1.The clinical characteristics of colorectal cancer in Guangdong province

Objective To investigate the clinical characteristics of colorectal cancer and its trend in Guangdong province.Methods The data from 6501 patients with pathologically confirmed colorectal cancer of 5 hospitals from 1990 to 2004 was analyzed.Results The mean age of 6501 patients with colo- rectal cancer was 59 years old,with the predominant age ranged from 61 to 70 years old.The proportion of young patients decreased from 7.1% to 3.5%.and the ratio between male and female was 1.47:1, which was gradually increased with increasing of age.The prevelence reached a high peak(60 years old) and dropped after that as a single peak curve.The ratio between rectal cancer and colon cancer was 1.1: 1,and the descending colon cancer accounted for 27.6%.With ageing,the rectal cancers decreased while the right hemi colon cancer increased gradually.Tubular adenocarinoma was predominant with his- tological typing,which accounted for 76.0%.The progressing stage(Dukes B,C,and D)was found in 93.9% of all cases.Conclusions The onset age of patients with colorectal cancer become close to west- ern country.The proportion of young patients are decreasing.The ratio between male and female dis- plays a single peak curve from rising to dropping.A tendency of decreasing percentage in rectal cancer and the gradual increasing in right hemi colon cancer emphasize the importance of colonoscopy in colorec- tal caner screening.
2.Influence of type 2 diabetes mellitus on coronary artery lesion and plaque using optical coherence tomo- graphy in patients with coronary artery disease

Objective:To study influence of type 2 diabetes mellitus (T2DM) on coronary artery lesion and plaque sta‐bility using optical coherence tomography (OCT) in patients with coronary artery disease (CAD) manifesting unsta‐ble angina pectoris (UAP) . Methods :A total of 200 patients diagnosed as CAD manifesting UAP were selected.According to complicated with diabetes mellitus or not ,they were divided into normal blood glucose group (n=98) and complicated T2DM group (T2DM group ,n=102). Coronary angiography (CAG) and OCT were used to eval‐uate coronary artery lesion ,assess plaque nature and measure the thickness of plaque fibrous cap in order to confirm plaque stability . Results:Compared with normal blood glucose group ,there were significant rise in percentages of multi-vessel coronary disease (48.98% vs. 78.43% ) ,severe artery stenosis (20.57% vs. 40.21% ) ,lipid plaques (25.00% vs. 39.84% ) and plaques with fibrous cap thickness <65μm (25.93% vs. 45.24% ) in T2DM group , P<0.05 or < 0.01 .Conclusion :Patients with coronary artery disease have more severe coronary artery lesion and high ‐ er incidence rate of multi‐vessel coronary disease and unstable plaque when complicated with type 2 diabetes mellitus .
3.Investigation and analysis in medication compliance of patients with ankylosing spondylitis

Objective To investigate the current status of medication compliance of patients with anky-losing spondylifis and analyze its related factors. Methods Seventy-four patients with ankylosing spondylitis were investigated about their general conditions,medication compliance and its related factors.The results were analyzed. Results Thirty-nine patients(52.7%) had good medication compliance while the rest 35 cases has poor medication compliance among which 24 patients with drug withdrawal exceeded 1 month.The medication compliance for patients with disease process of lower than 0.5 year,0.5 to 1.0 year,1 to 2 years and above 2 years was 82.0%,60.0%,25.0%and 31.0%.The logistic regression analysis showed that the disease process,dis-ease knowledge and media advertisement were the main influencing factor for medication compliance,while the age,sex,marital status,education background,adverse reaction,satisfaction degree with medication effect, economic conditions and depression were not major influencing factors. Conclusions About half of patients with anky-losing spondylitis had poor medication compliance,especialy with disease process of above 1 year.The main in-fluencing factors include disease process,disease knowledge and media advertisement.
4.Effects of 5-Aza-CdR on cell apoptosis in lung cancer A549/DDP cell line and expression of tumor suppressor gene hMLH1

