Intracytoplasmic sperm injection,which combined with in vitro fertilization and micromanipulation techniques,has been applied to research the molecular mechanisms of fertilization,and produce the sexing livestock and transgenic animals.The research advances in porcine intracytoplasmic sperm injection,including in vitro maturation and pretreatment of oocytes,selection and treatment of spermatozoon,artificial activation of injected oocytes after injection,and improvement of the operation technique were reviewed.

3.Control study on the treatment of refractory neuropathic pain with pregabalin and sertraline hydrochloride

Objective To investigate the effect,security and tolerance of pregabalin combined with sertraline hydrochloride for refractory neuropathic pain.Methods Forty-six patients with refractory neuropathic pain were divided into two groups by random digits table.The patients in pregabalin group (24 patients) were treated by pregabalin combined with sertraline hydrochloride,and the patients in carbamazepine group (22 patients) were treated by carbamazepine combined with sertraline hydrochloride.They were treated for 4 weeks.The scores of visual analogue scales (VAS),Hamilton rating scale of depression (HAMD),Hamilton rating scale for anxiety (HAMA) and the dose,adverse reaction were compared between two groups.Results The total effective rate of pregabalin group was 83.3%(20/24) and carbamazepine group was 86.4% (19/22),there was no significant difference (P> 0.05).After treatment of 1,2,4 weeks,the scores of VAS in two groups were significantly decreased compared with those before treatment[pregabalin group:(5.90 ± 1.11),(4.59 ± 1.33),(3.59 ± 1.94) scores vs.(6.50 ± 0.85) scores; carbamazepine group:(5.50 ± 1.06),(4.18 ± 1.71),(3.73 ± 1.88) scores vs.(6.50 ± 0.80) scores] (P <0.05).After treatment of 2 weeks,the scores of VAS in carbamazepine group was significantly lower than that in pregabalin group (P < 0.05).In pregabalin group,the scores of HAMD/HAMA after treatment of 1 week had no significant difference compared with before treatment (P > 0.05),and the scores of HAMD/HAMA after treatment of 2,4 weeks were significantly increased [(15.25 ± 4.15),(11.29 ± 4.16)scores vs.(16.08 ± 3.56) scores and (14.21 ± 4.72),(11.63 ± 4.50) scores vs.(15.25 ± 4.05) scores] (P <0.05).In carbamazepine group,the scores of HAMD/HAMA after treatment of 1,2 weeks was decreased compared with those before treatment,but there was no significant difference (P > 0.05).The scores of HAMD/HAMA after treatment of 4 weeks was significantly decreased [(13.18 ± 2.42) scores vs.(15.20 ±1.05) scores,(12.32 ± 2.64) scores vs.(14.55 ± 4.43) scores] (P < 0.05).The scores of HAMD/HAMA in pregabalin group after treatment of 4 weeks was significantly lower than that in carbamazepine group (P <0.05).There was no severe adverse reaction.Conclusion Pregabalin combined with sertraline hydrochloride is worth to recommend for refractory neuropathic pain,and has no serious adverse reaction.
4.Danhong Injection inhibits high glucose-induced injury in rat peritoneal mesothelial cells

AIM To investigate that Danhong Injection may decrease inflammation and oxidative stress response in rat peritoneal mesothelial cells (RPMCs) cultured with high glucose.METHODS The third passage of RPMCs were used for the experiment.After being incubated with DMEM for 24 hours,RPMCs were divided into normal control group,high glucose group (RPMCs were incubate with 100 mmol/L glucose for 12 h,24 h,48 h,72 h),Danhong Injection group (complete medium with 80 mL/L Danhong Injection for 48 h) and high glucose + Danhong Injection group (100 mmol/L high glucose with 40 mL/L,80 mL/L,160 mL/L Danhong Injection for 48 h respectively).The cell proliferation was detected by MTT method.The levels of interleukin-18 (IL-18),interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in culture fluid were determined by enzyme linked immunosorbent assay.The contents of malondialdehyde (MDA),superoxide dismutase (SOD),glutathione (GSH),in the culture fluid and intracellular reactive oxygen species (ROS) were measured by kits.The expressions of Bcl-2,Bax,fibronectin (FN),endothelin-1 (EDN1) and haem oxygenase-1 (HMOX1) mRNA were detected by quantitative real-time PCR.The protein expressions of EDN1 and HMOX1 were measured by Western blot.RESULTS High glucose can significantly inhibit RPMCs' viability.High glucose can also up-regulate the expressions of IL-18,IL-6,TNF-α,ROS,MDA,Bax mRNA,FN mRNA,EDN1 mRNA and down-regulate the levels of SOD,GSH,Bcl-2 mRNA,HMOX1 mRNA in RPMCs.Treatment with Danhong Injection can effectively reverse aforementioned effect induced by high glucose.CONCLUSION Danhong Injection can reverse the inhibition of cell viability,the inflammation and oxidative stress response induced by high glucose in RPMCs,thus protect peritoneal membrane.
6.Application of external application of fermented flour in parturient with engorgement and gas pains of breast

