Chronic kidney disease(CKD) is a major global health issue that leads to end-stage renal disease which untreated.The use of stem cell therapy provides a new perspective in this area.Reviewing the experimental studies of stem cell therapy on CKD,many different,and even contradictory reports in this area were found.In this paper,the recent reports,and present several key issues of experimental stem cell treatment on CKD were reviewed,including the source of stem cell,the sort of experimental animal,the time of treatment and other experimental details.Hoping these may lead more understanding in this area.
5.Clinical value and optimal diagnostic point of ultrasound in the diagnosis of thyroid lesions

Objective To discuss the combined value of gray-scale ultrasound ( GSUS) and contrast-enhanced ultrasound (CEUS) in the diagnosis of benign and malignant thyroid lesions and to explore the optimal diagnostic point of scoring method .Methods Ultrasound images of 178 thyroid lesions confirmed by pathology were synthetically reviewed by scoring 5 GSUS indicators including shape , orientation, interior echogenicity, halo sign, microcalcification and 6 CEUS indicators including relative arrival time of microbubbles in the periphery and interior, peak periphery and interior echogenicity, peripheral ring-enhancement, and homogeneity of enhancement .One positive indicator scored one point .The optimal diagnostic points and their clinical value were explored according to ROC curves .Results Scores of GSUS, CEUS and the combination of GSUS and CEUS were significantly different (Z =10.188,9.843,10.705,all P <0.001). Areas under ROC curves of GSUS, CEUS and the combination of GSUS and CEUS were 0.936, 0.919 and 0.964, respectively.Three or more positive GSUS indicators of five in a thyroid lesion predicted that the thyroid lesion was malignant , with the sensitivity of 79.6% and the specificity of 91.2%.Two or more positive CEUS indicators of six in a thyroid lesion predicted that the thyroid lesion was malignant , with the sensitivity of 91.8% and the specificity of 81.2%.Five or more positive GSUS and CEUS indicators of eleven in a thyroid lesion predicted that the thyroid lesion was malignant , with the sensitivity of 93.6%and the specificity of 92.3%.The areas under the ROC curve of GSUS and CEUS were 0.936 and 0.919.The area under the ROC curve of the combination of GSUS and CEUS was 0.964, larger than the areas under the GSUS ROC curve and the CEUS ROC curve.Conclusion Ultrasound is valuable in the differential diagnosis of benign and malignant thyroid lesions , and the combination of GSUS and CEUS is the most valuable with 5 points as the optimal diagnostic scoring method .
6.Nosocomial Infection in Hemodialysis Patients:Clinical Analysis and Preventive Measure

OBJECTIVE To study the situation of nosocomial infection of maintenance hemodialysis of chronic renal failure patients and discuss the risk factors and preventive measures.METHODS Retrospective analysis of our hospital clinical data in hemodialysis patients with nosocomial infection from Jan 2005 to Dec 2006 was taken.RESULTS There were 44 nosocomial infection cases from 89 cases.The infection rate was 49.43%.Hemodialysis patients were high-risk population of the nosocomial infection.The common infection site was the respiratory tract(52.27%),followed by central venous catheter-related infections(25.00%),urinary tract infections(15.91%) and intestinal infections(6.82%).The primary diseases of diabetic nephropathy,serious anemia,low albumin and elderly were the risk factors(P
7.Numerical computation on the scaffolds models with regular square holes using nonlinear fluid-solid-coupling approaches

Objective The influencing parameters of solid and fluid computing fields for the scaffolds models with regular square holes were discussed by nonlinear fluid-solid-coupling approaches.The numerical computational resuits of which the models were regarded as both rigid body and non-linear elasticity were compared as well.Method One direct fluid-solid-coupling approach and two indirect fluid-solid-coupling approaches were adopted,and the calculating reliability of three kinds of fluid-solid coupling methods was verified.Rasults The solid-fluidcoupling computational results are obtained in light of 12 kinds of scaffolds models which are constructed by 3 groups of square side length(50,100 and 150 μm)and 4 groups of porosity(61%,65%,77%and 84%).The field parameters of those solid models including stress,strain and displacement and those fluid models including static pressure,velocity,wall shear stress and strain rate are achieved and compared.Conclusion There appear some difference between the results of porous scaffold models as a rigid body and as non-linear elasticity.The different porosity with the same pore radius or the different pore radius with the same porosity would affect the field parameters of solid models and fluid models in varying degrees.

