeutic efficacy in both groups, while liver-kidney yin deficiency type responded better to acupuncture at distant acupoints.
4.Inhibition on Mast Cell Degranulation and Histamin Releasing of EtoH Ext. of Roasted Perilla Seed in Vitro

Objective To detect the inhibition of the EtOH ext. of roasted perilla seed on mast cell degranulation and histamin releasing in vitro. Methods Aliquots of OVA-sensitized rat peritoneal mast cells (PMC) were preincubated with 320, 640, 12 80 ?g/mL EtOH ext. of roasted perilla seed, disodium cromoglicate and luteolin, before an optimal concentration of OVA were added. Then the PMC degranulation response upon allergen triggering was detected by PMC degranulation percentage and histamine releasing assay. 6.Effect of maternal hypothyroidism on fetal glucose metabolism

[Summary] The theory of fetal origins of adult disease (FOAD) is now widely accepted by researchers who hold the opinion that adult degenerative and metabolism diseases have close relationship with the environment of fetal development inside and outside the womb. Some studies have proved that maternal hypothyroidism can negatively affect the glucose metabolism of their offsprings. However, the whole mechanism is not clear yet. Insufficient thyroid hormone during pregnancy was proved to slow down the formation of fetal pancreatic cytoskeleton, to decrease the proinsulin gene transcription, and to modulate series of cytokines and enzymes which are related to glucose dependent insulin secretion. Thyroid hormone receptor is also considered to be partially responsible for the relation between low thyroid hormone and β cell insufficiency. However, more studies in vivo should be carried out to prove this hypothesis. Epidemiologic studies have suggested that type 2 diabetes and low birth weight can be different phenotypes of the same genotype. The definite mechanism of maternal hypothyroidism in influencing fetal β-cell function should be studied by further investigation.
7.Effect of ultrasound-guided transverse abdominal plane block combined with dexmedetomidine on cesarean section

Objective:To investigate the effect of ultrasound-guided transverse abdominal plane block combined with dexmedetomidine on cesarean section.Methods:Eighty pregnant women undergoing elective cesarean section under general anesthesia in Binzhou Central Hospital from October 2017 to October 2019 were included in this study. They were randomly assigned to undergo either cesarean section under general anesthesia followed by patient- controlled intravenous analgesia (control group, n = 40) or ultrasound-guided transverse abdominal plane block combined with intravenous dexmedetomidine administration (observation group, n = 40). Analgesic efficacy and the analgesic effect on puerperas and their newborns were compared between the observation and control groups. Results:At 2, 4 and 12 hours after surgery, the Visual Analogue Scale score in the observation group was (2.1 ± 0.9) points, (2.2 ± 0.7) points, (3.0 ± 0.6) points, respectively, which was significantly lower than (4.4 ± 1.3) points, (4.9 ± 1.2) points, (3.9 ± 0.8) points in the control group, respectively ( t = 8.946, 7.854, 6.246, all P < 0.05). At 2, 4 and 12 hours after surgery, the comfort score in the observation group was (3.4 ± 0.8) points, (3.2 ± 0.7) points, (3.3 ± 0.7) points, respectively, which was significantly higher than (2.1 ± 0.7) points, (2.2 ± 0.9) points and (2.5 ± 0.8) points respectively in the control group ( t = 7.235, 6.130, 5.953, all P < 0.05). After surgery, the number of times the demand button of the anesthesia pump in the observation group was pressed was (8.4 ± 2.9), with the rate of patient satisfaction of 97.50% (39/40), which was significantly higher than that in the control group [13.1 ± 3.7, 77.50% (31/40), t = 7.036, χ2 = 9.347, both P < 0.05). At 1 and 2 days after surgery, the cumulative amount of anesthetics used for patient-controlled intravenous analgesia was (52.1 ± 1.9) mL and (121.7 ± 4.1) mL respectively, which were less than (63.6 ± 2.4) mL and (146.5 ± 5.3) mL in the control group, respectively ( t = 6.240 and 8.842, both P < 0.05). The score of extubation quality in the observation group was significantly lower than that in the control group [(1.3 ± 0.3) points vs. (3.2 ± 0.5) points, t = 6.664, P < 0.05]. There was statistically significant difference in cortisol content between the two groups at 10 minutes after extubation and at the time points of fetus removal, skin incision and intubation (all P < 0.05). In the observation group, the partial pressure of carbon dioxide in the umbilical cord blood was significantly reduced, the partial pressure of oxygen and the oxygenation index in the umbilical cord blood were significantly increased, and Apgar score at 1 minute after birth was significantly increased (all P < 0.05). Conclusion:Ultrasound-guided transverse abdominal plane block combined with dexmedetomidine can enhance the anesthetic effect on cesarean section, prolong analgesic time, reduce the amount of analgesics, improve the operation effect, reduce maternal stress responses, and improve maternal and infant health.
8.Determination of isofraxidin in Xiaoerkaiwei Oral Liquid by HPLC

Objective: To establish a method for determination of isofraxidin in Xiaoerkaiwei Oral Liquid (Radix Acanthopanacis Senticosi, Endothelium Corneum Gigeriae Galli, Semen Alpiniae Katsumadai, Semen Pharbitidis, etc.) by RP HPLC. Methods: The HPLC system consisted of Symmetry C 18 column(3.9m?150mm, 5?m), methanol water (0.1% formic acid) (30∶70) mixture as a mobile phase, detection wavelength at 342nm, 0.8mL?min -1 of flow rate and column temperature at 30℃.Results: The calibration curves of the isofraxidin were linear ( r =0.9993). The precision were perfect ( RSD =1.87). The samples were stable in 6 h. The average recovery of Xiaoerkaiwei Oral Liquid was 102.4% with RSD of 1.76%.Conclusion: This method is simple, quick and specific, suitable for the quality control.
9.Determination of Dracorhodin and Curcumin in Dieda Pills by UPLC

Objective:To determine the content of dracorhodin and curcumin in Dieda pills by UPLC .Methods:A UPLC method was adopted.The determination was performed on a Waters Acquity UPLC BEH C 18 column(100 mm ×2.1 mm,1.7 μm) with the mobile phase consisting of acetonitrile-0.05 mol· L-1 NaH2PO4(50 ∶50).The detection wavelength was 440 nm for dracorhodin and 431 nm for curcumin, the column temperature was 30℃and the flow rate was 0.1 ml· min-1 .Results:There was a good linear range of 0.001 8-0.036 4 μg(r=0.999 9)for dracorhodin and 0.000 8-0.015 6 μg(r=0.999 9) for curcumin.The average recovery for dracorhodin was 97.94%(RSD=0.89%) and that for curcumin was 98.45%(RSD=0.91%).Conclusion:The method is simple, rapid and reproducible ,which can be used for the determination of dracorhodin and curcumin in Dieda pills .
10.Study on binocular acuity and the effect on disability evaluation

Objective To research the relationship between binocular acuity and monocular acuity,and their effect on the evaluation of visual impairment and disability. Methods 1006 patients, aged between 18 and 60 years, are measured monocular and binocular acuities after the best lens correction. Results Binocular vision is higher than, equal to and less than monocular vision, three accounted for 27.14%, 70.78% and 70.78% respectively. Interocular difference has certain correlation with binocular interactions (χ2=54.939,P<0.05,r=0.228). Thirty-three patients have same binocular acuity but different monocular acuity. Conclusion Binocular acuity associated with the monocular acuity and the better eye vision plays a special role in visual disability evaluation.
