Objective: It was reported that major subclasses of anti-golmerular basement membrane (GBM) antibody were IgG1 and IgG4. The IgG1 subclass was mainly found in male patients and IgG4 subclass in female patients with anti-GBM disease. This study investigates the distribution of anti-GBM IgG subclasses and their association with clinical characteristics. Methods: Sera from 50 patients diagnosed as anti-GBM disease during 1991-2003 were collected. The anti-GBM antibodies with IgG1, IgG2, IgG3, and IgG4 subclasses were detected with purified bovine ?(IV)NC1 as solid phase ligand in ELISA. The association with clinical manifestations was further investigated. Results: The positive rates of IgG anti-GBM subclasses were IgG1 94%, IgG2 56%, IgG3 12%, IgG4 88%, respectively. IgG1 and IgG4 were the main subclasses in anti-GBM IgG subclasses (P

Objective:To observe effects of wrist-ankle acupuncture (WAA) onβ-endorphin (EP), nitric oxide (NO) in uterus tissue and prostaglandin F2α (PGF2α), substance P (SP) in serum of rats with primary dysmenorrhea. Methods:A total of 45 non-pregnant Wistar rats were randomly divided into a control group, a model group and a WAA group, 15 rats in each group. Rats in the model group and the WAA group received continuous abdominal subcutaneous injection of Diethylstilbestrol to establish dysmenorrhea rat models. On the first day after modeling, rats in the WAA group began to receive acupuncture on Point Lower 1 and Point Lower 2, once a day for 10 d. The control group and the model group didn’t receive any treatment. Writhing latencies and frequencies were recorded.β-EP and NO in uterus tissue homogenates and PGF2α, SP in serum were detected. Results:In the model group,β-EP and NO levels were the lowest among the groups, the serum PGF2α level was the highest, and serum SP level was the lowest. These measurements showed significantly difference between the model group and the control group (P<0.05). PGF2α in the WAA group was lower than that in the model group;β-EP, NO and SP levels were higher than those in the model group, with inter-group statistically significant differences (P<0.05). Conclusion: WAA may achieve analgesic effect through decreasing PGF2α, increasingβ-EP, NO and SP to relieve uterine cramps, increase blood flow and promote functional improvement.

BACKGROUND: Previous studies have proved that stress can affect immunity function and disturb homeostasis of bodies. Moderate exercise can alleviate the psychological stress reaction to some degree.OBJECTIVE: To observe the effects of moderate exercise on lymphocyte number in spleen and mesenteric lymph nod,interferon-y (IFN-γ) and interleukine-4(IL-4) level in serum, as well as Th1/Th2 balance in rats under chronic psychological stress.DESIGN, TIME AND SETTING: A randomized control animal experiment was carried out in the laboratory of Sports Human Science in Chengdu Sports University from May to October in 2007.MATERIALS: A total of 40 healthy male SD rats were divided by random digits table into 4 groups: control group, psychological stress group, exercise group and exercise+psychological stress group, with 10 ones in each group.METHODS: Before experiment, rats in exercise group and exercise+psychological stress group underwent 3 days of adaptive swimming training, 30 min/(t·d). During experiment, they were made swimming with no weight loading for 8 weeks, 60 min/(t·d).From day 2 post exercise on, rats in exercise+psychological stress group began to undergo the 21 days of chronic restraint stress,6 hours per day. At the same time, the chronic restraint stress models were established in the psychological stress group. Rats in the exercise group were fed for 22 days since the 8~(th) day of exercise ended. Rats in the control group were only fed without any intervention.MAIN OUTCOME MEASURES: The lymphocyte number in spleen and mesenteric lymph node was measured, and ELISA was employed to measure IFN-y and IL-4 levels as well as the ratio of IFN-y/IL-4 in serum.RESULTS: Compared with the control group, the lymphocyte number, the level of IFN-y and the ratio of IFN-y/IL-4 were significantly decreased in psychological stress group (P < 0.01 ), and the Thlf'rh2 balance was broken. Compared with the psychological group, the exercise+psychological stress group had got significantly higher lymphocyte number, IFN-γ level and ratio of IFN-γ/IL-4 (P<0.05). The lymphocyte number in spleen and mesenteric lymph node in the exercise group showed not a difference of significance compared with the control group, but a tendency towards going up. There was no difference of significance in IFN-γ level and ratio of IFN-γ/IL-4 among the exercise+psychological stress group, the exercise group and the control group.CONCLUSION: Moderate exercise can increase lymphocyte number in spleen and mesenteric lymph node significantly,up-regulate the IFN-γ level so as to regulate the disbalance of Th1/Th2, and maintain immunity homeostasis.

The efficacy of colorectal cancer treatment is not very satisfactory.Conventional morphologic imagings are limited in the diagnosis and in the applications of multimodality treatment for colorectal cancer.18 F-fluorodeoxyglucose PET-CT can play a pivotal role in detecting metabolism changes and in the localization of lesions concurrently. It is valuable in the assessment of responses to therapy, prediction of prognosis and selecting modality management and monitoring postoperative recurrence. The right-moment and proper application of PET-CT can increase the therapeutic efficacy of colorectal cancer.
9.Bacterial Protein Secretion Pathway with SecA as a Motor

Microbiology 2008;0(07):-
There are one third of synthesized proteins must be secreted to the cell surface or to the surrounding environment to acquire their native functional state. Most of them are exported by Sec translocase (secretion pathway). Sec translocase consists of a membrane embedded protein-conducting channel, termed SecYEG and a peripherally associated motor domain, the ATPase SecA. The SecDFyajC heterotrimeric membrane protein complex can facilitates protein translocation. SecB is a molecular chaperone that functions in the protein translocation pathway. SecM (secretion monitor) encoded by the 5' region of the secM-secA mRNA, which elongation arrest is required for upregulated expression of SecA. The signal sequence in the N terminus of the nascent peptide is first recognized by the signal recognition particle (SRP). SecB, the Sec-system-specific chaperone, channels the preprotein to the Sec translocation pathway and, ad- ditionally, actively targets the bound precursor to the translocase by its ability to bind SecA. The preprotein-bearing SecA then binds to the membrane, at a high-affinity SecA-binding site, SecYEG, which constitutes a channel for polypeptide movement. Continued translocation requires cycles of ATP hydrolysis bySecA, which is thought to occur in a step-wise fashion with a step of 20~30 amino acid residues.
