The short stature homeobox-containing(SHOX) gene,located in the short-arm pseudoautosomal region (PAR1) of the sex chromosomes,is one of the recently discovered genes,which is related to short stature.Its encoded protein,as a transcription activator,plays an important role in the regulation of growth.It has now been confirmed that the human SHOX gene mutation can cause L?ri-Weill syndrome,Turner syndrome,idiopathic short stature growth and its related characteristic skeletal deformities.This review makes a summary about SHOX gene defects,its clinical phenotype and treatment.

3.Current study on diagnostic values of various inflammatory cytokines in postoperative infection related to orthopedic implants

In recent years,along with social economic development and population aging,more and more fracture patients have received internal fixation and artificial joint replacement.Postoperative infection related to orthopedic implants is a catastrophic complication which imposes serious psychological,physiological and economic burdens on the patients.Therefore,it is necessary to be able to make an early diagnosis of the infection.However,diagnosis of implant-related infection is always a clinical problem for orthopedists.Currently,it is thought to be a valuable way to judge orthopedic implant infection by monitoring inflammatory cytokines.This article will offer an overview on the progress in research of diagnosis values of different inflammatory cytokines in postoperative infection related to orthopedic implants.
4.Expression of transient receptor potential M8 in human knee joint cartilage

BACKGROUND:Transient receptor potential protein channel is distributed in many kinds of tissue and cells in human body, and takes part in many physiologic functions and pathological changes of human body. However, up to now, transient receptor potential melastatin 8 (TRPM8) has not been reported in human cartilage.
OBJECTIVE:To study the expression of TRPM8, a subfamily member of transient receptor potential gene, in human knee joint cartilage.
METHODS:Fifteen samples were selected from patients after amputatioin due to severe trauma. Total protein was extracted from 300 mg cartilage tissue of every patient. To detect the distribution of TRPM8 in human
cartilage, cartilage sections were col ected in the same region. The expression and distribution of TRPM8 in human cartilage was detected by western blot analysis and immunofluorescence respectively.
RESULTS AND CONCLUSION:Western blot analysis showed that specific TRPM8 band was visible at molecular weight of 130 000, and as the age increased, TRPM8 expression in human cartilage was gradual y decreased. Immunofluorescence staining results showed that TRPM8 was evenly located in the cellmembrane of chondrocytes. TRPM8 is expressed in the human knee cartilage, specifical y in member of chondrocytes.
5.The relationship between air pollutants and mortality in Huairou District

Objective:
To evaluate the relationship between air pollutants and mortality of residents in Huairou District, Beijing, providing a basis for the formulation of air pollution control measures.
Methods :
The data of daily deaths, meteorological factors and air pollutants in Huairou District from 2014 to 2018 were collected from Beijing Disease Prevention Monitoring Information Integration and Analysis System, Huairou Meteorological Bureau and Environmental Monitoring Station. The generalized additive models were used to analyze the relationship between the average daily concentration of air pollutants and the daily deaths.
Results:
The medians of daily average concentrations of SO2, NO2, CO, O3, PM10 and PM2.5 were 5.00 μg/m3, 24.00 μg/m3, 0.71 mg/m3, 77.27 μg/m3, 64.25 μg/m3 and 44.13 μg/m3, respectively. Except for O3, the daily average concentrations of SO2, NO2, CO, PM10 and PM2.5 showed decreasing trends from 2014 to 2018. An increase of 10 μg/m3 of NO2 resulted in an elevation of 1.69% ( 95%CI: 0.31%-3.08% ) , 3.31% ( 95%CI: 1.24%-5.42% ) and 3.31% ( 95%CI: 0.51%-6.19% ) for non-accidental death in the whole population, females and people under 65 years old, respectively, with a delay of 2 days (lag2). For every 10 μg/m3 increase in the daily average concentrations of CO and PM2.5, the risk of non-accidental death among people under 65 years old at lag2 increased by 0.08% ( 95%CI: 0.01%-0.14% ) and 0.88% ( 95%CI: 0.12%-1.64% ) , respectively. For every 10 μg/m3 increase in daily average concentration of O3, there was 0.69% ( 95%CI: 0.02%-1.36% ) increase in daily male non-accidental death risk at lag4. The results of the multi-pollutant model showed that after adjusting the effects of the other two air pollutants, NO2, CO and PM2.5 had no statistically significant effects on the daily non-accidental deaths of people under 65 years old at lag2 ( P>0.05 ) .
Conclusion
The ambient NO2, CO, O3 and PM2.5 pollution increase daily non-accidental deaths, which shows a lag effect.
6.Clinical efficacy of Qizhengxiaotong plaster plus Xianling Gubao for osteoarthritis of knee

ObjectiveTo compare and observe the clinical effect of Qizhengxiaotong plaster and Votalin emulsion in curing osteoarthritis of knee (KOA) for outpatient treatment.MethodsIn 70 cases,the KOA patients are randomly divided into treatment group and control group,respectively given Qizhengxiaotong plaster plus Xianling Gubao and Votalin emulsion for one month.To observe the clinical effects of patients respectively pre and post treatment.ResultsAfter a course of treatment,Qizhengxiaotong plaster plus Xianling Gubao could improve the clinical symptoms and signs significantly.Its improvement for pain was the same as Votalin.But Qizhengxiaotong plaster plus Xianling Gubao was significantly better than Votalin in eliminating the swollen,tenderness and dysfunction.And there was no obvious difference between Qizhengxiaotong plaster plus Xianling Gubao and Votalin emulsion in total effective rate,evaluated by WOMAC Osteoarthritis Index Guide and VRS-5.The typical symptoms were improved after the therapeutic method.No adverse reaction was found during the treatment.ConclusionQizhengxiaotong plaster plus Xianling Gubao had obvious effect on the osteoarthritis of knee and could improve the patients' pain and limitation of activity.Hence,it was worthy to be generalized in the conservative treatment of the osteoarthritis of knee for out-patients.

