1.S100B and acute ischemic cerebral injury
International Journal of Cerebrovascular Diseases 2011;19(6):466-470
S100B is a class of acid calcium-binding protein. It mainly exists in neuroglial cells in the central nervous system, and it is one of the signs of astrocyte activation. S100B may reflect the function of neuroglial cells and regulate the complex interactions between the neurons and neuroglial cells. Studies in recent years have demonstrated that S100B protein is involved in the pathological processes of acute ischemic brain injury. Serum S100B protein can be used as an important marker for identifying the severity of cerebral infarction, progress and outcome.
2.Influences of basic fibroblast growth factor and epidermal growth factor on the proliferation and differentiation of neural stem cells
Chinese Journal of Tissue Engineering Research 2007;0(07):-
OBJECTIVE: How to promote the generation of neural stem cells (NSCs) and induce them differentiate into the goal cell type has been the focus of neuroscience. In this article, we review the adjustment of basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) to the proliferation and differentiation of NSCs. DATA SOURCES: A computer-based online search of Medline database was undertaken to identify the related articles published in English from January 1998 to March 2006 by using the keywords of "bFGF, EGF, nerve stem cells". At the same time, Chinese relevant articles were searched in China National Knowledge Infrastruction published between January 2000 and March 2006 with the same keywords in Chinese. STUDY SELECTION: The data were selected firstly to look over the citations of every literature. Inclusive criteria: Articles related to the effect of bFGF and EGF on the proliferation and differentiation of NSCs. Exclusive criteria: Duplicate research or Meta analysis. DATA EXTRACTION: A total of 78 related articles were collected, of which 32 accorded with the inclusive criteria and 46 with duplicate or old contents literatures were eliminated. Among the 32 literatures, 8 involved in the research status of NSCs, 10 about the effect of bFGF on the proliferation and differentiation of NSCs and the mechanism, 4 involved in the effect of EGF on the proliferation and differentiation of NSCs and the mechanism, respectively, 8 related to the effect of bFGF and EGF together, and 5 related to the comparison of the effect of bFGF and EGF. DATA SYNTHESIS: ①NSCs are self-renewed, and can differentiate into different stem cells and mature tissue cells in different microenvironment. Many cytokines in the microenvironment play essential roles in the differentiation direction of NSCs. ②bFGF has strong improvement effect on the proliferation of NSCs and can activate the potential regeneration capacity of neuronal precursor cell in distinct areas of central nervous system, proliferation and differentiation of cells present bFGF concentration dependent. ③EGF can improve the growth of NSCs, and promote them differentiate into neuron and glial cells; its effect of facilitating cell proliferation also presents concentration dependent. ④To elevate the efficacy of proliferation and differentiation of NSCs, allied application of bFGF and EGF is usual in NSCs culture. ⑤There is not a unified cognition to the effect difference of bFGF and EGF. CONCLUSION: bFGF and EGF have powerful effect to improve the proliferation and differentiation of NSCs; therefore, both of them are important neural somatomedin. Moreover, their effects present concentration dependent.

Objective To evalute the feasibility of a double hand allogroft to reconstuct the limbs'function. Methods A brain dead donor was chosen under the test of blood group in ABO and Rh,HLA match,PRA and the mixed leukocyte culture. The level of the transplantated hands was 5 cm proximal to the wrists The procedure of the allotransplantation was similar to that of the autologous replantation. The immunosuppressants were taken two days before the operation,in the operation and after the operation. The circulation, the vital signs and the rejection were under the supervision. Results The progress was satisfactory. No rejection was found postoperation. After 10 weeks the callus in both ends of radius and ulna was obvious. After 5 months sensation recovered. After 7 months the internal fixation(plates and screws) was removed and tenolysis was done. Now the recipient could use engrafted hands to wash face, make the bed, wear clothes, brush teeth, peel oranges, eat with a spoon, tie the shoelace slowly, pick up a telephoneor and use the conroller of Television. Conclusions With the immunosuppressive agents, the double hands allograft can survive. The growth rate of bone and nerve is faster than that of autologous replantation.

10.Preparation of a three-dimentional acellular cartilage matrix scaffold and its characteristics
Orthopedic Journal of China 2006;0(09):-
[Objective]To prepare a cartilage acellular matrix scaffold and to explore its feasibility in cartilage tissue engineering. [Methods]Microparticles about 100 ?m~154 ?m were prepared after calf cartilage physically shattered and experienced gradient centrifugation, and then treated by a modified Courtman's four-step method which was improved to produce acellular cartilage matrix. After this treatment the microparticles were made into 3% suspension which was placed into moulds. With the freeze-drying method,3-D cartilage acellular matrix (CACM) was prepared. The scaffolds were cross-linked by a neotype crosslinking agent genepin for 48h,and then placed into glycine solution server times for removing redundant genepin. The freeze-drying method was used to prepare CACM.The scaffolds were investigated by gross observation, histological staining (haematoxylin-eosin,toluidine blue) ,scanning electron microscope (SEM) observation and porosity measurement, water absorption rate and degradation rate analysis. After being cultivated for ten days, bone marrow stranal cells (BMSCs) of rabbit were seeded into the scaffold.MTT test and SEM were done to assess the growth and proliferation of BMSCs.[Results]Gross observation showed the scaffolds had a loosely porous and dark blue appearance after being cross-linked by genepin. The histological staining (haematoxylin-eosin,toluidine blue staining) showed that there were no chondrocyte fragments in the scaffold. The CACM scaffold had 90% porosity,(1314?337) % water absorption rate, and (13.69?7.3)% or (25.99?8.9) % degradation rate at 2 or 4 weeks.MTT test showed that BMSCs grew well in the 3-D CACM scaffolds of logarithmic trend, supporting that the scaffolds had no cytotoxic effect on BMSCs.SEM micrographs indicated that the scaffolds were porous and the cells covered the scaffolds firmly with cell processes.[Conclusion]The improved Courtman's four-step method makes a more thoroughly acellular scaffold. The 3-D CACM scaffold retains most of extracelluar matrix. After being cross-linked by genepin, the 3-D CACM scaffold has good biocompatibility and degradation rate of the scaffolds is decreased, which makes it a suitable carrier for cartilage tissue engineering.