Objective To evaluate the effect of external application of fermented flour on postpartum patients with engorgement and gas pains of breasts.Methods Totals of 109 postpartum cases with engorgement and gas pains of breasts in experimental group received external application using fermented flour,while 123 cases in control group received massage (clinical routine),then pain relieve,caring time,acceptable degree,breasts hardness and unobstructed degree of mammary gland during two groups were observed and compared.Results During intervention,in experimental group 33.02% patients felt pain significantly relieved and 27.52% felt pain relieved,onlyl2.19% patients felt pain relieved and 58.54% patients felt pain aggravated in control group,and the difference was statistically significant ( u =10.317,P < 0.01 ).After intervention,81.65% patients felt pain significantly relieved and 13.76% felt pain relieved in experimental group,and 18.70% patients felt pain significantly relieved,50.41% patients felt pain relieved and16.26% patients felt pain aggravated in control group,and the difference was statistically significant( u=9.356,P <0.01 ).The average time of nursing operation in experimental group was ( 28.4 ± 4.1 ) min significantly shorter than (98.3 ± 11.2 )min in control group ( t =8.069,P < 0.01 ).There was no significant difference on the breasts hardness improvement(P > 0.05). In experimental group 79.82% patients' mammary gland completely unobstructed,18.35% patients less unobstructed and 1.83% patients obstructed,while that in control respectively was 47.97%,35.77%,16.26%,and the difference was statistically significant ( u =5.258,P < 0.01 ).Conclusions External application of fermented flour has many advantages such as relieve pain well,unblock mammary gland,save the caring time and easier be accepted and so on.So,it is a favorable nursing care method for postpartum patients with engorgement and gas pains of breasts.
7.Expression of IP-10 and Its Significance in Pathological Scar

Objective To explore the expression of 10 kDa interferon-gamma-induced protein(IP-10)in pathological scar and its role in the pathogenesis of pathological scar.Methods Imrnunohistochemistry was performed to detect the expression and distribution of IP-10 in 28 patients with keloid(group K),34 patients with hypertrophic scar(group HS),and 20 normal controls(group N).The data were collected and analyzed statistically.Results The expression of IP-10 was significantly higher in groups K and HS than in group N(P<0.01),but no significant difference in the expression of IP-10 was found between groups K and HS.Conclusion IP-10 may enhance the formation of pathological scar by attracting T lymphocytes and inducing immune/inflammatory response.
8.Effects of periodontitis on the phenotype and secretion function of macrophages in C57 BL/6J mice model

Objective To explore the splenic macrophages phenotype and secretory function of mice with periodontitis, so as to explore effects of periodontitis on macrophages. Methods 22 mice were randomly divided into periodental ligation group (group P10d ) and pseudo periodental ligation control group (group C), with 11 mice in each group. The experimental periodental ligation on mice lasted for 10 days before they were sacrificed. Flow cytometry was applied to detect the expression of M1 and M2 in mononuclear macrophages. Real-time PCR was applied to detect the relative expression of pro-inflammatory cytokines IL-1β and anti inflammatory cytokines IL-10. Results Compared with the control group C, the proportion of M1 macrophages in the periodontitis group decreased, and the ratio of M1/M2 was also decreased significantly, and IL-1β mRNA also down-regulated. Conclusions Chronic periodontal infection could down regulate the proportion of M1 macrophages , decrease ratio of M1/M2 and the expression of inflammatory cytokines IL-1βmRNA.
