1.Biodistribution and Postmortem Redistribution of Emamectin Benzoate in Intoxicated Mice.

OBJECTIVE:
To investigate the lethal blood level, the target organs and tissues, the toxicant storage depots and the postmortem redistribution in mice died of emamectin benzoate poisoning.
METHODS:
The mice model of emamectin benzoate poisoning was established via intragastric injection. The main poisoning symptoms and the clinical death times of mice were observed and recorded dynamically in the acute poisoning group as well as the sub-acute poisoning death group. The pathological and histomorphological changes of organs and tissues were observed after poisoning death. The biodistribution and postmortem redistribution of emamectin benzoate in the organs and tissues of mice were assayed by the enzyme-linked immunosorbent assay (ELISA) at 0h, 24h, 48h and 72h after death. The lethal blood concentrations and the concentrations of emamectin benzoate were detected by high performance liquid chromatography (HPLC) at different time points after death.
RESULTS:
The symptoms of nervous and respiratory system were observed within 15-30 min after intragastric injection. The average time of death was (45.8 ± 7.9) min in the acute poisoning group and (8.0 ± 1.4) d in the sub-acute poisoning group, respectively. The range of acute lethal blood level was 447.164 0-524.463 5 mg/L. The pathological changes of the organs and tissues were observed via light microscope and immunofluorescence microscope. The changes of emamectin benzoate content in the blood, heart, liver, spleen, lung, kidney and brain of poisoning mice showed regularity within 72 h after death (P < 0.05).
CONCLUSION
The target organs of emamectin benzoate poisoning include heart, liver, kidney, lung, brain and contact position (stomach). The toxicant storage depots are kidney and liver. There is emamectin benzoate postmortem redistribution in mice.

2.The protection study of recombinant human erythropoietin on acute cerebral injury rats

Objective To investigate the effect of recombinant human erythropoietin(rh‐EPO ) on acute cerebral injury and expression of GLT‐1 and GLAST in rat .Methods Sixty SD rats were randomly divided into three groups by weight :control group (n=18) ,acute cerebral injury group(n=22) and rh‐EPO conditioning group(n=20) .Acute cerebral injury models were made by modified Feeney′s method .rh‐EPO was injected in abdominal cavity 15 min after acute cerebral injury in rh‐EPO conditioning group .Rats′brain were removed 48 h after experiments .Rat GLT‐1 and GLAST mRNA expression were determined by RT‐PCR , GLT‐1 and GLAST protein expression were determined by Western blot .Results GLT‐1/GLAST mRNA and protein expression decreased significantly after acute cerebral injury(all P<0 .01) ,but increased significantly in rh‐EPO preconditioning group com‐pared with acute cerebral injury group(all P<0 .01) .Conclusion rh‐EPO preconditioning may protect against acute cerebral injury by up regulating the expression of GLT‐1/GLAST .
3.Influence of Porphyromonas gingivalis on vascular intima adhesion factors and metalloproteinases

Objective: To observe influence of porphyromonas gingivalis (Pg, a main pathogenic bacterium of periodontal disease) on vascular intima adhesion factors and metalloproteinases. Methods: A total of 60 rats were randomly and equally divided into blank control group, Pg low dose group (Pg 2ml) and Pg high dose group (Pg 5ml) according to number table. The latter two groups respectively received intramuscular injection of corresponding dose Pg every three days without antibiotic intervention for 12 weeks. Then their venous blood was taken to measure levels of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), matrix metalloproteinase (MMP)-3 and MMP-9 in three groups. Results: Compared with blank control group, there were significant increase in contents of ICAM-1 [(175.79±14.30)ng/ml vs.(182.62±15.07)ng/ml, (189.39±14.93)ng/ml]、VCAM-1 [(256.49±37.17)ng/ml vs.(271.58±32.85)ng/ml , (286.66±30.66)ng/ml] 、 MMP-3 [(3.23±0.69)ng/ml vs.(3.61±0.82)ng/ml, (3.97±0.83)ng/ml]及 MMP-9 [(1.30±0.39)mg/L vs.(1.48±0.39)mg/L, 1.67±0.45)mg/L ](P <0.05,or <0.01),Compared with Pg low dose group, there were significant increase in levels of above indexes in Pg high dose group (P<0.05). Conclusion: Porphyromonas gingivalis can significantly increase serum contents of vascular intima adhesion factors and metalloproteinases, aggravating pathological development of coronary heart disease.

Objective To study the effect of Fas gene transfection on rectal carcinoma cells in vitro. Methods By using RT-PCR technique, a full length of Fas gene 1007 bp was cloned from actived peripheral mononuclear cells of healthy donors. The fragment was ligated with the pGEM-T Easy and sequenced. The constructed vector was transfected into 8348 cells with lipofectin, the change in expression of Fas gene was determined by RT-PCR. The apoptosis and proliferation of rectal carcinoma cells pre- and posttransfection induced by cisplatin were analysed by ladder and MTT methods. In this study gloxylic acid-induced fluorescent histochemical method was usedto reveal the innervation of adrenergic nerves,acetycholinesterase (AChE) methodto show the cholinergic nerves,and the immunohistochemical method to display thevasoactive intestinal peptide (VIP) and neuropeptide Y (NPY) immunoreactivenerves.This observation on the vas deferens showed that the VIP-immunoreactivenerves had the same distribution as the cholinergic nerves,and mainly existed inlamina propria,while NPY-immunoreactive nerves had the same distribution asadrenergic nerves and mainly existed in the muscular layer.One group of animalswas treated by intraperitoneal injections of 6-OHDA on the lst,2nd,6th and 7thday,and killed on the 8th day.Following treatment with 6-OHDA,significantevanescence of most adrenergic nerves was observed.NPY-immunoreactive nervesreduced remarkably and the extent of the decrease was similiar to that of adrenergicnerves,whereas no change was observed on the VIP and cholinergic nerves.Theimmunoelectronic microscopy (PAP pre-embedding method) demonstrated that VIPimmunoreactive complex deposited in the varicosity containing small clear vesicles(diameter of 40-55nm).Some varicosities also contained small number of largegranular vesicles with the diameter of 100-144mm.NPY immunoreactive complexdeposited in the large granular vesicles,and occasionally can be observed in thesmall granular vesicles,40-55nm in diameter.The results from this study suggestthat VIP probably co-exist with ACh and NPY with noradrenalin in rat vasdeferens.
7.Experimental study of the effect of recombination human growth hormone on the PingPang Racket flap survival

