1.Effects of ginkgo biloba extract 761 on contraction of gastrocnemius of fixed hindlimb in rats

Objective To study the preventive and therapeutic effects of ginkgo biloba extract 761 on disused skeletal muscle atrophy by observing its effects on the wet weight, protein content and contraction of the gastrocnemius of fixed hindlimb in rats. Methods 24 SD rats were divided into blank group, contrast group and experimental group. Their right hind limbs were fixed for 4 weeks with plastic plaster from metatarsus to 1 cm up the knee. The knee joint was fixed in 100? flexion and the ankle joint in 60? plantar flexion. Rats in the contrast group were perfused with 3mL distilled water, and rats in the experimental group were perfused with ginkgo biloba extract 761 (120 mg/kg, 14 mg/mL). The whole gastrocnemius muscle of the right hindlimb was dissected 4 weeks later to measure its contract tension, wet weight and protein content. Results Compared with the blank group, single maximal contraction, maximal tetanus, wet weight and protein content of the gastrocnemius muscle in the contrast and experimental groups decreased significantly (P
2.Biomechanical changes of proximal femur for posteriomedial fractures:a finite element analysis

[Objective]To research the biomechanical influence of posteriomedial fractures on proximal femur.[Method]Three dimensional finite element models of intact and osteotomied proximal femurs were established from computerized tomographic(CT)scanning images,and finite element analysis was undertaken.[Result]The maximum equivalent stress was located at the anteriomedial of the defection of the proximal femur.The stress of femoral neck,as well as the medial and lateral femoral cortex was not significantly changed after isolated lesser trochanteric fracture.However,with half of the medial cortex involved,the stress of the medial and lateral femur increased by 47% and 12%,respectively.If the entire medial cortex was involved,the stress of medial and lateral femur increased by 247% and 66%,respectively.[Conclusion]Different types of posteriomedial fragments have different effects on the stress distribution and quantity of proximal femur.When treating intertrochanteric fractures using dynamical hip screw,the fragments should be differently treated based on its types considering for biomechanical effects.
3.Long-term effects of articular cartilage derived scaffold/autologous chondrocytes composites for repairing full-thickness defects of knee articular cartilage in rabbits

[Objective]To observe the histological characteristics of autologous chondrocytes/human articular cartilage derived scaffold (HACDS) composites for repairing chondral defects in rabbit models. [Methods]A full-thickness articular-cartilage defect (diameter 4 mm,thickness 2 mm) was created in the femoral condyle of rabbits. The rabbits were divided into two groups: control group,HACDS scaffold only,and experimental group,chondrocytes/HACDS scaffold. Fifteen months after implantation,the specimens were observed by inverted phase contrast microscopy,and assessed by staining with haematoxylin-eosin,alcian blue,toluidine blue,safranin O,as well as by the immunohistochemistry of collagen type Ⅱ.[Results]Histologically,the generated neo-cartilage integrated well with its surrounding normal cartilage and subchondral bone in the defects of experimental group at 15 months post-implantation,whereas only fibrous tissue was filled in the defects of control group. Histochemical and immunohistochemical analysis revealed that alcian blue,toluidine blue,safranin,and were obviously positive in the experimental groups. However,alcian blue,toluidine blue,and safranino O were negative in the control group.[Conclusion]The current histological examination demonstrated that an engineered cartilage composed of autologous chondrocytes on HACDS scaffold could be successfully obtained and further applied to repair an articular cartilage defect in a rabbit model.
4.Studies on the distribution of interstitial Cajal-like cells in ampulla region and the myoelectric activities of the sphincter of Oddi of guinea-pigs

Objective To investigate the distribution of interstitial Cajal-like cells(ICLC) in ampulla and the myoelectric activities of the sphincter of Oddi(SO) of adult guinea-pigs.Methods C-Kit immunohistochemical staining was carried out on whole-mount preparations of guinea-pigs' ampulla region.The myoelectric activities of SO were recorded by using the contact electrode on mucosa.Result C-Kit-positive ICLC were observed in both exterior and inner wall of ampulla.Some of these cells were distributed in the circular and longitudinal smooth muscle cell(SMC) layers assuming a spindle-like shape.Another sub-group of ICLC,which were located between the longitudinal and circular SMC layers,were shown to have oval or triangle cell bodies with projection of more than three long processes.These cells interlaced into a cellular network,closely resembling the interstitial cells of Cajal(ICCs),around the myenteric nervous plexus(ICC-MY) in the duodenum.At the inner side of circular SMC layer of the exterior wall,ICLC were similar to ICCs associated with the deep muscular plexus(ICC-DMP) of small intestine.In the inner wall of the ampulla,a large amount of ICLC were found in the SMC layers or bundles,running parallel with them.At the opening of the ampulla,a number of ICLC surrounded the opening to form a ring.The spontaneous rhythmic myoelectric activities could be recorded in the SO.Conclusion It is suggested that the ICLC might contribute to the regulation of the spontaneous rhythmic contraction,and it might be related to development of motility disorders of the SO.
5.Treatment of Thoracic-lumbar Vertebra Tuberculosis by Focus Eliminate Through Anterior Approach in First Intention and Fixation by Bone Transplantation and Interfixation of Z-plate

