Objective To research the reversal the effect on multidrug resistance (MDR) and the effect on Ca~(2+) concentration of HL60/HT cell by psoralen, and observe its possible mechanism. Methods The cytotoxic effect of chemotherapeutic agents was determined by MTT assay, intracellular harringtonine (HT) content was assayed by HPLC, and intracellular Ca 2+ concentration in different periods and at different time points was detected by laser confocal microscope. Results Psoralen could reduce IC_ 50 value of HT to HL60/HT cells and increase HT content in cells in different degrees at the doses of 1-20 ?mol/L. Psoralen could influence Ca 2+ concentration with negative correlation at different time points (24, 48, and 96 h) at the doses of 1-20 ?mol/L, and influence Ca 2+ concentration with negative correlation at different times (5, 10, 15, 20, and 25 min) in the doses of 10 and 20 ?mol/L. Conclusion Psoralen can reverse MDR of HL60/HT cells, its reversal mechanism is associated with influence of intracellular Ca~(2+) concentration and the increase of intracellular accumulation of HT.

Microbiology 1992;0(06):-
N-carbamoylase is a part of hydantoinase operon which can transform N-carbamoylamino acid to corresponding ammo acids. The L-N-carbamoylase of Arthrobacter BT801, codied by the HyuC gene, is the rate-limiting and the only stereoselective enzyme. HyuC DNA fragment was amplified by PCR from the plasmid of pUC18-169. The target fragment was introduced into pPIC3. 5K plasmid to construct the pPIC3. 5K-hyuC expressing vector which was then transduced into Pichia pastoris GS115 cells after being linearized by BglⅡ digestion. Multi-copies insertion transformants were screened on G418 plates. The recombinant protein was proved to have biological activity of hydrolyzing N-carbamoylphenylalanine into phenylalanine through enzyme activity determination.

Objective:To study the use of digital subtraction angiography (DSA) guided transnasal ileus tube placement in management of abdominal compartment syndrome (ACS) after liver transplantation. Methods:From January 2015 to December 2019, a total of 30 patients who developed ACS after liver transplantation who were admitted to the Transplantation Intensive Care Unit of Tianjin First Central Hospital were retrospectively studied. According to the way of decompression, these patients were divided into the study group and the control group. Patients in the control group were treated with conventional abdominal decompression, while patients in the study group were treated with DSA guided transnasal ileus tube placement based on management principles developed in conventional abdominal decompression. Changes in intra-abdominal pressure, treatment efficacy rates and liver functions were monitored in the two groups up to 7 days after abdominal decompression. Results:There were 23 males and 7 females, aged (53.4±11.6) years. After treatment, the IAP, portal venous blood flow velocity, bile drainage volume, ALT and AST in the study group were significantly better when compared with the findings before treatment: [IAP: (7.13±3.87) vs (22.73±2.09) mmHg, portal vein blood flow velocity: (34.76±10.31) vs (21.45±6.47) cm/s, bile drainage volume: (198.43±19.94) vs (80.72±9.52) ml/d, ALT: (158.92±67.56) vs (278.73±99.17) U/L, AST: (79.36±15.63) vs (196.71±89.05) U/L], ( P<0.05). After treatment, when compared with the control group, the IAP, portal vein blood flow velocity, bile drainage and TBil in the study group were significantly better [IAP: (7.13±3.87) vs (13.47±6.19) mmHg, portal vein blood flow velocity: (34.76±10.31) vs (24.98±8.54) cm/s, bile drainage: (198.43±19.94) vs (108.73±21.30) ml/d, TBil: (258.85±91.95) vs (343.69±89.45) μmol/L], ( P<0.05). In the control group, the IAP significantly decreased on the fourth day after treatment, ( P<0.05); compared with the significant difference in the study group on the second day after treatment ( P<0.05). After 7 days of treatment, the efficacy rate of the control group was 46.7% (7/15), compared to 86.7% (13/15) in the study group. The difference between the two groups was significant (χ 2=5.400, P<0.05). Conclusion:DSA guided transnasal ileus tube placement for treatment of abdominal compartment syndrome after liver transplantation resulted in a better treatment efficacy rate than conventional treatment.

OBJECTIVETo investigate the significance of cytokine interleukin-12p40 (IL-12p40) and interferon-gamma (IFN-gamma) in tissues formation and development of human oral lichen planus (OLP).
METHODSThe tissues of 11 cases of normal oral epithelium and 43 cases of OLP were investigated for the expression of IL-12p40 and IFN-gamma proteins by using Envision two-step immunohistochemistry. The correlations between the expressions of these two cytokines, and their clinical and pathological significance in OLP were analyzed.
RESULTS1) IL-12p40 and IFN-gamma proteins were up-regulated in OLP comparing with that in normal oral mucosa and there was statistical significance between their difference (P < 0.05). 2) The percentage of positive IL-12p40 staining in OLP of IFN-gamma positive group was higher than IFN-gamma negative group and there was statistical significance between their difference (Chi2 = 5.828, P = 0.016). A positive correlation was found between IL-12p40 and IFN-gamma proteins in OLP (Spearman r = 0.357, P = 0.019). 3) The percentage of positive IL-12p40 staining in OLP with short course (< 6 months) was higher than that in OLP with long course (> 6 months; Chi2 = 7.935, P = 0.005), and a significant association was found between IFN-gamma over expression and the degeneration of base cells in OLP lesions (Chi2 = 9.070, P = 0.011).
CONCLUSIONThese results indicate that at the primary phase of OLP, IL-12 may drive the pathological destruction in OLP lesions by elevating IFN-gamma protein locally. IFN-gamma may play an important role for the pathological destruction in OLP lesions.

BACKGROUNDPatients with severe full-thickness burn injury suffer from their inability to maintain body temperature through perspiration because the complete destructed sweat glands can not be regenerated. Bone marrow-derived mesenchymal stem cells (BM-MSCs) represent an ideal stem-cell source for cell therapy because of their easy purification and multipotency. In this study, we attempted to induce human BM-MSCs to differentiate into sweat gland cells for sweat gland regeneration through ectodysplasin (EDA) gene transfection.
METHODSThe dynamic expression of EDA and EDA receptor (EDAR) were firstly observed in the sweat gland formation during embryological development. After transfection with EDA expression vector, human BM-MSCs were transplanted into the injured areas of burn animal models. The regeneration of sweat glands was identified by perspiration test and immunohistochemical analysis.
RESULTSEndogenous expression of EDA and EDAR correlated with sweat gland development in human fetal skin. After EDA transfection, BM-MSC acquired a sweat-gland-cell phenotype, evidenced by their expression of sweat gland markers by flow cytometry analysis. Immunohistochemical staining revealed a markedly contribution of EDA-transfected BM-MSCs to the regeneration of sweat glands in the scalded paws. Positive rate for perspiration test for the paws treated with EDA-transfected BM-MSCs was significantly higher than those treated with BM-MSCs or EDA expression vector (P < 0.05).
CONCLUSIONSOur results confirmed the important role of EDA in the development of sweat gland. BM-MSCs transfected with EDA significantly improved the sweat-gland regeneration. This study suggests the potential application of EDA-modified MSCs for the repair and regeneration of injured skin and its appendages.