Objective To investigate the effects of 5-Aza-2'-deoxycytidine (5-Aza-CdR) on the apoptosis of A549/DDP cells and the expression of hMLH1 gene.Methods A549/DDP cells were treated with 5-Aza-CdR at 0.5,5,50 μmol/L.The growth curve of A549/DDP cells was investigated by MTT assay.The methylation status of hMLH1 gene was detected by methylation specific PCR (MSP).The expression of hMLH1 mRNA was evaluated by FQ-PCR.The apoptosis rate of A549/DDP cells was analyzed by flow cytometry.Results A549/DDP cells treated with 5-Aza-CdR showed a slow growth in comparison with the control cells,and the growth rates were decreased with the increasing of 5-Aza-CdR concentration.The apoptosis rate after treatment was higher than that before treatment in A549/DDP cells (P < 0.05),and had a positive correlation with 5-Aza-CdR dose (P < 0.001).hMLH1 mRNA expression level was increased in a 5-Aza-CdR concentration dependent manner (P < 0.05).hMLH1 promoter in A549/DDP cells was methylated and hMLH1 mRNA was negatively expressed before treatment,but the mRNA was positively expressed after treatment with 5-Aza-CdR.Conclusions 5-Aza-2'-CdR can induce apoptosis of A549/DDP cells by inducing demethylation of hMLH1 promoter and thereby enhancing hMLH1 gene expression and its tumor suppressor function.
5.Experimental study on acute hypertensive cerebral edema at 7.0T MR

Objective To verify the feasibility of a rat model of acute hypertension, and to observe the imaging findings at 7.0T MR scanner of the model.Methods In experimental group, the rats were scanned at 7.0T Bruker MR scanner when phenylephrine was injected continuously to evoke an acute hypertension state.The real time blood pressure was monitored through a femoral arterial catheter connected to a pressure transducer.The control group was infused with saline.T2WI, DWI and T1 mapping were performed in the two rat groups.ADC maps and T1 maps were acquired after image post-processing, and a voxel wise analysis and a ROI analysis were applied.The brain morphology change was evaluated by HE staining.The blood-brain barrier permeability was evaluated by Evans blue staining.Results The T2 images and DWI images of the experiment group showed no abnormal signal intensity changes observed by naked eyes.But ROI analysis of the ADC maps showed that the brain ADC values of the experiment group was higher than the control group (t=3.291,P<0.01, the cortex;t=2.186,P<0.05, the subcortex).Voxel wise analysis of the ADC maps showed vasogenic edema mainly distributed in the parietal and occipital regions,subcortical nuclei (caudate nucleus),thalamus,brainstem and cerebellum,which mainly located in the posterior portions of cerebral hemispheres.T1 image analysis did not show any significant difference between two groups (P>0.05).The blue dye region was only performed in the experimental group,the results of HE staining corresponded with vasogenic brain edema.Conclusion Continuous infusing of phenylephrine hydrochloride can induce a rat model of acute hypertension.Vasogenic edema and blood-brain barrier permeability change can be observed in the rat model, and the imaging distribution of vasogenic edema can be detected by MRI.
6.Relationship of plasma homocysteine level and pathological change of large artery in stroke patients

Objective To determine the association between homocysteine and severity of large cerebral artery atherosclerosis.Methods Total 152 ischemic stroke patients were evaluated using transcranial Doppler(TCD),magnetic resonance angiography(MRA) or digital substraction angiography(DSA).Severity of cerebral artery atherosclerosis was scaled by number of stenosed or occluded artery.Results There were 83 patients in low level homocysteine group with mean Hcy level(15.31?3.08)?mol/L,while 69 patients in high level homocysteine group with mean Hcy(34.47?21.38)?mol/L.Number of abnormal intra-and extra-artery was 1.86?1.51 and 1.52?1.46 respectively.No significant difference was demonstrated in the number of abnormal artery between two groups.No correlation was noted between Hcy and number of abnormal artery.Conclusion There is no relationship between high homocysteine and severity of large cerebral artery atherosclerosis in ischemic stroke patients.Homocysteine might be the marker of the disease.

