Objective To evaluate the effect of metformin treatment on the thyroid hormone profile in patients with polycystic ovary syndrome (PCOS).Methods A total of 148 patients with PCOS was specifically selected for either being diagnosed of hypothyroidism or subclinically hypothyroid or euthyroid without levothyroxine treatment before the starting of metformin.The serum levels of thyroid stimulating hormone (TSH) and free thyroxine (FT4) were measured before and after a 6-month period of metformin therapy.Results Thyroid function parameters did not change after starting metformin therapy in euthyroid patients with PCOS.In the 9 hypothyroid patients and 38 subclinical hypothyroid patients with PCOS,the basal median serum levels of TSH (P ＜ 0.05) decreased after a 6-month course of mefformin treatment [(2.77 ±1.47) μIU/ml vs (6.77 ± 2.46) μIU/ml;(2.90 ± 1.08) μIU/ml vs (4.05 ± 0.94) μIU/ml,P ＜ 0.05].No significant change in the serum levels of FT4 was observed in these patients (P ＞ 0.05).Conclusions Mefformin treatment has no effect on thyroid hormone,but has a TSH-lowering effect in hypothyroid and subclinically hypothyroid patients with PCOS.

Objective To investigate the effect of streptozotocin (STZ) on learning,memory and expression of p-MKK4,p-JNK and p-Jun in APP transgenic mice.Methods Three-month-old APP/PS1 mice were intraperitoneally injected with STZ.Learning and memory function was performed by Morris water maze.The changes of neuronal morphology in hippocampum were observed by Nissl staining.The expression of MKK4,JNK and c-Jun were detected in hippocampal tissues by Western blotting.Results In the hidden platform experiment of Morris maze,the escape latencies and the pathway length of STZ-induced diabetic mice were longer than those in controls from the 2nd to 5th day(P ＜ 0.05 or P ＜ 0.01).The Nissl staining showed that some neurons in hippocampum of STZ-treated mice were displayed changes of injury.Compared with the vehicle control,the expression levels of pJNK and p-c-Jun were increased by 217.78 ± 23.49 and 189.27 ± 17.80,respectively (P ＜ 0.01).Conclusion STZ can decrease the learning and memory function and injury the neurons,which maybe relate to the JNK signal pathway.

Objective: To study the plasma concentrations and pharmacokinetic parameters of 6,7 dimethoxycoumarin after oral administration of Herba Artemisiae Scopariae decoction and Yinchenhao decoction. Methods: A HPLC UV method was developed to determine 6,7 dimethoxycoumarin in rat plasma. The drug was extracted with acetonitrile from plasma and separated on a Kromasil ODS (250mm?4.6mm,5?m)column with methanol 1% solution of acetic acid THF (30∶63∶7) as mobile phase, UV detecter was set at 340nm. The data obtained were analyzed with Topfit program. Results: The liner calibration curves were obtained in the concentration range of 0.026～2.5 ?g?mL -1 . With non compartmental analysis, the main pharmacokinetic parameters after oral administration of Artemisiae Scopariae decoction and Yinchenhao decoction were as follows: T 1/2 was 1.30 and 1.75h, AUC 0→t was 1215.00 and 2527.85ng?h?mL -1 , AUC 0→∝ was 1325.90 and 2612.58ng?h?mL -1 respectively. Conclusion: This method can be used to study the pharmacokinetics of 6,7 dimethoxycoumarin derived from different traditional Chinese medicines containing this component.

Methods Thirty-two gray rabbits were divided into four groups(n=8).The rabbits were intravitreally injected with buffered saline solution(control group),4 mg triamcinolone acetonide(group B),1.3 mg triamcinolone acetonide(group C) and vehicle(group D).Intraocular pressure,scotopic and photopic electroretinogram examinations were performed before injection and at different time points after injection.Histologic and ultrastructural changes were observed 1 week,1 month and 3 months after injection. [Objective]To study the means and curative effect of the total hip replacement for ankylosed hip joint with severely flexinal abnormality.[Method]Retrospective analysis of the operative effect and related problems of the total hip replacement for ankylosed hip joint with severely flexinal abnormality.[Result]Through follow up for 6 to 44 months(22 months averaged),the average range of motion of hip joint and of flexion was 220.5? and 103?,respectively.The postoperative Harrris Score was 84.4 versus the preoperative Harrris Score of 14.2.The pain of the hip was disappeared almost completely and pain of lower waist and knee was relieved obviously.Patient got their gait right and could care themselves approximately.[Conclusion]The total hip replacement can eliminate the pain of the joint effectively and meliorate the function of the joint rapidly for ankylosed hip joint with severely flexinal abnormality,but surgeons must grasp the indication and technique of the operation and postoperative functional exercise.

Objective To investigate the relationships between the expressions of cidA and lrgA and the biofilm formation of Staphylococcus epidermidis clinical isolates. Methods Thirty-nine S. epidermidis clinical isolates were divided into biofilm formation group and non-biofilm formation group according to adhesion assays and icaA amplification, 20 strains for biofilm formation group and 19 strains for another group respectively. Amplify cidA and lrgA fragment with PCR, detect mRNA levels of cidA and lrgA with RT-PCR and then calculate the ratios of cid/lrg mRNA. Results All of the 39 isolates of S. epidermidis existed cidA and lrgA gene.Select 2 strains of S. epidermidis, Y36 and Y26, at random for detecting the cidA and lrgA mRNA levels of different phases, the highest level of cidA mRNA shows on 4 h's culture while lrgA shows on6 h. The relative expression levels of cidA of 39 isolates of S. epidermidis were 0.340 + 0.250 in biofilm formation group and 0. 406 +0. 408( P =0. 541 ) in non-biofilm formation group for 4 h culture, while the relative expression levels of lrgA were 0.325 ± 0.218 and 0.253 ± 0.211 respectively ( P = 0. 299). There were no significant differences between these two groups. The ratios of cid/lrg mRNA were 1.067 ±0.529 and 1.958 ±1.877 respectively, the difference of population distribution was significant( P = 0.001 ). Conclusion The expressions of cidA and lrgA of S. epidermidis clinical isolates may be time-limited. There were no significant differeces of expressions of cidA and lrgA between biofilm formation group and non-biofilm formation group. The ratios of cid/lrg mRNA maybe have some biological significance.
