1.Determination of baicalin and hesperidin in Muxiangliqi Pills by HPLC

AIM: To establish the determination of baicalin and hesperidin in Muxiangliqi Pills (Radix Saussureae seu Vladimiriae, Pericarpium Citri Reticulatae Viride, Pericarpium Citri Reticulatae, Rhizoma Cyperi, etc.). METHODS: The determination was carried out by HPLC with octodelyl silicohydrides silanes bonded silica gel as a packing material, methanol water glacial acetic acid (35∶61∶4) as a mobile phase, detection wavelength at 283nm, flow rate at 1mL?min -1 . RESULTS: Hesperidin showed a good linear relationship at the range of 0.072?g ~0.360?g. Baicalin showed a good linear relationship at the range of 0.104?g~0.520?g. The average recoveries were 97.0% and 97.8%, respectively. RSD were 1.93% and 1.69%, respectively. CONCLUSION: The method is sensitive, accurate, reproducible, specific and can be used for quality control of Muxiangliqi Pills.

Herpes simplex keratitis (HSK),which is caused by infection of herpes simplex virus,is a kind of disease with high incidence and blinding rate.The mechanism of HSK is complex and always attracts the attention of researchers.It is considered as an immune response to viral infection and is related to several factors,such as pathway of infection and immunoregulation.This review reveals the latest advance of the mechanism of HSK in the aspects of viral infection and latency,apoptosis,immunity.
3.Impact of Mild Asphyxia in Delivery on Motor Development

Objective To explore the impact of intrapartum mild asphyxia on motor development in infants. Methods 30 infants with any of the following were as asphyxia group: heart rate more than 160 beats/min in the process of childbirth, and then down to 70-80 beats/ min, no cyanose; Apgar score 1 min more than 8. Other 30 normal infants were as control group. They were assessed with Neuropsychological Development Scale for 0-6 Years Old 3, 6 and 9 months after birth. Results Opisthotonos was more frequent in the asphyxia group than in the control group (P<0.01) 3 months after birth, but there was no significant difference in total score (P>0.05). Sitting forward was more frequent in the asphyxia group 6 months after birth, and the score of motor was less in the asphyxia group than in the control group (P< 0.05). More of the asphyxia group couldn't stand with help, and with poor motor score 9 months after birth (P<0.05). Conclusion Intrapartum mild asphyxia may infulence the motor development of infant in long-term.

6.Studies of the Effects of Hydrogen Peroxide on the Ultrastructure and Permeability of Candida Albicans

The Effects of hydrogen peroxide(H2o2) on the ultrastructure, cellular pea-meability barrier and 3H-TdR-DNA leakage of C. albicans were studied. Damage of the cell, especially leakage of cytoplasm were found under transmission electron microscope. Morphological changes and small hole like structures were also found under scanning electron microscope. Radioactivity assay of the leakage of 3H-TdR-DNA leakage of C. albicans further suggested that the 3H-TdR-DNA leakage of C. albicans was directly due to the damage of cellular permeability barrier. Negative correlation (r=+0.8392--0.9915)between the extent of 3H-TdR-DNA leakage and the number of surviving colony-forming units was found. This indicated that the damage of cellular permeability barrier and leakage of DNA of C. albicans caused by H2o2 was one of the most importent causes of cell killing.
7.Effect of laparoscopy and laparotomy surgery on immune function related indicators in ovarian benign tumor patients

