3.Recent progress of dry powder inhalation of proteins and peptides.

To provide theoretical and practical basis for the successful formulation design of physically-mixed inhalation dry powder of proteins and peptides, related references were collected, analyzed and summarized. In this review drug micronization technology and commonly used carriers for inhalation dry powder preparation were introduced. For proteins and peptides, supercritical fluid technology and spray-drying are more suitable because of their capabilities of keeping drug activity. Being approved by U. S. Food and Drug Administration, lactose has been extensively used as carriers in many inhalation products. Formulation and process factors influencing drug deposition in the lung, including carrier properties, drug-carrier ratio, blending order, mixing methods, mixing time and the interaction between drug and carrier, were elucidated. The size, shape and surface properties of carries all influence the interaction between drug and carrier. Besides, influence of micromeritic properties of the dry powder, such as particle size, shape, density, flowability, charge, dispersibility and hygroscopicity, on drug deposition in the lung was elaborated. Among these particle size plays the most crucial role in particle deposition in the lung. Moreover, based on the mechanisms of powder dispersity, some strategies to improve drug lung deposition were put forward, such as adding carrier fines, adding adhesive-controlling materials and reprocessing micronized drug. In order to design physically-mixed inhalation dry powder for proteins and peptides with high lung deposition, it is essential to study drug-carriers interactions systematically and illustrate the potential influence of formulation, process parameters and micromeritic properties of the powder.

4.Effect of matrine on cell apoptosis and proliferation and the apoptosis related proteins of human medulloblastoma D341 cells in vitro.

OBJECTIVETo investigate the apoptosis and proliferation effect of matrine on human medulloblastoma cell line D341 in vitro and the effect of the expression of the related caspase 3 and caspase 9 proteins.
METHODSThe D341 cells were cultivated successfully in vitro. Then the cells were divided into 5 groups according to the concentration of matrine (0.5 mg/mI group, 1.0 mg/ml group, 1.5 mg/ml group, 2.0 mg/ml group and the control group was 0 mg/ml). All the experiments were repeated three times. The cell morphologic and structure change was observed with the optical microscope and the transmission electron microscope. The proliferation of D341 cell was analyzed using Cell Counting Kit-8 assay. Apoptosis was detected by Annexin V-FITC/PI double staining. The expression of Caspase3 and Caspase9 was detected by Western blot.
RESULTSWith the effect of matrine, the proliferation inhibition rate gradually increased with drug concentrations increasing, and there was a significant difference (P < 0.01). The inhibitory effect of matrine on cell proliferation was different with the different treatment time, there was a significant difference between the 24 h to 72 h groups (P < 0.01). The apoptotic rate increased with matrine concentrations increasing. There were significant differences between the group of 0.5 mg/mI or 1.0 mg/mI to the group of 1.5 mg/mI or 2.0 mg/mI (P < 0.05). The apoptotic rate increased with the prolonged treatment time. There were significant differences between the group of 24 h or 48 h to the group of 72 h ( P < 0.05). With the increase of matrine concentration, the expression of Caspase 3 and Caspase 9 increased (P < 0.01).
CONCLUSIONMatrine induces the apoptosis, and inhibits the proliferation of human medulloblastoma D341 cells in vitro by up-regulation of the expression level of Caspase3, Caspase9.

6.Study on MR tracking of magnetically labeled swine mesenchymal stem cells after autologous transplantation into acute injured liver in vivo

Objective To evaluate in vivo tracking of swine mesenchymal stem cells (MSCs) la-beled with super paramagnetic iron oxide (SPIO) in intraportal transplantation by a clinical 1.5T MR.Methods MSCs were isolated from swine and cultured as well as expanded, which were then incuba-ted with SPIO (Feridex I. V.). Prussian blue staining was performed for showing intracelluar irons.To establish a swine model of acute liver necrosis, 0.5 g/kg of D-galactosamine was administrated to 10 pigs. MSCs(labeled cells in six, unlabeled cells in four)were injected into liver via portal veins. MR imaging was performed with a clinical 1.5T MR immediately before and at 6 h, 3 d, 7 d, 14 d after transplantation, respectively. Results Prussian blue staining of SPIO labeled MSCs could be effec-tively labeled and the labeling efficiency was almost 100%. Signal intensity loss in liver by SPIO labe-ling on FFE sequence persisted until 14 days after transplantation. Histological analysis by Prussian blue staining showed homing of labeled MSCs in liver after 14 days, primarily distributing in hepatic sinusoids and liver parenchyma. Conclusion MSCs can be labeled with SPIO in vitro successfully.MRI can monitor magnetically labeled MSCs transplanted into liver.
9.Clinical Study on Thread Embedding at the Holographic Point of the Second Metacarpal Bone for Tinnitus

Objective To observe the clinical efficacy of thread embedding at the holographic point Kidney of the second metacarpal bone in treating tinnitus.Method Eighty-two patients with tinnitus were randomized into a treatment group and a control group, 41 cases in each group. The treatment group was intervened by thread embedding at the holographic point Kidney of the second metacarpal bone, while the control group was by intragluteal injection with Mecobalamin 0.5 mg. The Tinnitus Handicap Inventory (THI) was compared before intervention and after 1 treatment course.Result The THI scores were significantly changed in both groups after intervention (P<0.01). The change of THI score after intervention in the treatment group was significantly different from that in the control group (P<0.05).Conclusion Thread embedding at the holographic point of the second metacarpal bone in an effective approach in treating tinnitus.
10.Analysis of international literatures on geriatrics and gerontology in mainland of China

Objective To investigate the situation and development of international literatures on geriatrics and gerontology in mainland China.Methods The literatures on geriatrics and gerontology written by mainland Chinese scholars as the first author were selected from Thomson Scientific database in 1979-2011 period.Researches on basic theory,summary reports of clinical practice and reviews were collected.The quantity of published articles,article types,international cooperation research and funding programs,hospital rankings,as well as the 2011 geriatrics professional journals,thesis topic disciplines distribution and high-performance papers were analyzed by Thomson Data Analyzer software.Results From 1979 to 2011,the quantity of literatures written by Chinese scholars as the first author (not including Hong Kong,Macao and Taiwan regions) was 1927,including 1769 research articles,86 reviews,18 editorials,54 letters.Average paper was cited 7.6 times and the average number of article citation was 30.6.International cooperation research articles occupied 379 (19.7 %).Articles with research sponsorship were 734 (38.1 %),in which most subject discipline was geriatric and gerontology.Conclusions The development of geriatric medicine in China is rapid.The international scientific and technological cooperation is very extensive.It is important to further improve the world impact of geriatric medicine from China.The development of geriatric medicine in a large,cooperative,comprehensive,interdisciplinary,interagency direction,and the prospective,multicenter,cooperative study need to promote.It is extremely urgent to establish the English version of Chinese journals of geriatrics with a international influence.