9.Effect of lentivirus-mediated RNA interference silencing IL-1 type Ⅰ receptor on signal transduction in a rat model of osteoarthritis

Objective To observe the effects of lentivirus-mediated RNA interference silencing IL-1 type Ⅰ receptor on the expression of mitogen-activated protein kinase (MAPK) in a rat model of osteoarthritis (OA), and to explore the role of IL-1 and MAPK in OA. Methods Forty SD rats were randomly divided into four groups(A-D, each group: n = 10). The rats (A-C group) were underwent unilateral anterior cruciate ligament transaction and partial excision of the medial meniscus. At day 7 and 9 after operation, the rats of group A were respectively received 40 μL of lentivirus-mediated RNAi silencing IL-1 type Ⅰ receptor by intra-articular injection, while the rats of group B were received lentivirus-mediated RNAi non-silencing IL-1 type Ⅰ receptor, and the rats of group C were received saline. The rats of group D were taken for normal control. All rats were sacrificed four weeks after the surgery. The knees were harvested to observe macropathologic changes of the joint cartilage. ERK1/2, JNK1/2 and p38 proteins in the cartilage were detected by Western blot. Results Cartilage degradation in group A was milder than that in group B and C (P<0.05),but was worse compared to group D (P<0.05). Level of p38 expression in cartilage in group A was lower than that in group B and C (P<0. 05), but had no significant difference compared to group D (P>0.05). Levels of ERK1/2 and JNK1/2 expressions in cartilage in group A were lower than that in group B and C(P<0.05),but was significantly increased compared to group D(P<0.05). Conclusions Lentivirus-mediated RNAi silencing IL-1 type Ⅰ receptor can inhibit the expression of MAPK, in particular,p38 protein was strongly inhibited.
10.Metformin regulate the balance of Th17/regulatory T cells and expression of serum cytokines through AMPK-mTOR signaling pathway in collagen-induced arthritis

Objective To evaluate therapeutic effects of metformin on the expression of serum cytokines,balance of splenic Th17/regulatory T cells (Treg) and expression of splenic AMPK-mTOR in collagen-induced arthritis (CIA) rats,and the mechanism thereof.Methods The rat model of CIA was established by injecting bovine type Ⅱ collagen.Forty rats were randomly divided into five groups:the CIA model group(sterile water by gavage),Met-30 mg/kg group (metformin 30 mg ·kg-1 ·d-1 by gavage),Met100 mg/kg group(metformin 100 mg ·kg-1 ·d-1 by gavage),Met-300 mg/kg group(metformin 300 mg ·kg-1 ·d-1 by gavage) and control group (sterile water by gavage).The hind paw volume was recorded once a week.The serum level of cytokines,tumor necrosis factor (TNF)-α,interleukin (IL)-1β,IL-6,and IL-17,were measured by enzyme linked immunosorbent assay (ELISA) 35 days after the initial immunization.The quantity of Th17 and Treg cells were examined by flow cytometry.The expression of AMPK,p-AMPK,mTOR and p-mTOR were examined by western blotting.One-way analysis of variance was used to evaluate the experimental data.Results The values of hind paw volume were decreased on the 35 d of the initial immunization in the Met100 mg/kg [(2.43±0.37) ml,t=2.97,P<0.05] and Met-300 mg/kg groups [(2.58±0.21) ml,t=2.96,P<0.05] than those of CIA model group (2.97±0.23) ml.The serum levels of TNF-α,IL-1β,IL-6 and IL-17 on the 35-d after the initial immunization were significantly lower in the metformin therapeutic groups [Met-300 mg/kg group TNF-α (104±8) pg/ml,t=42.77,P<0.05;IL-1β (183±24) pg/ml,t=60.457,P<0.05;IL-6 (19.3±2.8) pg/ml,t=53.62,P<0.05;IL-17 (64.5±6.7) pg/ml,t=47.92,P<0.05] than those of the CIA model group [TNF-α (246±8) pg/ml;IL-1β (1 336±40) pg/ml;IL-6 (87.0±5.1) pg/ml;IL-17 (282.3±6.8) pg/ml].The quantity of Th17 and Treg cells were significantly different in the Met-300 mg/kg group than those of the CIA model group 35 d after the initial immunization [Th17(6.57±0.39) vs (9.89±0.53),t=8.74,P<0.05;Treg (7.60±0.45) vs (3.94±0.61),t =8.37,P<0.05].The expression of p-AMPK on the 35 d after the initial immunization in the metformin therapeutic groups was significantly higher than in the CIA model group(P<0.05),and the expression of p-mTOR was significantly lower (P<0.05).Conclusion Metformin can significantly inhibit the serum levels of TNF-α,IL-1β,IL-6 and IL-17 in CIA model rats,and regulate the balance of Th17/Treg through AMPK-mTOR signaling pathway.