Objective To explore the effect of laparoscopy and laparotomy surgery on immune function related indicators in ovarian benign tumor patients. Methods From January 2015 to January 2016, 86 cases of ovarian benign tumor patients were set as study objects. All the patients were randomly divided into control group and observation group, 43 cases in each. Patients in control group were given traditional laparotomy surgery, while patients in observation group were given laparoscopic surgery. The changes of immune function related indexes before and after operation were recorded, before operation, day 1 and day 3 postoperative, number of peripheral white blood cells (WBC), neutrophil ratio (NGR), immunoglobulin (IgA, IgG, IgM) levels, C reactive protein (CRP) content and the distribution of T lymphocyte subsets (CD3+, CD4+ and CD3+/CD4+ ratio) distribution were respectively determined, and the incidence of postoperative complications was recorded, and were further statistical analyzed. Results There was no significant difference in immunoglobulin IgA, IgG and IgM between the twogroups before operation (P > 0.05). Postoperative, only the IgG content of the two groups was significantly lower than that before the operation (P < 0.05 ); The levels of WBC, NGR and CRP in the control group patients in the昀rst day and the third day after surgery were significantly increased compared with preoperative period, while CRP in the observation group was higher than that before the operation, there was significant difference between the two groups (P < 0.05). There was no significant difference in CD3+, CD4+ and CD3+/CD4+ ratios between the two groups before operation (P > 0.05), the difference between the two groups was significant (P < 0.05). The complication rate of the observation group after operation was 6.98%, the control group was 16.28%. Conclusion Clinical use of laparoscopic treatment of benign ovarian tumors relative to the traditional laparotomy can effectively reduce the immune system of patients with immune function, with high safety, contribute to the recovery of patients, it is worthy of clinical promoting.
8.Application of induced hypothermia in vasoplegia syndrome after postcardiopulmonary bypass

Objective To investigate the effects of induced hypothermia in vasoplegia syndrome after post-cardiopulmonary bypass.Methods 4 individuals diagnosed as CPB-induced vasoplegia syndrome was given hypothermia by surface cooling using hypo-hyperthermia blanket based on vasopressors and adjustment of blood volume.The surface temperature,heart rate,systolic pressure,mean arterial blood pressure,central venous pressure,oxygen saturation of blood,urine volume,systemic vascular resistance (SVR),pulmonary vascular resistance (PVR) and cardiac output (CO) were recorded.Results Systemic vascular resistance gradually improved along with normalization of cardiac output,and increased mean arterial blood pressure.Conclusions The mild hypothermia can increase blood pressure,decrease dosage of vasopressors,and may play a decisive role in some cases.So mild hypothermia may be an effec-tive alternative to treat CPB-induced vasoplegia syndrome.
9.Optimization on formulation of sinomenine hydrochloride pulsatile drug delivery tablets by central composite design-response surface method

Objective To optimize the formulation of sinomenine hydrochloride pulsatile drug delivery tablets by central composite design-response surface method.Methods The tablets containing sinomenine hydrochloride were prepared by dry-compression coating technique.The influence factors included the amount of sodium carboxymethyl starch in core tablets, the ratio of HPMC/carrrageenan,and the amount of matrix materials in coating film.The evaluation parameter was lag time.Experiments were done on the central composite design,and the data were simulated using multi-linear equation and second-order polynomial equation.The possibly optimal formulation was predicted by response surface method.The lag time of the tablets prepared under the optimum condition was compared with the predicted.Results The lag time was simulated using second-order polynomial equation and the regression coefficient was 0.993 7.The lag time in vitro of the tablets prepared under the optimum conditions was about 6 h,then drug released in pulsatilerelease character.Bias between the observed and predicted values of the lag time was within ?4.43%.Conclusion The sinomenine hydrochloride pulsatile tablets could release drug quickly in vitro at the predetermined time.Central composite design-response surface method can be used to optimize the formulation and the model developed in this study proves to be highly predictable.
10.Study on the mechanism of themolecular biology of hepatocyte growth factor and connective tissue factor in proliferative vitreoretinopathy

Objective To determine the role of CTGF in PVR, we successfully down - regulated the expression of CTGF by RNA interference( RNAi) technology in ARPE-19 cell line. Methods Design and synthesis three CTGF siRNA, screen effective targeted points in Western blot, combine Oligo DNA of effective targeted point, anneal to form double chain DNA, connect with linearing pGC-LV-GFP carrier, sequence DNA in PCR screening positive clones. Results The shRNA-expression vector of CTGF was constructed successfully. And RT-PCR and Western Blot results showed that CTGF expression was significantly inhibited by siRNA transfectants in ARPE-19 cells at mRNA and protein levels. Conclusion We successfully construct shRNA-expression vector of CTGF which paves a way for CTGF-targeted gene therapy of proliferative vitreoretinopathy.