Objective To discuss the significance and effect of tuberculosis of thoracic-lumbar vertebra treated by interfixation with Z-plate in anterior approach. Methods Summarize was made in 32 cases of patient with tuberculosis of thoracic-lumbar vertebra from January 2000 to June 2004 ,All were treated by focus eliminate through anterior approach in first intention, autobody bone transplantation inter vertebra and interfixation with Z-plate in anterior approach. Results Followed up for a average of 25 months. 32 cases with tuberculosis of vertebra were cure, whole transplantation bones were bone fusion ,the fusion time was a average of 5 months. rectification angle of back protruding was 17.50,no surgery complication of hemothorax,pheumothorax, aggravation of paraplegia, leak of cerebrospinal fluid,looseness of interfixation and rupture . Conclusion Treatment of tuberculosis of thoracic-lumbar vertebra by focus eliminate through anterior approach in first intention, fixation by bone transplantation and interfixation of Z-plate have importance significance and marked effect.
6.The predictive value of five factors to the maternal difficulty airway

Objective To research the most commonly used five method to evaluated the difficulty airway , and compare which methods were more suited for the pregnant woman in general anesthesia. Methods 214 patients with full-term pregnancy who requested emergency or elective caesarean-section were assigned. During the pre-anesthetic visit,we evaluated patients from Mallampati score, thyromental distance, body mass index (BMI), inter-incisor gap, and upper lip bite test. After endotracheal intubation ,patients were divided into 2 groups based on Cormack classification. Results Five ways sensitivity descending order were upper lip bite test (79.5%)、Mallampati score (76.9%)、BMI (56.4%)、inter-incisor gap (51.3%)、thyromental distance (35.9%); specificity descending order were upper lip bite test (93.1%)、Mallampati scores (86.3%)、inter-incisor gap (85.1%)、thyromental distance (76.6%)、BMI (62.3%). Conclusions In pregnant women ,Mallampati score and the upper lip bite test are the better indicators to predict difficult airway.
7.Development of time-resolved immunofluorometric assays for Dickkopf-1 and application on sera of patients with lung cancer

Objective To develop a highly sensitive and accurate time-resolved immunofluorometric assay (TR-IFMA) for measurement of Dickkopf-1(DKK-1),as a novel serologic biomarker for lung cancer. Methods The study constructed a two-manoclonal-antibody "sandwich"-type assay and the sensitivity, within run CV and between run CV and accuracy were evaluated. Serum DKK-1 concentrations were measured by TR-IFMA in 120 healthy controls, in 72 benign lung disease patients, and in 212 lung cancer patients before surgery. The association between serum DKK-1 levels and clinicopathological features were evaluated. Results A standard curve for DKK-1 TR-IFMA had been developed with good sensitivity (0.08 μg/L). Both within run CV and between run CV were less than 6.5%. Accuracy studies, parallelism and precision data were determined and all found to be satisfactory. The validity of DKK-1 assay was confirmed by the good correlation between the results obtained by TR-IFMA and commercial ELISA (r=0.972, P=0.01). The serum levels of DKK-1 were higher in lung cancer patients 31.93(79.47-18.03) μg/L than in benign lung diseases 15.25(18.41-11.49) μg/L and in healthy controls 13. 90( 16. 91-11.02) μg/L DKK-1 levels were significant associated with the presence of distant metastases, as well as lymph node metastases and TNM stage, but not with patients' age, gender and tumor histology. At the cutoff of 22.63 μg/L, the diagnostic sensitivity, specificity, accuracy, positive predictive value and negative predictive value of the TR-IFMA for lung carcinoma were 68.4%, 92.2%, 82.1% ,90.6% and 72.5%. Diagnostic sensitivity and accuracy were higher for small cell carcinoma than for non-small cell carcinoma (70.7% vs 69.5% and 85.6% vs 80.7%, respectively). Conclusions A highly sensitive and reliable TR-IFMA for DKK-1 has been developed. The determination of serum DKK-1 levels may be useful for diagnosis and tumor staging of lung cancer.

9.The clinical analysis of diabetic peripheral neuropathy

Objective To investigate the clinical characteristics of diabetes complicated with peripheral neu- ropathy and increase the cognition of diabetic peripheral neuropathy in clinical work.Methods The clinical data of 160 diabetic patients were analyzed with retrospective analysis.Results 118 diabetic patients had different levels neuroelectrophysiologic abnormality,and the incidence rate was 73.8 %.The cure rate of diabetic peripheral neuropa- thy was 11.0%.Conclusion The incidence of peripheral neuropathy is high in diabetic patients,neuroelec- trophysiologic examination is a sensitive index in diagnosis of diabetic peripheral neuropathy,and the therapeutic ef- fect of diabetic peripheral neuropathy is poor.
10.Effects of hepatocyte growth factors on contractile function of disused gastrocnemius atrophy in rats

Objective To explore the possible effects of hepatocyte growth factors (HGF) on prevention and therapy of disused gastrocnemius atrophy in rats by observing their effects on the wet weight and contractile function of the gastrocnemius of fixed hind limbs. Methods Twenty-four Wister male rats were randomly divided into a blank group, a contrast group and an experimental group. Their right hind limbs were fixed for four weeks with plastic piaster from metatarsus to 1 cm up the knee. Their knee joint was fixed in 100?flexion and their ankle joint in 60?plantar flexion. Rats in the contrast group were injected with 0. 2 mL of normal saline, and rats in the ex- perimental group were injected with HGF (10 mg/kg, 10 mg/mL). Their whole gastrocnemius of the right hind limb was dissected four weeks later to measure its contractile function and wet weight. Results Compared with the blank group, single maximal contraction, maximal tetanus, and wet weight of the gastrocnemius in the contrast and experimental groups decreased significantly (P<0. 05), and fields of electric stimulation also decreased signifi- cantly (P<0. 05). Compared with the contrast group, single maximal contraction, maximal tetanus, and wet weight of the gastrocnemius in the experimental group were significantly superior (P<0.05), and fields of electric stimulation significantly increased. Conclusion Since after a local injection of HGF, the contractile function and wet weight can be significantly increased, HGF may have a preventive effect on disused atrophy.