Cancer cell exfoliation,invasion and entry into circulation system is the early event with metastasis,which provide the possibility to formation of clinical metastase.Further research about the circulating cancer cells can help us to understand the mechanism of metastasis and offer the scientific proof against anti-metastasis.The detection of circulating tumor cells and clinical significance were reviewed.

9.The relationship between plasma triglycerides, cholesterol levels and breast cancer

Objective To investigate the relationship between plasma triglycerides ( TG ) , cholesterol (TC)levels and breast cancer.Methods 105 cases of breast cancer(age from 22 to 69 years)and 261 cases of healthy control(age from 16 to 52 years)admitted from May.2014 to Oct.2014 were studied.TG and TC were tested.Subgroup analysis was performed by age (<35, 35-45, ≥45 years) and BMI( <18.5, 18.5-24, ≥24).Results TG level in breast cancer group was significantly higher than that of the healthy control group (t=-3.97,P=0.00), and TC level was lower than that in the healthy control group (t=2.49, P=0.01).TG lev-el of breast cancer patients≥45 years old was higher than that in the healthy control group ( t=-4.31, P=0.00).TC level of breast cancer patients <35 years and from 35 to 45 years was lower than that of the healthy control group(t=3.12, P=0.00及t=2.41, P=0.02); TG levels in breast cancer group were significantly lower than those of the healthy control group for all BMI subgroup ( t=-3.67, P=0.01 for BMI<18.5; t=-3.87, P=0.00 for BMI from 18.5 to 24.0;t=-2.01, P=0.04 for BMI≥24).TC level of breast cancer patients was higher than that of the healthy control group for patients whose BMI <18.5(t=-3.41, P=0.01) and it was lower than that of the healthy control group for patients whose BMI≥24.Conclusions The relation-ship between TG , TC and breast cancer is associated with age .Compared with healthy group , breast cancer pa-tients≥45 years have a higher TG level and breast cancer patients <45 years have a lower TC level .
10.Effect of genistein on the angiogenesis-related factors secreted by fibroblast-like synoviocytes from rats with collagen Ⅱ induced arthritis

Objective To investigate the effect of genistein (gen) on the secretion of vascular endo-thelial growth factor (VEGF) and matrix metalloproteinases (MMPs-1, 2, 3, 9) by fibroblast-like synoviocytes (FLS) from rats with collagen Ⅱ induced arthritis (CIA). Methods The CIA was induced by collagen Ⅱ and complete Freund's adjuvant (CFA) into rats. The rats were scored based on the arthritis index(AI) once a week. At the sixth week, the X-ray of joints was taken. The synovial tissues from knee joints were examined pathologically. The primary fibroblast-like synowocytes were separated from the synovial tissue by collagenase digestion and cultured. Then the expression of VCAM-1 was estimated by flow cytometry. After adding gen (100, 200, 400 μmol/L) at different concentrations into the FLS, VEGF and MMP-1, 2, 3, and 9 of the supernatants were tested by indirect ELISA. Results After 3 days of type Ⅱ collagen and CFA injec-tion, the rats started to catch arthrocele and their arthritis index increased gradually. The arthrocele was most remarkable at the 3rd week. The AI, X-ray and pathological examination indicated that the model group were significantly different from the control group. After the synoviocytes were cultured to the 4th generation, the expression of VCAM-1 was as high as about 85.5%. It showed that most synoviocytes were changed to fibro-blast-like synoviocytes. Different concentrations of gen (100, 200, 400 μmol/L) added to FLS were compared and revealed that the VEGF and MMP-1, 2, 3, and 9 in the supematants were suppressed evidently and in a dose-dependent manner. Conclusion The CIA model can be successfully constructed by collagen type Ⅱ and CFA. Tthe primary FLS of rats' joint can be separated and cultured well by collagenase digestion. Certain levels of gen can suppress the secretion of VEGF and MMP-1, 2, 3 and 9 hy FLS. The affect is dose-depen-dent.