Objective To investigate the effect of recombinant human growth hormone on the PingPang Racket flap survival.Methods Every two PingPang Racket flaps were designed on the both sides of 40 adult SD rats's back.The pedicle size was 1.0 cm × 1.0 cm,while the flap size was 3.0 cm in diameter circular.Longitudinal axis of flap was perpendicular to the center line of the rats back,to which the distance from proximal pedicle was about 1 cm.The flaps on the left side served as Ⅰ group,and the other side served as Ⅱ group,which were subdivided into Ⅰa and Ⅰb,Ⅱa and Ⅱb,respectively.And there were 20 rats in each subgroup.On the flap surfaces in group Ⅰ,it was 6 uniform injection poinsts,subcutaneously injecting with rhGF (the dose was 0.1IU · Kg-1 · d-1) for 7 days from the beginning of operation,that were designed.It goes the same way to the group Ⅱ,while normal saline was instead of rhGF.In subgroup Ⅰa and Ⅱa,the flaps were generally observed every day.The percentage of the flap survival area was determinated 7 days after operation.In subgroup Ⅰb and Ⅱb,specimens were collected at the distal end of flap at intraoperative(before injecting rhGF)and 1 st,3rd,5th,7th day after operation.Immunohistochemistry and enzyme-linked immunosorbent were applied to examine the expression of TGF-β1 and CD34,and the microvessel density of the flaps was calculated.Results According to the 7 days' observation after the surgery,the flap survival area percentage of subgroup Ⅰa was (97.00 + 2.12) %,which was significantly higher (P < 0.05) than that of subgroup Ⅱ a,whose was (81.00 +3.43)%.On 1st,3rd,5th and 7th day postoperatively,the expression of TGF-β1,CD34 in both subgroup Ⅰb and Ⅱb were elevated and reached peak on the 5th day.Content of GF-β1 and CD34 in Ⅰb were 1571.40 ± 13.32 pg/ml and 60.40 ±0.32 pg/ml,respectively,and in Ⅱb were 691.43 ± 11.06 pg/ml and 20.43 ± 0.06 pg/ml.At the same point of time,the expression of TGF-β1,CD34 were significant higher in Ⅰb subgroup than that in Ⅱb (P < 0.05).In subgroup Ⅰb and Ⅱb,the number of microvessels increased on postoperative 1 st,3rd,5th and 7th day,especially on 3rd,5th and tended to be stable at 7th day.At the same point of time,the number of microvessels in Ⅰb was always higher than that in Ⅱb (P < 0.05).Conclusion Subcutaneous injection of rhGH on flaps can enhance the expression of TGF-β1,CD34,promote microvascular generation of the flap tissue directly or indirectly,and also improve the survival of PingPang Racket flaps.
8.Investigation and research in constipation causes of patients undergoing replantation of severed finger

Objective To investigate the factors that induce constipation in bedridden patients undergoing replantation of severed finger and find countermeasures. Methods A retrospective investigation was carried out in 92 patients undergoing replantation of severed finger, their constipation and factors influencing constipation were recorded. Results 61 patients suffered from constipation in total 92 cases,the rate being 66.30%. 76 patients reduced food intake, and 59 cases presented constipation (the rate being 77.63%). Factors influencing constipation included being unaccustomed to defecate in bed (the rate that present with constipation being 87.09%),reluctant to trouble others(75.47%), side effects of drugs(55.56%) and lack of dietary knowledge (71.87%), decreasing food intake was closely bound up with constipation. Conclusions Proper psychological counseling, strengthening of health propaganda, comfortable facility for defecation, improved family support and oral food intake can significantly reduce the proportion of constipation and increase comfort degree of patients.
9.X-ray implanted in fixing the fracture localization and navigation in orthopedics

BACKGROUND:The traditional orthopedic fixation by C-arm positioning surface is completed, but the large C-arm injury on the human body and the long fixed time increase the suffering of patients. OBJECTIVE:To investigate the X-ray fixed in position within the orthopedic implants, navigation and effect. METHODS:Twenty-six New Zealand white rabbits were randomly divided into C-arm machine group and X-ray group, with 13 in each group. Rabbits in both groups were used to simulate soft tissue foreign body localization, intramedul ary nail implantation at distal fracture end and spinal pedicle screw entry point position. In the C-arm machine group, positioning navigation was conducted with C-arm machine. In the X-ray group, X-ray positioning navigation was used. The positioning and navigation effects were compared between the two groups. RESULTS AND CONCLUSION:(1) Compared with the C-arm machine group, the time required for navigation in the X-ray group targeting soft tissue foreign body localization, fracture distal locking intramedul ary nail implantation and pedicle screw spinal needle point location was significantly shorter (P<0.05);navigation displacement and deviation produced were significantly less (P<0.05). (2) These findings suggested that the X-ray positioning for orthopedic fixation method is relatively simple, with high availability, and can obtain a high performance-price ratio. Meanwhile, the X-ray localization can improve accuracy and shorten the fixed time.