9.Clinical features of non-Hodgkin's lymphoma in cheek misdiagnosed as inflammatory myopathy

Objective To assess the clinical features of non-Hodgkin's lymphoma in cheek misdiagnosed as inflammatory myopathy.Methods Six patients presenting with unilateral swollen cheek were admitted at the Department of General Internal Medicine,Peking Union Medical College Hospital between January 2010 and December 2014.These patients were misdiagnosed as inflammatory myopathy and finally confirmed as Non-Hodgkin's lymphoma.The clinical characteristics,findings of laboratory tests and the diagnostic procedures were retrospectively analyzed.Results The median age of 6 patients (3 males and 3 females) was 30 years.The median duration from onset of unilateral swollen cheek to the final diagnosis was 13 months;the patients were referred to average 4 hospitals and experienced average 3 biopsies before admission.All patients had been treated with prednisone at a dose of 1 mg/kg or 0.5mg/kg per day.However,the symptoms relapsed in all patients after glucocorticoid tapering.No patient presented with proximal muscle weakness or myalgia.Two of 6 patients presented with unilateral forearm swollen.Creatine kinase was mildly elevated in 3 patients.Electromyographic abnormalities were seen in 4 patients.The biopsy showed muscle fiber necrosis,degeneration and a large amount of lymphocytes infiltration with or without vessel wall involvement.Erythrocyte sedimentation rate and high-sensitive C-reactive protein levels were normal.Anti-Jo-1,ANA and anti-U1RNP were all negative.EBV-DNV was significantly elevated in peripheral blood in 5 patients and it was positive in biopsy specimen in 3 patients.PET-CT scan was performed in 3 patients and suggested high metabolic activity with a SUV of 2.0-8.5 in cheek region.The biopsy specimen taken before admission did not show evidence of lymphoma.Repeated biopsies from cheek regions (3/6),lips (2/6) and forearm confirmed the final diagnosis as Non-Hodgkin's lymphoma,including 2 cases of T-cell lymphoma and 4 cases of NK/T cell lymphoma.Conclusion When patients present with unilateral swollen cheek without proximal skeletal muscle weakness or myalgia,even the pathologic evaluation implies inflammatory myopathy,non-Hodgkin's lymphoma should be considered.Repeated biopsy may be necessary to confirm the diagnosis.

OBJECTIVETo investigate the potential protective effect of the mitochondria-targeted antioxidant Mitoquinone (MitoQ) on post-thaw human sperm.
METHODSSemen samples were collected from 60 normal fertile men, each divided into six parts of equal volume to be incubated at 37 °C in normal saline (G0, control) or in the extender with 2 nmol/L (G1), 20 nmol/L (G2), 200 nmol/L (G3), 2 µmol/L (G4), and 20 µmol/L of MitoQ (G5). After one hour of incubation, the samples were subjected to computer-assisted semen analysis (CASA) for sperm motility, flow cytometry for reactive oxygen species (ROS), thiobarbituric acid assay for the concentration of malondialdehyde (MDA), and MitoTracker fluorescent staining and flow cytometry for the sperm mitochondrial membrane potential (MMP). Then, the semen were cryopreserved with none (B0), 200 nmol/L (B1), and 2 µmol/L of MitoQ (B2), followed by detection of the changes in the ROS, MDA, and MMP of the post-thaw sperm.
RESULTSThe percentage of progressively motile sperm and total rate of sperm motility were significantly higher in G3 ([30.8 ± 10.2]% and [70.6 ± 9.0]%) and G4 ([32.7 ± 13.5]% and [70.3 ± 11.9]%) than in G0 ([17.6 ± 5.0]% and [54.9 ± 11.5]%) (P < 0.05). The level of ROS dropped markedly with the increased concentration of MitoQ, 86.5 ± 31.6 in G3, 93.6 ± 42.0 in G4, and 45.1 ± 15.0 in G5, as compared with 160.8 ± 39.7 in G0 (P < 0.05). The content of MDA was remarkably lower in G3 ([0.9 ± 0.5] µmol/mg) and G4 ([0.9 ± 0.5] µmol/mg) than in G0 ([1.9 ± 1.1] µmol/mg) (P < 0.05), but not in G5 ([1.7 ± 0.7] µmol/mg), which was even higher than in G3 and G4 (P < 0.05). The MMP showed a significant reduction in G5 (1156 ± 216) in comparison with G0 (1701 ± 251) (P < 0.05) but exhibited no remarkable difference between G0 and G1 (1810 ± 298), G2 (1995 ± 437), G3 (1950 ± 334), or G4 (1582 ± 314). The percentage of progressively motile sperm and total rate of sperm motility after freezing-thawing were significantly decreased as compared with those of the fresh semen (P < 0.01), but both were remarkably higher in B1 ([3.2 ± 2.3]% and [ 43.0 ± 9.5]%) than in B0 ([0.8 ± 0.6]% and [26.5 ± 11.4]%) (P < 0.05). The ROS level was significantly lower in B1 and B2 than in B0 (34.6 ± 12. 3 and 37.0 ± 10.5 vs 56.9 ± 14.3, P < 0.05), and so was the MDA content ([1.4 ± 0.5] and [1.4 ± 0.6] µmol/mg vs [2.6 ± 1.0] µmol/mg, P < 0.05), but the MMP was markedly higher in B1 and B2 than in B0 (1010.0 ± 130.5 and 880.6 ± 128.6 vs 721.1 ± 24.8, P < 0.05).
CONCLUSIONAddition of MitoQ to the freezing extender at 200 nmol/L may effectively improve the quality of human sperm and MitoQ is a good protective addictive for human sperm cryopreservation.
